Leaf explants were cultured on a basic nutrient agar medium to which were added growth substances (adenine, benzyladenine, kinetin, 2-iP [isopentenyladenine], zeatin or 2,4-D) and NH4NO3, (NH4)2SO4 or NaNO3 in various concentrations. The effects of using the surfactant Tween 20 during leaf sterilization and of growing explants in light and/or darkness for 20 weeks were also investigated. There was a strong positive correlation between callus formation and shoot regeneration. Regeneration was optimal under the following conditions: addition of 0.25-1.00 ml/litre Tween 20 during leaf sterilization, adding a growth substance (adenine 0.1 mg/litre, zeatin 1 mg/litre, or 2,4-D 0.08 mg/litre), culture during 16 weeks in darkness followed by 4 weeks of light, and including 206 mg/litre NH4NO3 in the medium. The promoting effect of low levels of NH4NO3 on shoot regeneration in callus was caused by the NH4+ ion and not by the NO3- ion. (Abstract retrieved from CAB Abstracts by CABI’s permission)

Histological analysis of somatic embryos derived from in vitro-grown lamina of Anthurium andraeanumshowed bipolarity with the presence of shoot and root poles connected by procambium. Vascular connections between the explant and somatic embryos were not observed. Storage of proteins, starch and raphides as well as a suspensor-like structure and an epidermis were observed in the somatic embryos. Origin of the somatic embryos was from a proembryonic cell complex or possibly from a single cell by direct embryogenesis. Both modes of somatic embryogenesis arose from the mesophyll.

Novos híbridos de <i>Anthurium andraeanum</i> Lind., resultantes de um programa de seleção em andamento na Seção de Floricultura e Plantas Ornamentais do Instituto Agronômico de Campinas, vêm sendo micropropagados via cultura in vitro de explantes foliares. O melhor procedimento definido para a micropropagação desta espécie é: a) Esterilização: álcool 70° GL por 10 seg., solução de 1,5% de CaOCl2 + Tween 20 por 10 min., seguidas de três lavagens com água esterilizada; b) Meio de inoculação: conforme PIERIK (1976) modificado; c) Meio de repicagens: conforme PIERIK (1976) modificado, eliminado-se a fase líquida; d) Meio de enraizamento: conforme PIERIK (1976) modificado ou MS com 20g/l de sacarose.
