An alkaliphilic actinomycete, strain YIM 80305T, which was isolated from a muddy sample in Chaka salt lake, Qinghai Province of China, was characterized using a polyphasic approach. The isolate produced light-yellow substrate and yellow–white aerial mycelia on most tested media. Optimum pH for growth was 9·0–10·0 with scant growth at pH 7·0. Results showed that strain YIM 80305T was obligately Na+-dependent, and showed sensitivity to K+. The DNA G+C content was 70·5 mol%. 16S rRNA gene sequence analysis together with these characteristics consistently assigned strain YIM 80305T to the genus Streptomyces. It formed a distinct clade based on analyses of the almost-complete and 120-nucleotide variable γ region of the 16S rRNA gene. It could be differentiated by phenotypic and genotypic analysis from all the Streptomyces species whose names have been validly published. On the basis of polyphasic evidence, Streptomyces sodiiphilus sp. nov. is proposed. The type strain is YIM 80305T (=CCTCC AA 203015T=CIP 107975T).

A novel marine bacterium, strain KMD 001T, was isolated from the starfish Asterias amurensis, which inhabits the East Sea of Korea. Strain KMD 001T was aerobic, light-yellow pigmented and Gram-stain-negative. Analyses of the 16S rRNA gene sequence revealed that strain KMD 001T represents a novel lineage within the class Gammaproteobacteria. Strain KMD 001T is closely related to the genera Endozoicomonas and Zooshikella, which belong to the family Hahellaceae and to the order Oceanospirillales. The 16S rRNA gene sequence of strain KMD 001T shows similarities of approximately 91.8–94.6 % with the above-mentioned genera. The DNA G+C content of KMD 001T is 47.6 mol%. It contains Q-9 as the major isoprenoid quinone. The predominant fatty acids were determined to be anteiso-C15 : 0, iso-C15 : 0, iso-C14 : 0 and iso-C16 : 0. Strain KMD 001T should be assigned to a novel bacterial genus within the class Gammaproteobacteria based on its phylogenetic, chemotaxonomic and phenotypic characteristics. The name Kistimonas asteriae gen. nov., sp. nov. is proposed. The type strain is KMD 001T (=KCCM 90076T =JCM 15607T).

A yellowish-brown bacterium was isolated from enrichment cultures inoculated with seawater samples from the eastern coast of India (Visakhapatnam) under photoheterotrophic conditions. Enrichment and isolation in a medium containing 2 % NaCl (w/v) yielded strain JA128T, which has ovoid to rod-shaped cells, also forms chains and is non-motile. Phylogenetic analysis on the basis of 16S rRNA gene sequences showed that strain JA128T clusters with the Alphaproteobacteria and the sequence similarity with its closest relatives, Rhodovulum iodosum and Rhodovulum sulfidophilum, was 95 %. Strain JA128T contained vesicular intracytoplasmic membranes, bacteriochlorophyll a and carotenoids of the spheroidene series. Strain JA128T was mesophilic, slightly acidophilic, slightly halophilic and grew photoheterotrophically with a number of organic compounds as carbon source and electron donor. It was unable to grow photoautotrophically, chemoautotrophically or by fermentative modes. It did not utilize sulfide, thiosulfate or hydrogen as electron donors. Thiamine was required as a growth factor. Based on the 16S rRNA gene sequence analysis, morphological and physiological characteristics, strain JA128T was significantly different from other species of the genus Rhodovulum and was recognized as a novel species for which the name Rhodovulum marinum sp. nov. is proposed. The type strain is JA128T (=ATCC BAA 1215T=CCUG 52183T=JCM 13300T).

