Increasing the species diversity in the Aspergillus section Nidulantes: Six novel species mainly from the indoor environment

A novel species in Aspergillus section Fumigati is described as Aspergillus sibiricus. The new species was isolated from an open coal mine in Altai, Siberia, Russia and is represented by ex-type strain CBS 143307. We used a polyphasic taxonomic approach to characterise the species. A description based on morphological features is provided and shows that the new species differs morphologically from closely related species (A. assulatus, A. nishimurae and A. waksmanii). Partial sequences of the loci encoding β-tubulin (BenA), calmodulin (CaM), ribosomal polymerase II second largest subunit (RPB2) and internal transcribed spacer rDNA region (ITS1-5.8S-ITS2) were analyzed as well. Sequences data, macro- and micromorphological and physiological characteristics distinguish A. sibiricus from all known species in section Fumigati, series Unilaterales. A. sibiricus, isolated from an acidic habitat (pH 3.2–3.4), is able to grow at pH 2 and is considered an acid-resistant species.

Entoloma subgenus Claudopus is widely distributed, yet the taxonomy and systematics of its species are still poorly documented. In the present study, more than forty collections of Claudopus were gathered in China and subsequently analysed, based on morphological and molecular data. The results revealed first a high level of species diversity of Claudopus in China and second, there is a wide ecological range regarding the substrates and the habitats ranging from temperate, tropical to subalpine locations. Based on morphological and molecular evidence, five novel species from China are proposed, viz. E. conchatum, E. flabellatum, E. gregarium, E. pleurotoides and E. reductum. Molecular phylogeny of Entoloma s.l. was also reconstructed, based on 187 representatives of Entoloma s.l. by employing the combined ITS, LSU, mtSSU and RPB2 sequences. Ten monophyletic clades (Claudopus, Leptonia, Nolanea, Cuboid-spored Inocephalus, “Alboleptonia”, Cyanula, Pouzarella, Rhodopolia, Prunuloides and Rusticoides) were recovered, while 13 taxa could not be placed in any defined clades. The results confirmed that Claudopus in a traditional morphological sense is not monophyletic and the Rusticoides-group, previously considered within Claudopus, formed a separate clade; but section Claudopus and relatives of E. undatum belong to a distinctive monophyletic group. Despite some monophyletic groups in Entoloma s.l. being distinctive in both morphology and molecular phylogeny, they were still treated as subgenera of Entoloma s.l. temporarily, because accepting them as genera will make Entoloma s.l. paraphyletic.

The article presents the results of mycological analysis of samples taken from open surfaces in residential and working areas of the Arctic and Antarctic Research Institute (AARI) station on the Bolshevik island of the Northern Land archipelago. 114 samples were studied during the expedition work from 2014 to 2016. In total, 47 species of micromycetes were detected in the course of the research. Most of the species (89%) were identified as Ascomycetes. Mucor group of fungi was composed 11% out of all the identified species. The genus Penicillium was the richest in species diversity (16 species). On the surface of materials in places of the increased moisture, there was an open growth of colonies of the Penicillium species. P. expansum was the dominant one, being noted during all the years of observations with the occurrence of about 60%. Among the frequently occurring species, there were P. brevicompactum, P. herqueri, P. purpurogenum and P. waksmanii, which were also noted during all the years of observation (the total occurrence exceeded 15%, although in some years it was much higher). Dark-colored fungus Cladosporium cladosporioides was also dominated species, which in some cases formed black mold on the surface of the materials. Among the potential pathogens, 41 species were registered. A comparison of the data obtained over the years indicates in 2016 the largest number of microfungi to be detected in the indoor environment of AARI station (35 species), whereas in 2014 and 2015 only 29 species of fungi were identified. The similarity of the identified groups of fungi over the years turned out to be quite high. Approximately 36% of microfungi (17 species) were observed annually, although their occurrence over the years varied significantly. Locally accumulation of micromycetes was noted in the indoor environment: household rooms, storage places, workrooms. In these cases, the microfungi colonized the colorful coating, synthetic materials, plywood. The formation of mycobiota in the studied areas is due to anthropogenic invasion of microfungi.

We investigated ochratoxin A (OTA) contamination in raisin samples purchased from Slovak markets and determined the diversity of black-spored aspergilli as potential OTA and fumonisin (FB1 and FB2) producers. The taxonomic identification was performed using sequences of the nuclear ITS1-5.8s-ITS2 region, the calmodulin and beta-tubulin genes. We obtained 239 isolates from eight fungal genera, of which 197 belonged to Aspergillus (82%) and 42 strains (18%) to other fungal genera. OTA contamination was evidenced in 75% of the samples and its level ranged from 0.8 to 10.6 µg/kg. The combination of all three markers used enabled unambiguous identification of A. carbonarius, A. luchuensis, A. niger, A. tubingensis and A. welwitschiae. The dominant coloniser, simultaneously having the highest within-species diversity isolated from our raisin samples, was A. tubingensis. Out of all analysed strains, only A. carbonarius was found to produce OTA, but in relatively high quantity (2477–4382 µg/kg). The production of FB1 and FB2 was evidenced in A. niger strains only.
