A Novel Algicidal Bacterium, Microbulbifer sp. YX04, Triggered Oxidative Damage and Autophagic Cell Death in Phaeocystis globosa , Which Causes Harmful Algal Blooms

P. globosa is one of the most notorious harmful algal bloom (HAB)-causing species, which can secrete hemolytic toxins, frequently cause serious ecological pollution, and pose a health hazard to animals and humans. Hence, screening for bacteria with high algicidal activity against P. globosa and studies on the algicidal characteristics and mechanism will contribute to providing an ecofriendly microorganism-controlling agent for preventing the occurrence of algal blooms and reducing the harm of algal blooms to the environment.

Cyanopeptides Restriction and Degradation Co-mediate Microbiota Assembly During a Freshwater Cyanobacterial Harmful Algal Bloom (CyanoHAB)

Cyanobacterial harmful algal blooms (CyanoHABs) are globally intensifying and exacerbated by climate change and eutrophication. However, microbiota assembly mechanisms underlying CyanoHABs remain scenario specific and elusive. Especially, cyanopeptides, as a group of bioactive secondary metabolites of cyanobacteria, could affect microbiota assembly and ecosystem function. Here, the trajectory of cyanopeptides were followed and linked to microbiota during Microcystis-dominated CyanoHABs in lake Taihu, China. The most abundant cyanopeptide classes detected included microginin, spumigin, microcystin, nodularin and cyanopeptolin with total MC-LR-equivalent concentrations between 0.23 and 2051.54 ppb, of which cyanotoxins beyond microcystins (e.g., cyanostatin B and nodularin_R etc.) far exceeded reported organismal IC50 and negatively correlated with microbiota diversity, exerting potential collective eco-toxicities stronger than microcystins alone. The microbial communities were differentiated by size fraction and sampling date throughout CyanoHABs, and surprisingly, their variances were better explained by cyanopeptides (19-38%) than nutrients (0-16%). Cyanopeptides restriction (e.g., inhibition) and degradation are first quantitatively verified as the deterministic drivers governing community assembly, with stochastic processes being mediated by interplay between cyanopeptide dynamics and lake microbiota. This study presents an emerging paradigm in which cyanopeptides restriction and degradation co-mediate lake water microbiota assembly, unveiling new insights about the ecotoxicological significance of CyanoHABs to freshwater ecosystems.

Temporal and Spatial Patterns of Harmful Algae Affecting Scottish Shellfish Aquaculture

Consistent patterns of Harmful Algal Bloom (HAB) events are not evident across the scientific literature, suggesting that local or regional variability is likely to be important in modulating any overall trend. This study summarizes Scotland-wide temporal and spatial patterns in a robust 15-year high temporal frequency time series (2006–2020) of the incidence of HABs and shellfish biotoxins in blue Mussels (Mytilus edulis), collected as part of the Food Standards Scotland (FSS) regulatory monitoring program. The relationship between the countrywide annual incidence of HAB events and biotoxins with environmental variables was also explored. Temporal patterns exhibited interannual variability, with no year-on-year increase, nor any correlation between annual occurrences. Within years, there was a summer increase in bloom frequency, peaking in July for Dinophysis spp. and Pseudo-nitzschia spp., and a plateau from May to July for Alexandrium spp. Temporal-spatial patterns were analyzed with multivariate statistics on data from monitoring sites aggregated monthly into 50-km grid cells, using Principal Component Analysis (PCA) and cluster K-means analysis. PCA analyses showed correlation between areas with similar temporal dynamics, identifying seasonality as one of the main elements of HAB variability with temporal-spatial patterns being explained by the first and second principal components. Similar patterns among regions in timing and magnitude of blooms were evaluated using K-means clusters. The analysis confirmed that the highest risk from HABs generally occurred during summer, but demonstrated that areas that respond in a similar manner (high or low risk) are not always geographically close. For example, the occurrence of the most prevalent HAB genus, Dinophysis spp., is similar countrywide, but there is a regional trend in risk level with “very-high” and “high” clusters located primarily on the southwest coast, the islands of the central and northern west coast and the Shetland Islands. “Early” and “late” blooms were also associated with certain areas and level of risk. Overall, high risk areas mainly face in a southwest direction, whilst low risk locations face a south or southeast direction. We found relatively few countrywide relationships between environmental variables and HABs, confirming the need for regional analysis to support HAB early warning.

Biochemical Mapping of Pyrodinium bahamense Unveils Molecular Underpinnings behind Organismal Processes

Proteins, lipids, and carbohydrates from the harmful algal bloom (HAB)-causing organism Pyrodinium bahamense were characterized to obtain insights into the biochemical processes in this environmentally relevant dinoflagellate. Shotgun proteomics using label-free quantitation followed by proteome mapping using the P. bahamense transcriptome and translated protein databases of Marinovum algicola, Alexandrium sp., Cylindrospermopsis raciborskii, and Symbiodinium kawagutii for annotation enabled the characterization of the proteins in P. bahamense. The highest number of annotated hits were obtained from M. algicola and highlighted the contribution of microorganisms associated with P. bahamense. Proteins involved in dimethylsulfoniopropionate (DMSP) degradation such as propionyl CoA synthethase and acryloyl-CoA reductase were identified, suggesting the DMSP cleavage pathway as the preferred route in this dinoflagellate. Most of the annotated proteins were involved in amino acid biosynthesis and carbohydrate degradation and metabolism, indicating the active roles of these molecules in the vegetative stage of P. bahamense. This characterization provides baseline information on the cellular machinery and the molecular basis of the ecophysiology of P. bahamense.

A theoretical modeling framework for motile and colonial harmful algae

1. Harmful algal blooms are increasing in both severity and frequency across the globe. Many bloom-forming species are capable of vertical motility and colony formation. The cyanobacterium Microcystis aeruginosa is a common example of such a species, yet current models poorly predict vertical distributions of M. aeruginosa. 2. To couple the hydrodynamics, buoyancy, and the colony dynamics of Microcystis, we present a system of one-dimensional advection-diffusion-aggregation equations with Smoluchowski aggregation terms. 3. Results indicate Smoluchowski aggregation accurately describes the colony dynamics of M. aeruginosa. Further, transport dynamics are strongly dependent on colony size, and aggregation processes are highly sensitive to algal concentration and wind-induced mixing. Both of these findings have direct consequences to harmful algal bloom formation. 4. While the theoretical framework outlined in this manuscript was derived for M. aeruginosa, both motility and colony formation are common among bloom-forming algae. As such, this coupling of vertical transport and colony dynamics is a useful step for improving forecasts of surface harmful algal blooms.