A halophilic, aerobic bacterium, designated PO-M2T, was isolated from seashore sand, from Pohang, Korea and characterized on the basis of physiological and biochemical features. Phylogenetic analysis of 16S rRNA gene sequences revealed a clear affiliation of the novel strain with members of the genus Idiomarina. Sequence similarities between strain PO-M2T and the type strains of species belonging to the genus Idiomarina ranged from 94.3 to 95.5 %. Cells of strain PO-M2T were straight or slightly curved rods and formed light-yellow colonies on marine agar medium. The major isoprenoid quinone was ubiquinone (Q-8) and the predominant cellular fatty acids were C15 : 0 iso (19.3 %), C17 : 1 ω9c iso (11.9 %), C17 : 0 iso (10.9 %), C18 : 1 ω7c (10.4 %), C16 : 0 (9.0 %) and C16 : 1 ω7c and/or C15 : 0 iso 2-OH (7.2 %). The G+C content of the DNA was 45.1 mol%. Based on physiological, biochemical and chemotaxonomic traits and comparative 16S rRNA gene sequence analysis, it is demonstrated that the isolate represents a novel species of the genus Idiomarina, for which the name Idiomarina homiensis sp. nov. is proposed. The type strain is PO-M2T (=KACC 11514T=DSM 17923T).

The actinobacterial strain YIM 70185T was isolated from a saline soil sample collected from Qinghai province, north-west China, and subjected to a taxonomic investigation. Phylogenetic analysis based on 16S rRNA gene sequences revealed 93.5–96.4 % similarity to members of related genera in the family Micrococcaceae. In the phylogenetic dendrogram based on 16S rRNA gene sequence analysis, strain YIM 70185T formed a separate clade next to the genera Micrococcus and Citricoccus within the family Micrococcaceae. The peptidoglycan type was A4α, l-lys–l-ala–l-Glu. Cell-wall sugars contained glucose and tyvelose. The polar lipids were phosphatidylglycerol, diphosphatidylglycerol, phosphatidylinositol, an unknown phospholipid and an unknown glycolipid. The menaquinones were MK-9, MK-10 and MK-8 (molar ratio 5 : 2 : 1). The major fatty acids were ai-C15 : 0 and i-C15 : 0 and the DNA G+C content was 66.5 mol%. These chemotaxonomic profiles supported the assignment of strain YIM 70185T to a novel genus within the family Micrococcaceae. The name Zhihengliuella halotolerans gen. nov., sp. nov. is proposed. The type strain of Zhihengliuella halotolerans is YIM 70185T (=DSM 17364T=KCTC 19085T).

A Gram-negative, short rod-shaped micro-organism (CC-BB4T) was isolated on nutrient agar from soil from Sinshe in Taichung County, Taiwan. Analysis of the 16S rRNA gene sequence demonstrated that this isolate was novel, as it showed <92 % similarity to the sequences of species of the genera Labrys, Beijerinckia and Methylocystis. The micro-organism did not utilize methylamine or methanol as a substrate, but was able to use several organic acids. The fatty acid profile was different from those reported for the genera Labrys, Beijerinckia, Methylocystis, Angulomicrobium, Methylorhabdus and Methyloarcula. On the basis of the 16S rRNA gene sequence analysis, in combination with chemotaxonomic and physiological data, strain CC-BB4T represents a novel genus, for which the name Pseudolabrys gen. nov. is proposed. The type species is Pseudolabrys taiwanensis sp. nov., and the type strain of P. taiwanensis is CC-BB4T (=CCUG 51779T=CIP 108932T).

Strain EB21T was isolated from a brine sample from Aran-Bidgol salt lake, a saline playa in Iran. Strain EB21T was an orange–red-pigmented, motile rod and required at least 2 M NaCl but not MgCl2 for growth. Optimal growth was achieved at 3.5 M NaCl and 0.2 M MgCl2. The optimum pH and temperature for growth were pH 7.5 and 40 °C, while it was able to grow at pH 6.0–8.0 and 25–55 °C. Analysis of the 16S rRNA gene sequence revealed that strain EB21T is a member of the family Halobacteriaceae , showing low levels of similarity to other members of the family. The highest sequence similarities, 91.8, 91.7 and 91.5 %, were obtained with the 16S rRNA gene sequences of the type strains of Halobiforma lacisalsi , Haloterrigena thermotolerans and Halalkalicoccus tibetensis , respectively. Polar lipid analyses revealed that strain EB21T contains phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and phosphatidylglycerol sulfate. Three unidentified glycolipids and one minor phospholipid were also observed. The only quinone present was MK-8(II-H2). The G+C content of its DNA was 67.7 mol%. On the basis of the data obtained, the new isolate could not be classified in any recognized genus. Strain EB21T is thus considered to represent a novel species in a new genus within the family Halobacteriaceae , order Halobacteriales , for which the name Haloarchaeobius iranensis gen. nov., sp. nov. is proposed. The type strain of Haloarchaeobius iranensis is EB21T ( = IBRC-M 10013T  = KCTC 4048T).

