1. Salmonella typhi, strain Ty2, grown in vivo and employed as acetone-dried vaccine possessed a higher immunizing potency than the descendants of the same parent strain grown in vitro and employed as vaccine.2. When 2 × 108in vitro-grown bacteria were employed as challenge, the immunizing effects of both types of vaccine were more marked than after administration of 2 × 108in vivo-grown bacteria as challenge.3. The higher potency of the in vivo-grown vaccine was apparent in all experiments, whether the challenge strain was grown in vivo or in vitro.4. Immunogenic substances were isolated from infected organs of mice and guinea-pigs, and an immunogenic substance from the peritoneal fluid of the infected guinea-pigs was concentrated by precipitation with ethanol.

Abstract Background Resistance to anti-tuberculosis (TB) drug is a major issue in TB control, and demands the discovery of new drugs targeting virulence factor ESX-1. Methods We first established a high-throughput screen (HTS) assay for the discovery of ESX-1 secretion inhibitors. The positive hits were then evaluated for the potency of diminishing the survival of virulent mycobacterium and reducing bacterial virulence. We further investigated the probability of inducing drug-resistance and the underlying mechanism using M-PFC. Results A robust HTS assay was developed to identify small molecules that inhibit ESX-1 secretion without impairing bacterial growth in vitro. A hit named IMB-BZ specifically inhibits the secretion of CFP-10 and reduces virulence in an ESX-1-dependent manner, therefore resulting in significant reduction in intracellular and in vivo survival of mycobacteria. Blocking the CFP-10-EccCb1 interaction directly or indirectly underlies the inhibitory effect of IMB-BZ on the secretion of CFP-10. Importantly, our finding shows that the ESX-1 inhibitors pose low risk of drug resistance development by mycobacteria in vitro as compared with traditional anti-TB drug, and exhibit high potency against chronic mycobacterial infection. Conclusion Targeting ESX-1 may lead to the development of novel therapeutics for tuberculosis. IMB-BZ holds the potential for future development into a new anti-TB drug.

Chick embryo fibroblasts infected in vitro with Rous sarcoma virus have properties similar to tumor cells when injected into virus-immune chickens. When such virus-transformed fibroblasts are injected into normal chickens, they apparently participate in the production of tumors independent of their release of virus and are thus apparently malignant in vivo.

Abstract Four methods for measuring the survival of homologous platelets in rabbits were studied: (1) transfusion of nonradioactive platelet concentrates to thrombocytopenic recipients, (2) transfusion of concentrates of platelets labeled in vitro with Cr51-sodium chromate, (3) transfusion of concentrates of platelets labeled in vivo with P32-orthophosphate and (4) transfusion of whole blood labeled in vivo with P32-orthophosphate. The survival time of platelets in normal rabbits was 3-4 days. From comparison of the 3 methods using platelet concentrates, the following conclusions were drawn. (1) All the platelets in a platelet concentrate were capable of recirculating after transfusion. (2) Labeling with P32 or Cr51 did not damage platelets. (3) About one-third of the Cr51 was immediately eluted from viable platelets after they were transfused. (4) Further exchange of the label in vivo did not occur to a significant degree with either Cr51 or P32. (5) Cr51 did not elute from platelets during storage of the platelets. (6) Studies of rabbit platelets had applicability in predicting the behavior of human platelets.
