Molecular detection, characterization of Anaplasma spp. in domestic cats from Rio de Janeiro state

Abstract Ehrlichiosis is caused by agents belonging to Ehrlichia genus. Despite the frequent reports on the serological and molecular detection of E. canis in dogs in Brazil, there is scant data on ehrlichiosis in brazilian cats. This study aimed at investigating the occurrence of Ehrlichia spp. in domestic cats from Greater Rio de Janeiro, and evaluating hematological changes associated with this rickettsial infection. We searched for IgG antibodies against E. canis on blood samples of 216 cats by Indirect Fluorescence Assay (IFA). Additionally, we performed nested PCR (nPCR) and real-time PCR (qPCR) assays targeting E. canis-16S rRNA and dsb gene, respectively. Fifty-seven (26.4%) cats were seropositive for Ehrlichia spp. by IFA. Ehrlichia spp.-16S rRNA gene fragments were detected in 3 cats (1.4%). Although the obtained 16S rRNA sequences showed 99 to 100% identity with E. canis, cats were negative in qPCR. Anemia, thrombocytopenia, leukocytosis, left shift neutrophil and hyperproteinemia were observed. Anemia was statistically associated with seropositivity to E. canis and kittens showed lower positivity rates (p<0.05). This study showed that Ehrlichia spp. occur in domestic cats from Greater Rio de Janeiro. Further studies involving culture isolation are much needed to more precisely characterize these organisms.

Abstract Triatoma vitticeps is a triatomine with geographic distribution restrict to Brazil, which exhibits high prevalence of Trypanosoma cruzi natural infection. Of special epidemiologic concern, this species often invades households in the states of Rio de Janeiro, Minas Gerais and Espírito Santo. The objective of this study was to evaluate morphological and ultrastructural parameters on three T. cruzi isolates obtained from wild T. vitticeps specimens. The growth and cell differentiation of the parasite was evaluated through epimastigote and trypomastigote forms obtained in the growth curves for three distinct isolates. The maximum growth showed differences at the 20th day of the curve. Our in vitro results show a heterogeneity, regarding these features for samples cultivated under the same conditions. Morphometric analyzes based on the shape of epimastigotes and trypomastigotes corroborated such differentiation. These results highlight the need of better understanding the meaning of this diversity under an eco-epidemiological perspective.

Sugarcane (Saccharum spp.) cultivation is one of the major agricultural activities in the Brazilian states. This study aimed to molecularly identify the pathogen associated with rust in sugarcane cultivars in the state of Rio de Janeiro and to suggest a control strategy. Among the 14 PCR-tested cultivars, Puccinia kuehnii infection was identified for RB947520, RB92606, RB835486, RB72454, SP89-11I5, SP83-2847, both from infected leaf sample and from urediniospores. Puccinia kuehnii was not detected by PCR for the cultivars RB955971, RB951541, RB92579, RB867515, RB855536, SP91-1049, SP80-3280, SP80-1816. This is the first molecular detection of this fungus in the state of Rio de Janeiro for six of the 14 analyzed cultivars.

Sanitary sewage collection and treatment is a serious environmental problem in Brazilian cities, as well as the destination of solid waste resulting from this process, i.e. the sewage sludge, a substance rich in organic matter and nutrients, which is normally discarded in landfills. The aim of this study was to characterize the sewage sludge generated in four treatment stations in Rio de Janeiro State, Brazil and check if they meet the legal criteria of the National Environment Council (CONAMA), Resolution No. 375/2006. It also focused on analyzing the perspectives for its agricultural recycling based on the potential demand for main agricultural crops grown in Rio de Janeiro State. Samples from eight sewage sludge lots from four treatment stations located in the metropolitan area of Rio de Janeiro were analyzed. These stations receive and treat only domestic sewage by activated sludge system. For chemical and biological characterization of these lots, representative samples were collected and analyzed according to parameters of CONAMA Resolution No. 375/2006. In order to analyze the perspectives of agricultural recycling of sewage sludge in Rio de Janeiro State, 10 crops with the largest cultivated area in the state were surveyed and analyzed which of them are apt to receive sewage sludge as fertilizer and/or soil amendment. To determine the potential demand for sewage sludge in agriculture, the area occupied by these crops were multiplied by each fertilizer recommendation considering the sewage sludge as fertilizer. The analyzed sludge presented a high content of nutrients and organic matter and was included in the parameters of heavy metals, pathogenic agents, and bacteriological indicators stipulated by CONAMA Resolution No. 375/2006. The agricultural panorama of Rio de Janeiro State is favorable for agricultural recycling of sewage sludge since there is a great potential demand for this residue and, among the 10 agricultural crops with the largest cultivated area in the state, sludge can be used as fertilizer or soil amendment in eight of them. The implementation of a State Sewage Sludge Recycling Program could dispose of this material in a more sustainable way, as well as improve the quality of agricultural soils and reduce costs with chemical fertilizers, bringing positive results for sanitation companies and producers.