hsp60 gene sequences were determined for members of the genus Bacteroides and sequence similarities were compared with those obtained for the 16S rRNA gene. Among the 29 Bacteroides type strains, the mean sequence similarity of the hsp60 gene (84.5 %) was significantly less than that of the 16S rRNA gene (90.7 %), indicating a high discriminatory power of the hsp60 gene. Species of the genus Bacteroides were differentiated well by hsp60 gene sequence analysis, except for Bacteroides pyogenes JCM 6294T, Bacteroides suis JCM 6292T and Bacteroides tectus JCM 10003T. The hsp60 gene sequence analysis and the levels of DNA–DNA relatedness observed demonstrated that these three type strains are a single species. Consequently, B. suis and B. tectus are heterotypic synonyms of B. pyogenes. This study suggests that the hsp60 gene is an alternative phylogenetic marker for the classification of species of the genus Bacteroides.

A Gram-negative, motile, neutrophilic and extremely halophilic strain, AJ5T, was isolated from a salt lake in Xinjiang, China, and subjected to polyphasic taxonomic study. The major polar lipids of the isolate were C20C20 and C20C25 derivatives of phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and glycolipid. The DNA G+C content was 64·9 mol%. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain AJ5T clustered with members of the genus Halobiforma, exhibiting high sequence similarity to the 16S rRNA gene sequences of Halobiforma nitratireducens (96·3 %) and Halobiforma haloterrestris (99·0 %). Comparative analysis of phenotypic characteristics and DNA–DNA hybridization between strain AJ5T and Halobiforma species supported the conclusion that AJ5T represents a novel species within this genus, for which the name Halobiforma lacisalsi sp. nov. is proposed. The type strain is AJ5T (=CGMCC 1.3738T=JCM 12983T).

A thorough phenotypic and genotypic analysis of 150 strains belonging to the Mycobacterium terrae complex resulted in the identification of a number of previously unreported sequevars (sqvs) within the species known to belong to the complex. For the species Mycobacterium arupense , three sqvs were detected in the 16S rRNA gene, six sqvs in the hsp65 gene and 15 sqvs in the rpoB gene; in Mycobacterium senuense two sqvs were present in each of the three genetic regions; in Mycobacterium kumamotonense four, two and nine sqvs were found, respectively, and in M. terrae three, four and six sqvs were found, respectively. The inappropriate inclusion of Mycobacterium triviale within the M. terrae complex was confirmed. The limited utility of biochemical tests and of mycolic acid analyses for the differentiation of the members of M. terrae complex was also confirmed. The survey allowed the recognition of three previously undescribed species that were characterized by unique sequences in the 16S rRNA, hsp65 and rpoB genes. Mycobacterium engbaekii sp. nov. (proposed previously 40 years ago but never validly published) was characterized by pink photochromogenic pigmentation and rapid growth; phylogenetically it was related to Mycobacterium hiberniae . The type strain of this species, of which eight strains were investigated, is ATCC 27353T ( = DSM 45694T). A cluster of 24 strains was the basis for the description of Mycobacterium heraklionense sp. nov., which has an intermediate growth rate and is unpigmented; nitrate reductase activity is typically strong. Closely related to M. arupense with respect to the 16S rRNA gene, M. heraklionense sp. nov. could be clearly differentiated from the latter species in the other genetic regions investigated. The type strain is NCTC 13432T ( = LMG 24735T = CECT 7509T). Mycobacterium longobardum sp. nov., represented in the study by seven strains, was characterized by a unique phylogenetic location within the M. terrae complex, clearly divergent from any other species. The type strain is DSM 45394T ( = CCUG 58460T).
