ABSTRACT Twenty Hereford heifers approximately 9 months of age were vaccinated with saline (control) or 2 × 1010 CFU of the Brucella abortus strain RB51 (RB51) vaccine. Immunologic responses after inoculation demonstrated significantly greater (P < 0.05) antibody and proliferative responses to RB51 antigens in cattle vaccinated with RB51 than in the controls. Pregnant cattle received a conjunctival challenge at approximately 6 months of gestation with 107 CFU of B. suis bv. 1 strains isolated from naturally infected cattle. The fluorescence polarization assay and the buffered acid plate agglutination test had the highest sensitivities in detecting B. suis-infected cattle between 2 and 12 weeks after experimental infection. Serologic responses and lymphocyte proliferative responses to B. suis antigens did not differ between control and RB51 vaccinees after experimental infection. No abortions occurred in cattle in either treatment group after challenge, although there appeared to be an increased incidence of retained placenta after parturition in both the control and the RB51 vaccination treatment groups. Our data suggest that the mammary gland is a preferred site for B. suis localization in cattle. Vaccination with RB51 did not reduce B. suis infection rates in maternal or fetal tissues. In conclusion, although B. suis is unlikely to cause abortions and fetal losses in cattle, our data suggest that RB51 vaccination will not protect cattle against B. suis infection after exposure.

A bacteriological and physicochemical study was conducted in the waters of farms in mountainous regions of Cameroon. The different water samples were collected during two campaigns in December 2018 and February 2019. Ten stations representing the drinking water supply points in our study area were identified. The results showed that these waters contained both pathogenic and commensal fecal bacteria. The highest abundances reached 10x103 CFU/100mL for Salmonella sp., 72x103 CFU/100mL for Staphylococcus aureus, 102x103 CFU/100mL for Brucella suis and 40x103 CFU/100mL for Brucella abortus. These abundances were subject to space-time fluctuations. Water contamination by tweezers was not general and was present only in surface waters (rivers and ponds). The water in the ponds, rivers and wells analyzed were all basic, with low mineralization on average. The dissolved oxygen ranged from 37.5 to 70.6%, nitrate from 3.7 to 19.8 mg/L and iron from 0.01 to 3.5 mg/L. Most of the physicochemical parameters were relatively stable during the two campaigns. The degree of correlation between the physicochemical parameters and the abundance dynamics of the isolated bacteria was heterogeneous. This was clearly more pronounced with Salmonella sp. and Brucella suis. This would be due to the fraction of metabolically active cells present when the bacteria are exposed to unfavorable conditions. Correlations with the abiotic factors were less marked with Staphylococcus aureus and Brucella abortus; this would be due to their tolerance to environmental stresses.

Twelve reindeer (Rangifer tarandus) were vaccinated with Brucella abortus strain 19 vaccine and challenge exposed with B. suis type 4 two and one-half months later during mid-gestation. An additional 10 reindeer served as non-vaccinated controls. A sharp serologic titer response was observed in both vaccinates and controls. Brucella suis type 4 was isolated from tissues and blood from most controls (8 of 10, and 7 of 10 respectively). Seven of 11 vaccinated cows aborted, gave birth to weak fawns that died, or were not pregnant at the completion of the experiment. Brucella suis type 4 was isolated from the tissue of 4 of 12 vaccinates at necropsy. It was concluded that, under the conditions of this experiment, B. abortus strain 19 vaccine in reindeer did not provide adequate protection against challenge exposure with virulent B. suis type 4 organisms.
