Plant tissue culture involves the culture of all types of plant cells, tissues and organs under aseptic conditions. This definition also extends to the culture of excised embryos and to protoplast culture. An overview of tissue culture techniques and their applications in plant propagation and genetic improvement of plants is presented. The areas under review include: (1) embyro culture, (2) meristem culture, (3) micropropagation, (4) somatic embryogenesis, (5) somaclonal variation, (6) in vitro selection, (7) anther culture and (8) protoplast culture. Problems and limitations of each of the techniques are also discussed. Examples are given of work that has been undertaken or that is currently in progress on the application of these techniques to the improvement of Queensland's subtropical horticultural industries. Key examples are: (1) embryo culture to facilitate incorporation of genes conferring disease-resistance from wild Cucurbita species into cultivated varieties, (2) meristem culture for virus elimination in strawberries (Fragaria × ananassa) and sweet potato (Ipomoea batatas), (3) micropropagation for rapid increase in new varieties of ginger (Zingiber officinale) and pineapple (Ananas comosus) to enable more rapid field evaluation and early release, (4) micropropagation of disease-free, genetically uniform planting material of superior female papaya (Carica papaya) selections and banana (Musa spp.) selections and (5) the use of somaclonal variation and gamma-irradiation for the genetic improvement of banana. Finally, future opportunities for the utilisation of tissue culture in plant propagation and improvement in Queensland's horticultural industries are summarised.

Objetivo: Elaborar películas comestibles con la pectina extraída de la cáscara de piña, (Ananas comosus L.) y evaluar características de color, resistencia a la perforación y degradabilidad. Diseño/metodología/aproximación: Las películas comestibles se elaboraron utilizando el método de vaciado en placa (7ml y 10ml). Resultados: Las películas de pectina de cáscara de piña tuvieron valores de luminosidad y resistencia a la perforación menores (entre 42.18 ±0.47 y 31.07 ±0.66; entre 8.87 ±0.82 N y 3.61 ±0.4 N) que las del control (cítricos) (entre 86.0 ±0.26 y 85.46 ±0.99; entre 26.01 ±2.9 N y 18.82 ±1.9 N) pero su degradabilidad (% pérdida de peso) al primer día fue significativamente mayor (P<0.05); al final (7 d) ambos tipos de películas mostraron una degradabilidad similar (99%). Limitaciones del estudio/implicaciones: Se deben mejorar las propiedades físicas y mecánicas de las películas comestibles hechas con pectina extraída de la cáscara de piña. Hallazgos/conclusiones: Este trabajo demostró que las películas a partir de pectina de cáscara de piña son altamente biodegradables y no se descarta su uso y aplicación en alimentos; sin embargo, se requieren más estudios para la optimización de sus características físicas y mecánicas.

On the basis of the studied materials from the ultraoligotrophic lake Elgygytgyn (Chukotka), a rare for the flora of Russia diatom species Navicula schmassmannii was recorded. In the studied populations of the species a wide variability of morphological quantitative (valve length and width, the number of stria in 10 μm) and qualitative (the shape of the valve and the central area) features was marked. It made the basis for emending of the species diagnosis.
