New series of copper (II), cobalt (II), zinc (II), nickel (II), manganese (II), iron (II) complexes of a novel Schiff base were prepared by the condensation of sulphadizine and pyridoxal hydrochloride. The ligand and metal complexes were characterized by utilizing different instrumental procedures like microanalysis, thermogravimetric examination and spectroscopy. The integrated ligand and transition metal complexes were screened against various bacteria and fungus. The studies demonstrated the enhanced activity of metal complexes against reported microbes when compared with free ligand.

Two step synthesis of Schiff base ligand and its transition metal complexes was done by condensation reaction. In first step, the drug and aldehyde in equimolar ratio were refluxed for one hour at pH 8-9 in order to get Schiff base ligand. In second step, ligand and metal salts were refluxed for 2 hour. The ligand and Cu(II), Ni(II), Co(II), Fe(II), Mn(II), Zn(II) complexes were characterized by using different instruments like FT-IR, 1H-NMR, 13C-NMR, Mass, Atomic absorption spectrometer, Elemental analyzer, UV-visible spectrophotometer, Evans balance, Conductivitymeter and Thermogravimeter. In vitro antibacterial, antifungal and anti-inflammatory activities were also studied. The synthesized ligand and transition metal complexes were tested against Escherichia coli, Enterobacter aerogenes, Staphylococcus aureus, Bacillus pumilus, Klebsiella oxytoca, Clostridium butyrium, Mucor and Aspergillus niger. These studies demonstrated the enhanced activity of metal complexes against reported bacterial and fungal strains when compared with free Schiff base ligand. The Cu(II) complex recognized as anti-inflammatory agent while the parent drug showed no activity.

In this study, In Vitro antibacterial and antifungal activities of azo series based on transition metal complexes (Cu2+, Zn2+, Mn2+, Co2+ and Ni2+) with already our reported ligands named as; [(E)-1-(1, 3-dioxolan-2-yl)-2-phenyldiazene] (K-1), [(E)-1-(1, 3-dioxolan-2-yl)-2-(4-methylphenyl)diazene] (K-2), 2-[(E)-phenyl diazenyl]-1H-benzimidazole] (K-3), [(E)-1-(1, 3-dioxolan-2-yl)-2-(4-ethylphenyl)diazene] (K-4), and [(E)-1-(1, 3-dioxolan-2-yl)-2-(2-methylphenyl)-diazene] (K-5) were studied. FTIR 1H-NMR and mass spectrometric techniques were applied for interpretation of synthesized complexes. 4.05-4.07 ppm chemical shift appearance of azo group confirms azo coupling with transition metal complexes. N=N, C-H, C-N and C-O groups are also confirmed by FTIR which exhibited peaks at 1400-1500, 2090-3090, 1100-1180, 1010-1060 and 625-780 cm-1. Furthermore, mass spectroscopic data also gives strong indication for the synthesis of metal complexes. All the newly synthesized complexes were screened for their antibacterial and antifungal activities. Antibacterial and antifungal activity against S. aureus, E.coli and A.niger, A.ustus and C.albican at conc. 250 and#181;g/ml showed excellent activity by K-1 complexes (Co2+, Cu2+, Ni2+), K-5 complexes (Zn2+, Mn2+, Cu2+), K-2 complexes (Co2+, Cu2+, Mn2+) and K-3 (Zn2+, Mn2+, Co2+, Cu2+) as compared to standard drug (Ampicillin). Hence, based on this study, it was concluded that these azo based complexes may act as a platform for designing more active antibacterial and antifungal agents.

The current study was designed to investigate the interactions of an antimicrobial drug secnidazole and its two transition metal complexes with bovine serum albumin (BSA). The interactions of secnidazole and its both transition metal complexes were confirmed by the extingushing of fluorescence intensity of the protein. The fluorescence quenching of BSA by the drug and its both metal complexes showed a static quenching process and the reactions followed exothermic mechanism. The fluorescence spectroscopic method was utilized to evaluate the thermodynamic parameters like change of enthalpy (ΔH), entropy (ΔS) and Gibb’s free energy (ΔG) which indicated the bindings of the antimicrobial agent and its both metal chelates were hydrogen bonding and van der Waals interactions. The binding constant and the number of binding sites were also measured by double log plot that indicated the drug or its metal complexes bound with BSA at 1:1 ratio.
