Since 2017, a new leaf wilt syndrome was observed in plantations of date palm in Tunisia. Its incidence increases sharply from year to year, especially in ‘Deglet Nour’ trees, aged between 5 and 15 years. In severe cases, the large number of dried leaves per tree can lead to complete cessation of date production. Symptoms appear on one or more leaves in the center of the crown. Whitening and drying start at the top of the leaflets and proceed to their base, while the midrib remains green. Then the whole leaf dies. Small white-creamy leaflet fragments and roots were collected from five different regions in the Djerid Oases. They were disinfected with diluted bleach (0,8 % NaOCl) and ethanol (80%) (each 2 min), rinsed with sterile distilled water, dried and finally plated in Petri dishes containing Potato Dextrose Agar (PDA) amended with 50mg/l neomycin. After incubation for 7 days at 25ºC±2, emerging fungal colonies were single-spored by serial dilution. They were transferred to PDA, Carnation Leaf Agar (CLA) and Spezieller Nahrstoffarmer Agar (SNA) for morphological identification. Based on the colony color on PDA, conidial morphology and phialide structures on CLA and/or SNA, of the 85 Fusarium isolates, around 90% were identified as F. proliferatum and around 10% as F. brachygibbosum (Leslie and Summerell, 2006). Fusarium proliferatum colonies rapidly developed white aerial mycelium that became purple in old cultures. Microconidia were abundant in the aerial mycelium and formed chains of variable length, on monophialides and polyphialids, a characteristic that distinguishes F. proliferatum from F. verticilloides. Less often, they were observed in false heads. Chlamydospores were absent. On CLA, microconidia were mostly 2 × 15 µm in size, a large number of sickle shaped macroconidia (2 × 25 µm) had one septum, some were larger (2 × 50 µm) with 3 septa and tips at both ends. Molecular identification was carried out based on elongation factor (EF-1α) gene sequencing. The region between the EF1 and EF2 primers (O’Donnell et al., 1998) was amplified and the sequences were compared to Fusarium reference sequences (GenBank). The sequences of the isolates Fus 1953 (539 bp), Fus 1962 (618 bp), and Fus 1965 (605 bp) shared respectively 100%, 99.51% and 99.51% homology with that of F. proliferatum JF740713.1 and were deposited in GenBank with the following accession numbers: MT630418, MT630419, and MT630420, respectively. The sequences of isolates 7F, 28F, Fus 1955 and Fus 1956 shared 100 % homology with that of F. brachygibbosum (GQ505418.1) while those of Fus 1955 and Fus 1956 showed 99.02 and 98.91 % identity, respectively, with F. brachygibbosum JX118981.1. The sequences of 7F (535 bp), 28F (535 bp), Fus 1955 (608 bp), and Fus 1956 (647 bp) were deposited in GenBank with the following accession numbers: MT630409, MT630410, MT630411, and MT630412, respectively. Two ml suspension of 106 conidia / ml of each isolate was sprayed separately or in combinations on in vitro cloned ‘Deglet Nour’ plants, placed in a greenhouse at 28°±2 °C and 70% R.H.. Isolates of F. proliferatum led to dryness and wilting leaflets after 3 weeks. Fusarium brachygibbosum only induced mild leaf yellowing, while in combination they were more virulent. Fungal isolates of both species were re-isolated and their identity confirmed to be the same of those isolated from leaflets infected in the open field, confirming Koch’s postulates. Control plants lacked symptoms. Fusarium proliferatum is known as date palm pathogen in many countries (Saleh et al. 2017), however, to our knowledge, this is the first report of F. proliferatum and also F. brachygibbosum causing Leaf Wilt symptoms on P. dactylifera in Tunisia.

