Salsuginibacillus kocurii gen. nov., sp. nov., a moderately halophilic bacterium from soda-lake sediment

A Gram-positive, endospore-forming, alkali-tolerant, moderately halophilic bacterium, designated strain CH9dT, was isolated from the sediment of Lake Chagannor in the Inner Mongolia Autonomous Region, China. The cells were rod-shaped and motile. Isolate CH9dT grew at pH 5.8–10.0 (optimally at pH 8.5), at salinities of 3–20 % (w/v) marine salts (optimally at 10.0 %, w/v) and between 20 and 50 °C (optimally at 37 °C). The cell wall contained meso-diaminopimelic acid and the major respiratory isoprenoid quinone was MK-7. The predominant cellular fatty acids of strain CH9dT were anteiso-C15 : 0, anteiso-C17 : 0, iso-C17 : 0 and iso-C15 : 0 and its polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and two unknown phospholipids. The G+C content of the DNA was 44.7 mol%. Strain CH9dT exhibited a 16S rRNA gene sequence similarity value of only 91 % with respect to Thalassobacillus devorans DSM 16966T and showed values below 91 % with respect to members of the genera Bacillus, Halobacillus and Marinococcus. Strain CH9dT could be clearly differentiated from its closest phylogenetic neighbours on the basis of several phenotypic, genotypic and chemotaxonomic features. Therefore, data from the polyphasic study support the placement of strain CH9dT in a novel genus and species, for which the name Salsuginibacillus kocurii gen. nov., sp. nov. is proposed. The type strain is CH9dT (=CCM 7365T=CECT 7154T=CGMCC 1.6287T=DSM 18087T).

A moderately halophilic, Gram-positive, rod-shaped bacterium (BH030004T) was isolated from a solar saltern in Korea. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain BH030004T belonged to the genus Pontibacillus. Chemotaxonomic data (DNA G+C content, 42 mol%; major isoprenoid quinone, MK-7; cell-wall type, A1γ-type meso-diaminopimelic acid; major fatty acids, iso-C15 : 0 and anteiso-C15 : 0) also supported the affiliation of the isolate to the genus Pontibacillus. Although the 16S rRNA gene sequence similarity between strain BH030004T and Pontibacillus chungwhensis DSM 16287T was relatively high (99·1 %), physiological properties and DNA–DNA hybridization (about 7 % DNA–DNA relatedness) allowed genotypic and phenotypic differentiation of strain BH030004T from the type strain of P. chungwhensis. Therefore, strain BH030004T represents a novel species of the genus Pontibacillus, for which the name Pontibacillus marinus sp. nov. is proposed. The type strain is BH030004T (=KCTC 3917T=DSM 16465T).

An aerobic, spore-forming, moderately halophilic bacterium, strain BH260T, was isolated from a salt lake in China. Cells of this strain were found to be motile rods with flagella. The organism grew optimally at 30–32 °C and pH 8·0 in the presence of 12–15 % (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequences showed that the isolate formed a clade with the type strains of the genus Lentibacillus. The levels of 16S rRNA gene sequence similarity for strain BH260T with respect to Lentibacillus salsus KCTC 3911T, Lentibacillus juripiscarius JCM 12147T and Lentibacillus salicampi KCCM 41560T were 96·0, 95·5 and 95·3 %, respectively. Chemotaxonomic data (a DNA G+C content of 44 mol%; MK-7 as the major isoprenoid quinone; A1γ-meso-diaminopimelic acid as the cell-wall type; phosphatidylglycerol and diphosphatidylglycerol as the major polar lipids; anteiso-C15 : 0, anteiso-C17 : 0 and iso-C16 : 0 as the major fatty acids) also supported assignment of the isolate to the genus Lentibacillus. On the basis of physiological and molecular properties, the isolate represents a novel species within the genus Lentibacillus, for which the name Lentibacillus lacisalsi sp. nov. is proposed. The type strain is BH260T (=KCTC 3915T=DSM 16462T).

Strain halo-1T grew in the presence of 9–30 % (w/v) NaCl (optimum 19 %) and at pH 5–10 (optimum pH 9). The cell-wall peptidoglycan contained meso-diaminopimelic acid and the major respiratory isoprenoid quinone was MK-7. The predominant cellular fatty acids of the isolate were anteiso-C15 : 0 (58.35 %), anteiso-C17 : 0 (12.89 %) and C16 : 0 (6.52 %). The polar lipids contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, glycolipid and a phospholipid of unknown structure. The DNA G+C content of the strain was 46.4 mol%. On the basis of 16S rRNA gene sequence similarity, strain halo-1T showed the highest similarity (93.9 %) to Salsuginibacillus kocurii CH9dT. Strain halo-1T could be clearly differentiated from its closest phylogenetic relative on the basis of several phenotypic, genotypic and chemotaxonomic features. Therefore, strain halo-1T represents a novel species, for which the name Salsuginibacillus halophilus sp. nov. is proposed, with the type strain halo-1T (=CGMCC 1.7653T =NBRC 104934T).

