AbstractA total of 791 microsatellites (SSRs) were isolated from 7055 Panax ginseng expressed sequence tags (ESTs). According to primer design criteria, 68 primer pairs for EST-SSR were designed. Under an appropriate polymerase chain reaction (PCR) system, all EST-SSR primer pairs were screened against genomic DNA of Ji'anchangbo and Fusong'ermaya from Panax ginseng, and 43 EST-SSR primer pairs out of the above 68 resulted in PCR products. Then, all 43 pairs were detected in nine P. ginseng, two Panax quinquefolius and two Acanthopanax senticosus cultivars for polymorphisms, and 26 pairs (60.47%) were found to be polymorphic, accounting for 38.23% of the total number of designed primer pairs. These results demonstrate the possibility of developing EST-SSR markers using P. ginseng ESTs.

Abstract:Asian ginseng (Panax ginseng) and American ginseng (P. quinquefolius) are valuable medicinal herbs whose roots have been used for ages in traditional medicine in China and North America as vitalizing and stimulating agents. The roots are obtained mainly from field cultivation, which is a slow (5-7 years long), laborious, and troublesome process; so in vitro methods started to be used to produce ginseng biomass. In our study, non-organogenic callus of P. quinquefolius synthesized the same active substances like field roots, for more than 6 years. The ginsenosides are derivatives of protopanaxadiol (Rb1, Rb2, Rc, Rd) or protopanaxatriol (Rg1, Re). The synthesis of Rg1and Re metabolites is preferred in calli cultured in the dark and with ageing of culture.

We adopted a new extraction method - Accelerated Solvent Extractor (ASE)[1] to extract the liposoluble constituents of Panax quinquefolius and Panax ginseng. GC-MS was used to analyze extract constituents with liposoluble, and which main peaks were identified. It was the fingerprints of liposoluble constituents of Panax quinquefolius and Panax ginseng [2]. This method was able to expose liposoluble constituents with a full range offering a new method for identification and controlling quality of Panax quinquefolius and Panax ginseng.
