An enhanced trunk injection formulation of potassium phosphite against chestnut ink disease

Recently in Florida, foliar treatments using products with the antibiotics oxytetracycline and streptomycin have been approved for the treatment of citrus Huanglongbing (HLB), which is caused by the putative bacterial pathogen ‘Candidatus Liberibacter asiaticus’. Herein, we assessed the levels of oxytetracycline and ‘Ca. L. asiaticus’ titers in citrus trees upon foliar applications with and without a variety of commercial penetrant adjuvants and upon trunk injection. The level of oxytetracycline in citrus leaves was measured using an oxytetracycline ELISA kit and ‘Ca. L. asiaticus’ titer was measured using quantitative PCR. Low levels of oxytetracycline were taken up by citrus leaves after foliar sprays of oxytetracycline in water. Addition of various adjuvants to the oxytetracycline solution showed minimal effects on its uptake by citrus leaves. The level of oxytetracycline in leaves from trunk-injected trees was higher than those treated with all foliar applications. The titer of ‘Ca. L. asiaticus’ in the midrib of leaves from trees receiving oxytetracycline by foliar application was not affected after four days and thirty days of application, whereas the titer was significantly reduced in oxytetracycline-injected trees thirty days after treatment. Investigation of citrus leaves using microscopy showed that they are covered by a thick lipidized cuticle. Perforation of citrus leaf cuticle with a laser significantly increased the uptake of oxytetracycline, decreasing the titer of ‘Ca. L. asiaticus’ in citrus leaves upon foliar application. Taken together, our findings indicate that trunk injection is more efficient than foliar spray even after the use of adjuvants. Our conclusion could help in setting useful recommendations for the application of oxytetracycline in citrus to improve tree health, minimize the amount of applied antibiotic, reduce environmental exposure, and limit off-target effects.

Citrus huanglongbing (HLB) or greening is a devastating bacterial disease that has destroyed millions of trees and is associated with phloem-residing ‘Candidatus Liberibacter asiaticus’ (Las) in Florida. In this study, we evaluated the spatiotemporal dynamics of oxytetracycline in planta and its control effect against HLB via trunk injection. Las-infected ‘Hamlin’ sweet orange trees on ‘Swingle’ citrumelo rootstock at the early stage of decline were treated with oxytetracycline hydrochloride (OTC) using trunk injection with varying number of injection ports. Spatiotemporal distribution of OTC and dynamics of Las populations were monitored by high-performance liquid chromatography method and qPCR assay, respectively. Uniform distribution of OTC throughout tree canopies and root system was achieved 2 days postinjection. High levels of OTC (>850 µg/kg) were maintained in leaf and root for at least 1 month and moderate OTC (>500 µg/kg) persisted for more than 9 months. Reduction of Las populations in root system and leaves of OTC-treated trees were over 95% and 99% (i.e., 1.76 and 2.19 log reduction) between 2 and 28 days postinjection. Conditions of trees receiving OTC treatment were improved, fruit yield was increased, and juice acidity was lowered than water-injected control even though their differences were not statistically significant during the test period. Our study demonstrated that trunk injection of OTC could be used as an effective measure for integrated management of citrus HLB.

Downy mildew of collard occurs frequently in the southeastern United States, and fungicides have become an essential part of economical control strategies for the disease. Fungicides were evaluated in 2007 and 2008 either alone or combined with a reduced rate (2 pt/acre) of potassium phosphite. Two formulations of potassium phosphite also were tested alone at the full rate (4 pt/acre). Presidio, Presidio plus Pro-Phyt (2007) or K-Phite (2008), K-Phite, Amistar plus ProPhyt, and Aliette reduced downy mildew severity at the final rating and also reduced AUDPC over the two years. Presidio plus potassium phosphite and Amistar plus ProPhyt increased the weight of healthy leaves and stems over the water control and also increased economic return calculated after fungicide and other production costs were subtracted from the crop value. The yields with Ranman, Sonata, and Presidio applied in combination with potassium phosphite were 16% greater than with the fungicides applied alone. Potassium phosphite may be useful in a tank-mix with other fungicides applied to collard to prevent yield loss to downy mildew. Accepted for publication 7 July 2010. Published 23 August 2010.

The plant disease Moko, caused by Ralstonia solanacearum, is the most important bacterial disease in banana and plantain crops worldwide. In the present study, chlorine dioxide and seven resistance inducers in banana plants (Musa sp.) infected with this bacterium were evaluated under greenhouse conditions. For the evaluation of chlorine dioxide, three doses were used (10, 30 and 50 mg L-1). The evaluation of the resistance inducers included the following: sodium salicylate 0.4 g L-1; hydrogen peroxide 1 mM; potassium phosphite 1.5 mL L-1; 3-aminobutanoic acid 1.0 g L-1; methyl jasmonate 0.2 g L-1; acibenzolar-s-methyl 0.3 mL L-1 and chitosan 3.0 mg mL-1. The results showed a significant reduction of 74% in the area under the disease progress curve (AUDPC) value, which was calculated for the disease development when the injected chlorine dioxide dose was 50 mg L-1. The AUDPC value for the resistance inducers was reduced by 45.4% for chitosan, 75.5% for methyl jasmonate and 65.5% for 3-aminobutanoic acid. Therefore, the results indicated that these molecules have the potential to be used for control of the Moko disease.