A Gram-positive, rod-shaped, spore-forming and moderately halophilic bacterium (strain 8-1T) was isolated from a sediment sample of a neutral salt lake in Xin-Jiang, China. The strain grew optimally at 30 °C, pH 7.0–7.5 and 8–12 % (w/v) NaCl. The major cellular fatty acids were anteiso-C15 : 0, iso-C15 : 0 and anteiso-C17 : 0. The predominant isoprenoid quinone was MK-7 and the cell-wall peptidoglycan contained meso-diaminopimelic acid. Phosphatidylglycerol, diphosphatidylglycerol and two unidentified glycolipids were found to be the major polar lipid components. The genomic DNA G+C content was 43.4 mol%. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that strain 8-1T was closely related to Lentibacillus salarius KCTC 3911T (98.0 % sequence similarity) and other recognized species within the genus Lentibacillus (94.5–95.9 %). The level of DNA–DNA relatedness between strain 8-1T and L. salarius KCTC 3911T was 40 %. Based on the phenotypic and genotypic data presented, strain 8-1T is considered to represent a novel species of the genus Lentibacillus, for which the name Lentibacillus halodurans sp. nov. is proposed. The type strain is 8-1T (=CGMCC 1.3702T=DSM 18342T).

A Gram reaction-positive, moderately halophilic bacterium, designated H57B72T, was isolated from subsurface saline soil of Qaidam basin in the Qinghai province, China. Cells were rod-shaped, strictly aerobic, spore-forming and motile. The isolate grew optimally at 9 % (w/v) NaCl, pH 7.5 and 30 °C. The cell-wall peptidoglycan of strain H57B72T contained meso-diaminopimelic acid as the diagnostic diamino acid. The predominant isoprenoid quinone was MK-7. The major cellular fatty acids were anteiso-C15 : 0 (59.97 %) and anteiso-C17 : 0 (17.14 %). Phosphatidylglycerol, diphosphatidylglycerol and a glycolipid were found to be the predominant polar lipids. The genomic DNA G+C content of strain H57B72T was 37.1 mol%. 16S rRNA gene sequence analysis showed that strain H57B72T was a member of the genus Virgibacillus and was most closely related to Virgibacillus salinus DSM 21756T (98.3 % gene sequence similarity). The level of DNA–DNA relatedness between strain H57B72T and V. salinus DSM 21756T was 8.5 %. Based on the phenotypic, genotypic and phylogenetic data presented, strain H57B72T represents a novel species, for which the name Virgibacillus subterraneus sp. nov. is proposed. The type strain is H57B72T (=DSM 22441T =CGMCC 1.7734T).

An orange-pigmented, Gram-reaction-positive, non-spore-forming, halophilic, alkali-tolerant rod, designated strain halo-2T, was isolated from sediment of Xiarinaoer soda lake, in China’s Inner Mongolia Autonomous Region. Strain halo-2T grew in a complex medium with 3–30 % (w/v) NaCl and at pH 5–10. The cell-wall peptidoglycan contained meso-diaminopimelic acid and the major respiratory isoprenoid quinone was MK-7. The predominant cellular fatty acids were anteiso-C15 : 0 (43.6 %), anteiso-C17 : 0 (14.8 %) and iso-C15 : 0 (6.8 %) and the polar lipids consisted of diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylglycerol. The genomic DNA G+C content of the novel strain was 48.2 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain halo-2T was most closely related to Bacillus agaradhaerens DSM 8721T (93.9 % sequence similarity). However, strain halo-2T could be clearly differentiated from its closest phylogenetic relatives on the basis of several phenotypic, genotypic and chemotaxonomic characteristics. Strain halo-2T therefore represents a novel species in a new genus for which the name Salisediminibacterium halotolerans gen. nov., sp. nov. is proposed. The type strain of the type species is halo-2T ( = CGMCC 1.7654T = NBRC 104935T).

