SARS-CoV-2 surveillance is crucial to identify variants with altered epidemiological properties. Wastewater-based epidemiology (WBE) provides an unbiased and complementary approach to sequencing individual cases. Yet, national WBE surveillance programs have not been widely implemented and data analyses remain challenging. We deep-sequenced 2,093 wastewater samples representing 95 municipal catchments, covering >57% of Austria's population, from December 2020 to September 2021. Our Variant Quantification in Sewage pipeline designed for Robustness (VaQuERo) enabled us to deduce variant abundance from complex wastewater samples and delineate the spatiotemporal dynamics of the dominant Alpha and Delta variants as well as regional clusters of other variants of concern. These results were cross validated by epidemiological records of >130,000 individual cases. Finally, we provide a framework to predict emerging variants de novo and infer variant-specific reproduction numbers from wastewater. This study demonstrates the power of national-scale WBE to support public health and promises particular value for countries without dense individual monitoring.

AimsDiabetes-related cerebral microangiopathy can manifest as cerebral small vessel disease (CSVD) and exhibit cognitive decline. To find the early change of function in advance, this study examined the spatiotemporal dynamics of cerebral vascular permeability (Ktrans) in the progression of type 2 diabetes mellitus (T2DM).MethodsKtrans was cross-sectionally measured in T2DM and non-diabetes groups with or without CSVD using dynamic contrast-enhanced MRI (DCE-MRI).ResultsIn all patients with T2DM, the Ktrans of white matter (WM) was increased, whereas the Ktrans of gray matter (GM) was increased only in T2DM with CSVD. The involvement of WM was earlier than GM and was before the CSVD features could be visualized on MRI. Among the commonly available four CSVD items of MRI, microbleeds were the most sensitive, indicating the increased permeability in all patients. Increased Ktrans in T2DM was more associated with moderate WM hyperintensity but less with the presence of lacunae or multiple perivascular spaces, in contrast to patients without diabetes. The differential correlation suggested distinct mechanisms underlying diabetes-related CSVD and other CSVDs.ConclusionsThis study highlights the early development of cerebral microangiopathy with increased BBB leakage in T2DM, before the CSVD features can be visualized on MRI. The results may increase the proactivity of clinicians in recognizing the subsequent neurological comorbidities.

Estimating the grassland carbon budget is critically important for ensuring that grassland resources are used sustainably. However, the spatiotemporal dynamics of the carbon budget and the response to grazing have not yet been characterized in Qinghai grasslands. Here, we estimated the gross primary productivity (GPP) and net ecosystem exchange (NEE) in Qinghai grasslands using the improved Biome-BGCMuSo model to characterize the spatiotemporal dynamics of the carbon budget and the response to grazing in this region from 1979 to 2018. The GPP of Qinghai grasslands fluctuated during the study period, with an average annual value of 118.78 gC/m2. The NEE of Qinghai grasslands fluctuated from 1979 to 2018, with an average value of −5.16 gC/m2. After 2,000, GPP increased, and NEE decreased in a fluctuating manner. There were clear regional differences in GPP and NEE. GPP was low in most areas of Qinghai, and GPP was high in eastern and southern Qinghai. The southern, southeastern, and northeastern parts of Qinghai were mainly carbon sinks, and the northwestern part of Qinghai and the region between the southeastern and northeastern parts of Qinghai were mainly carbon sources. Grazing generally decreased GPP and increased NEE in Qinghai grasslands from 1979 to 2018. There was spatial heterogeneity in the effect of grazing on GPP and NEE. Under grazing, GPP and NEE were significantly decreased mainly in eastern Qinghai, and GPP and NEE were significantly increased mainly in southern and eastern Qinghai. NEE was most affected by grazing in eastern Qinghai. The results of this study aid our understanding of the mechanism driving variation in the grassland carbon budget and provide new data that could be used to support local grassland management.

AbstractThe bromate–sulfite reaction-based pH-oscillators represent one of the most useful subgroup among the chemical oscillators. They provide strong H+-pulses which can generate temporal oscillations in other systems coupled to them and they show wide variety of spatiotemporal dynamics when they are carried out in different gel reactors. Some examples are discussed. When pH-dependent chemical and physical processes are linked to a bromate–sulfite-based oscillator, rhythmic changes can appear in the concentration of some cations and anions, in the distribution of the species in a pH-sensitive stepwise complex formation, in the oxidation number of the central cation in a chelate complex, in the volume or the desorption-adsorption ability of a piece of gel. These reactions are quite suitable for generating spatiotemporal patterns in open reactors. Many reaction–diffusion phenomena, moving and stationary patterns, have been recently observed experimentally using different reactor configurations, which allow exploring the effect of different initial and boundary conditions. Here, we summarize the most relevant aspects of these experimental and numerical studies on bromate–sulfite reaction-based reaction–diffusion systems.