A survey of postharvest losses of commercially handled and cold-stored fruit of fresh date palm (Phoenix dactylifera L.), cvs. Medjool and Hayani, was conducted in the 2009 and 2010 seasons in the grove of Elx (Alacant Province, Southeast Spain). Disease symptoms consisting of circular, light brown, soft spots located in any part of the fruit skin were observed in 2 to 5% of the fruit. At room temperature, the lesions expanded rapidly and blue mold symptoms were apparent. The potential causal agent (isolate IVIA NiAA-2) was transferred to PDA and incubated at 25°C. The identification was performed at the Spanish Type Culture Collection (CECT, University of Valencia, Spain) based on colony morphology of the isolate grown on Czapeck yeast extract agar (CYA) and malt extract agar (MEA) at 26°C. Colonies were circular (average diameter of 40 mm at 7 days), radially sulcate, with dense velvety white mycelium, and very abundant, bluish green conidia. The underside of the plates showed light brown and pale green colonies on CYA and MEA, respectively. On CYA, but not on MEA, a light yellow exudate was produced and a brownish pigment diffused into the medium. At 5 and 37°C on CYA, white microcolonies and no colonies were observed, respectively. Conidia were ellipsoidal to subglobose, smooth and thin walled, measuring 3.0 to 3.5 × 2.5 to 3.0 μm (n = 50) (4). Based on these morphological characteristics, the isolate IVIA NiAA-2 was tentatively identified as Penicillium expansum L. To confirm the identity, we amplified and sequenced the rDNA internal transcribed spacer (ITS) region with primers ITS1 and ITS4 (GenBank Accession No. KC169942). A BLAST search showed 99% identity and 100% query coverage with P. expansum strain NRRL 6069 (DQ339562) (2). Selected healthy dates cv. Medjool were surface disinfected by dipping in 0.5% sodium hypochlorite for 2 min followed by thorough rinsing in deionized water. Pathogenicity was tested by pipetting 20 μl of a spore suspension (1 × 106 spores per ml), prepared from 7-day PDA cultures, onto fresh skin wounds, which were made on disinfected fruit using a sterile, stainless steel rod with a probe tip 1 mm in width × 2 mm in length (one wound on each of nine dates, incubated in one humid chamber). Disinfected, wounded, and non-inoculated dates were used as controls. The procedure was repeated three times. Disease symptoms were observed on all inoculated fruit (average lesion size of 6, 15, and 22 mm after 4, 7, and 10 days of incubation at 20°C, respectively) and P. expansum was consistently reisolated, thereby fulfilling Koch's postulates. No decay was observed on any of the non-inoculated fruit. Unidentified species of Penicillium have been reported to cause date palm fruit rot (1,3). Pygmy date palm (Phoenix roebelenii O'Brien), native to Laos and southeast Asia, is one of the most commonly cultivated date palms both indoors and out. In early April 2003, a new and widespread leaf disease was detected on 2- to 3-feet high, potted, pygmy date palms growing in open fields or shade-houses in four commercial nurseries of the eastern Sicily Region of southern Italy. Initial symptoms on infected plants were small, chlorotic spots on blades and edges of leaflets. As spots enlarged, yellow or reddish-brown margins and brown or gray, wrinkled, sunken centers developed. Sometimes, spots or lesions coalesced to cause blight of young expanding leaves or death of apical buds. Botrytis cinerea Pers.:Fr was recovered consistently from sections of infected tissues (disinfected for 1 min in 1% NaOCl and rinsed in sterile water) plated on malt extract agar. On potato dextrose agar (PDA), colonies of B. cinerea were first hyaline, then turned white, and later turned dark gray when spores differentiated. Six- to eight-day-old-cultures developed white sclerotia that turned black after three more days. Conidia in 1-month-old-cultures were hyaline or gray, ovoid, and ranged from 5.2 to 8.8 × 8 to 14 μm (average 6.6 × 9.9 μm). Black microsclerotia were round or irregular in shape and ranged from 0.6 to 2.8 × 0.3 to 2.5 mm (average 1.66 × 0.98 mm) (1). Koch's postulates were performed by spraying potted, 9-month-old pygmy date palms (2 feet high) with a spore suspension (1 × 106 CFU per ml from 15-day-old cultures grown on PDA). Six plants were wounded and inoculated, while six plants were inoculated without wounding. An equal number of noninoculated plants sprayed with sterile water served as controls. All plants were maintained in a humid environment at 20°C. Sunken-leaf and rachis lesions were observed on all wounded and most nonwounded plants within 10 to 14 days after inoculation. Symptoms did not develop on the control plants. Koch's postulates were fulfilled by reisolation of the fungus from affected tissues. A field survey revealed the occasional presence of similar leaf spots on canary island date palms (Phoenix canariensis Hort. ex Chabaud), from which B. cinerea was consistently recovered. The unusually cool and humid weather conditions recorded in Sicily during this disease outbreak were very conducive for occurrence of the B. cinerea infections. Although the disease only occasionally caused death of plants, evidence indicated that B. cinerea reduced commercial value of infected pygmy date palms. B. cinerea was previously recorded in northern Italy on canary island date palm (2) and was recently detected in southern Italy on majesty palm (3). To our knowledge, this is the first report of leaf spot and rachis blight caused by B. cinerea on pygmy date palm. References: (1) M. B. Ellis. Dematiaceous Hyphomycetes. During the survey of the forest fungal disease, of Jalgaon district, two severe leaf spot diseases on Lannae coromandelica and ( Ougenia dalbergioides (Papilionaceae) were observed in Jalgaon, forest during July to September 2016-17. The casual organism was identified as Stigmina lanneae and Phomopsis sp. respectively1-4,7. These are first report from Jalgaon and Maharashtra state.
