Рассматривается построение и уравнивание аналитической пространственной фототриангуляции, ос- нованной на совместном использованием условия коллинеарности и условия компланарности векторов. Получена структура системы нормальных уравнений. Предлагаемое решение позволяет выполнять по- строение фототриангуляции с одновременным определением (уточнением) элементов внешнего ори- ентирования и внутреннего ориентирования съёмочной камеры. Для назначения начальных значений весов уравнений, составленных с использованием условия компланарности, для некоторой точки ис- следуемого объекта (местности) предлагается использовать ошибки определения пространственных координат точки, рассчитанные методом прямой фотограмметрической засечки для произвольного случая съёмки.

The purpose of this study was to evaluate the effects of sample handling, storage, and culture techniques on the isolation of Pythium insidiosum from infected equine tissues. Tissue and kunker samples obtained immediately posteuthanasia from a horse with subcutaneous pythiosis were used to assess the effects of sample type (kunkers vs. tissues), media type (selective vs. nonselective), storage technique, and storage time on P. insidiosum isolation rate. Overall, isolation rates were higher from fresh kunkers (94.6%) and stored kunkers (76.4%) than from fresh tissues (8.3%) or stored tissues (4.6%). Isolation of P. insidiosum also occurred more often on antibiotic-containing media than on nonselective media for both fresh and stored samples. For samples that were stored for 1–3 days prior to culture, P. insidiosum isolation rates were highest for the following techniques: kunkers stored at room temperature and plated on selective media (100%), kunkers stored at 4 C and then plated on either nonselective (91.7%) or selective (95.8%) media, kunkers stored on cold packs and then plated on either nonselective (93.8%) or selective (100%) media, kunkers stored in ampicillin solution and plated on selective media (100%), and kunkers stored in ampicillin/gentocin solution and plated on selective media (87.5%). For samples stored for 4–5 days, P. insidiosum isolation rates were highest for kunkers stored at 4 C and then plated on either nonselective (81.3%) or selective (87.5%) media, kunkers stored in ampicillin solution and then plated on selective media (87.5%), and kunkers stored in ampicillin/gentocin solution and plated on selective media (87.5%). Results of this study suggest that optimal isolation rates of P. insidiosum from infected equine tissues are achieved by culturing fresh kunkers on selective media. For samples that cannot be processed immediately, acceptable handling techniques include storage at room temperature for up to 3 days, refrigeration for up to 5 days, shipping on cold packs, and storage in antibiotic solution, each combined with subsequent inoculation on selective media.

Abstrak Teknik Penyimpanan Krioproservasi  Biakan Agrobacterium tumefaciens : Bahan Penelitian  di  Laboratorium Terpadu dan Sentra Inovasi Teknologi Universitas Jember.  Penelitian merupakan kegiatan yang memerlukan bahan yang berkualitas, salah satu cara menjaga kualitas bahan dengan tehnik penyimpanan yang baik. Pemilihan penggunaan tehnik kriopreservasi pada penelitian ini bertujuan menjaga viabilitas  bakteri dalam jangka waktu tertentu. Teknik kriopreservasi yang digunakan  disesuaikan dengan ketersediaan alat pendingin yang ada. Diharapkan dengan penggunaan tehnik kriopreservasi dapat meningkatkan efiisiensi  dan viabilitas sel dalam jangka waktu enam bulan penyimpanan. Penelitian dilakukan di laboratorium Biologi Molekul dan Bioteknologi  menggunakan Agrobacterium tumefaciens strain GV yang sudah terinsersi gen SPS dalam plasmid pKYS (GVpKYS SPS). Bakteri ditumbuhkan pada media yeast, peptone NaCL dengan penambahan antibiotik 100 ppm rifampisin, 12,5 ppm gentamisin dan 50 ppm kanamisin. Dalam biakan bakteri ditambahkan 15% gliserol sebagai kreoprotektan, kemudian dilakukan pembekuan menggunakan nitrogen cair (-196 °C) dan disimpan selama 6 bulan pada suhu dingin. Analisa yang dilakukan adalah uji viabilitas bakteri dan stabilitas genetik diawal dan akhir masa simpan. Hasil penelitian ini diharapkan dapat menyiapkan bahan penelitian yang berkulaitas dan memberikan informasi teknik penyimpanan dingin yang baik dan effisien pada biakan A. tumefaciens khususnya dan bakteri lain pada umumnya.     Abstrac   Cryoproservation Storage Technique for Agrobacterium tumefaciens Culture : Research Material in Center for Devolepment of Advance Science and Technology (CDAST) University of Jember. This research has tole requires quality materials, one of the way to maintain the quality of materials with good storage techniques. Choosing cryopreservation techniques in this study aims to maintain viability of bacteria in a certain period of time. The cryopreservation technique used is adjusted to the availability of existing cooling devices. It is expected that the use of cryopreservation techniques can improve efficiency and viability of cell within six months of storage. The research was conducted in the Molecular Biology and Biotechnology laboratory using Agrobacterium tumefaciens strain GV which was inserted into the SPS gene in plasmid pKYS (GVpKYS SPS). The bacteria are grown on the yeast, NaCL, peptone media. with the addition of 100 ppm antibiotic rifampicin, 12.5 ppm gentamicin and 50 ppm kanamycin. In culture the bacteria added 15% glycerol for cryoprotectant, then it is freezeed using liquid nitrogen (-196 °C) and stored for 6 months in cold temperatures. The analysis carried out was a viability of bacterial test and genetic stability at the beginning and end of the shelf life. The results of this study are expected to be able to prepare quality research materials and provide information on good and efficient cold storage techniques in particular culture of A. tumefaciens and other bacteria.

The purpose of this paper is to indicate a method for estimating values of specified aquifer parameters from an investigation of the reservoir performance of an associated oilfield. To achieve this objective an analysis was made of the simultaneous solution of the material-balance and diffusivity equations, followed by an application of the method of least squares. Three analytical functions evolved, which in dimensionless form were numerically evaluated by computer and tabulated herein. Application of the proffered method requires the simultaneous solution of the three normal equations developed in the paper.