A Gram-stain-positive, rod-shaped, non-sporulating, motile and moderately halophilic bacterium, designated strain H96B60T, was isolated from a saline soil sample of the Qaidam basin, China. The strain was facultatively anaerobic. Major end products formed from glucose fermentation were acetate, ethanol and lactic acid. The cell-wall peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. The isoprenoid quinone component was menaquinone-6 (MK-6). The predominant cellular fatty acids were C16 : 0, anteiso-C13 : 0 and anteiso-C15 : 0. The genomic DNA G+C content of strain H96B60T was 36.2 mol%. Phylogenetic analysis based on comparative 16S rRNA gene sequences indicated that strain H96B60T represented a novel phyletic lineage within the family Bacillaceae and was related most closely to Halolactibacillus species (96.1–96.4 % similarity). Based on the phenotypic, chemotaxonomic and phylogenetic data presented, strain H96B60T is considered to represent a novel species of a new genus, for which the name Streptohalobacillus salinus gen. nov., sp. nov. is proposed. The type strain of Streptohalobacillus salinus is H96B60T ( = DSM 22440T  = CGMCC 1.7733T).

An aerobic, Gram-stain-positive, endospore-forming, rod-shaped and moderately halophilic strain SKP4-6T, was isolated from shrimp paste (Ka-pi) collected from Samut Sakhon Province, Thailand. Phylogenetic analysis revealed that strain SKP4-6T belonged to the genus Halobacillus and was most closely related to Halobacillus salinus JCM 11546T (98.6 %), Halobacillus locisalis KCTC 3788T (98.6 %) and Halobacillus yeomjeoni KCTC 3957T (98.6 %) based on 16S rRNA gene sequence similarity. The digital DNA–DNA hybridization (dDDH) and average nucleotide identity (ANI) values between strain SKP4-6T and its related species were 18.2–19.3 % and 69.84–84.51 %, respectively, which were lower than the threshold recommended for species delineation. The strain grew optimally at 30–40 °C, at pH 7.0 and with 10–15 % (w/v) NaCl. It contained l-Orn–d-Asp in the cell wall peptidoglycan. The DNA G+C content was 44.8 mol%. The major fatty acids were iso-C15 : 0, anteiso-C15 : 0 and anteiso-C17 : 0. The predominant isoprenoid quinone was MK-7. Phosphatidylglycerol and diphosphatidylglycerol were present as major polar lipids. Based on this polyphasic approach, digital DNA–DNA relatedness and ANI values, strain SKP4-6T represents a novel species of the genus Halobacillus , for which the name Halobacillus fulvus sp. nov. is proposed. The type strain is SKP4-6T (=JCM 32624T=TISTR 2595T).

A Gram-stain-negative, aerobic, moderately halophilic bacterium, strain DY53T, was isolated from a deep-seawater sample collected from the eastern Pacific Ocean. This isolate grew in the presence of 0.5–10.0 % (w/v) NaCl, at pH 6.5–8.5 and at 15–40 °C. The optimum NaCl concentration for growth of DY53T was 2 % (w/v) at 35 °C. Chemotaxonomic analysis showed MK-7 as the predominant menaquinone and iso-C15 : 0, summed feature 3 (iso-C15 : 0 2-OH and/or C16 : 1ω7c), iso-C15 : 1 G, iso-C15 : 0 3-OH and iso-C17 : 0 3-OH as major cellular fatty acids. The genomic DNA G+C content was 40.8 mol%. Phylogenetic trees based on 16S rRNA gene sequences revealed that Fabibacter halotolerans UST030701-097T was the closest neighbour, with 96.7 % sequence similarity. Based on phylogenetic, chemotaxonomic and phenotypic data, we propose that strain DY53T represents a novel species of the genus Fabibacter , for which the name Fabibacter pacificus sp. nov. is proposed. The type strain is DY53T( = CGMCC 1.12402T = JCM 18885T).

A moderately halophilic bacterium, strain CG2.1T, isolated from a solar saltern at Cabo de Gata, a wildlife reserve located in the province of Almería, southern Spain, was subjected to a polyphasic taxonomic study. This organism was an aerobic, motile, Gram-negative rod that produced orange-pigmented colonies. Strain CG2.1T was able to grow at salinities of 3–25 % (w/v) and at temperatures of 15–40 °C. The pH range for growth was 5–9. Strain CG2.1T was a heterotroph capable of utilizing various carbohydrates as carbon sources. The organism reduced nitrate and showed phenylalanine deaminase activity. The major fatty acids were C18 : 1 ω7c, C16 : 0 and C19 : 0 cyclo ω8c. The DNA G+C content was 60.9 mol%. On the basis of the phenotypic and phylogenetic data, strain CG2.1T appeared to be a member of the genus Halomonas and clustered closely with Halomonas marisflavi (97.1 % 16S rRNA gene sequence similarity). However, the level of DNA–DNA relatedness between the novel isolate and the most closely related Halomonas species was low. On the basis of these data, strain CG2.1T represents a novel member of the genus Halomonas, for which the name Halomonas indalinina is proposed. The type strain is CG2.1T (=CECT 5902T=LMG 23625T).
