Artificial seed technology is a rapidly growing area of research in plant cell and tissue culture. Application of this technology opens an alternative route for mass scale production, efficient delivery of cloned plantlets and fulfils the increasing demand of local growers. An attempt was made to produce artificial seeds and their subsequent regeneration of a highly valuable medicinal orchid of Nepal i.e. Cymbidium aloifolium. Artificial seeds were obtained through encapsulation of protocorms in calcium alginate beads. Protocorms were encapsulated by using 3% sodium alginate and 0.2 M calcium chloride solution. Murashige and Skoog (MS) medium (1962) was used as the basal medium for in vitro germination and seedling development of artificial seed. In Cymbidium aloifolium, 20-25 days old in vitro grown protocorms were used for production of artificial seeds. Artificial seeds were inoculated on two different culture conditions of MS medium i.e. MS solid & MS liquid with four different treatments i.e. strength of 1.0, ½, ¼ and MS media supplemented with plant growth regulators viz. BAP (0.5 mg/l) and NAA (0.5 mg/l). Highest percentage of germination (100%) and plantlet conversion was found on hormone free full strength (1.0 MS) of MS liquid medium after 13-14 weeks of culture. Plantlets regenerated from artificial seeds with well developed shoot and root systems were successfully acclimatized in potting mixture of cocopeat, litter and sphagnum moss in a ratio 2:1:1.J. Nat. Arbutus unedo L. is a Mediterranean species used for fruit production; it is tolerant to drought and shows regeneration ability following forest fires. Mycorrhizal plants with Tuber borchii add resilience and value. This study aims to test the effect of quercetin on mycorrhizal synthesis between T. borchii and A. unedo. Two genotypes selected for fruit production and hydric stress tolerance, were micropropagated for mycorrhizal synthesis, accomplished during ex vitro rooting in perlite, using lyophilized spores of T. borchii suspended in culture media with different quercetin levels (0–10 µM). Six months after inoculation, plants were transferred to pots and maintained in nursery. Ten and 12 months after inoculation, roots were morphological examined and molecularly characterized using ITS1-5.8SITS2 rDNA region and specific primers. Results showed that mycorrhizae establishment was dependent on studied factors (genotype, quercetin level, and culture medium) and their interaction (genotype X culture medium). Quercetin levels up to 2.0 µM favored mycorrhizae establishment and plant growth, although levels superior to 4 µM showed a toxic effect. Quercetin showed to be an efficient factor on inducing mycorrhiza thriving independent of the genotype. Morphological observations and molecular analysis confirmed the permanence of the fungus association 10 and 12 months after inoculation.

In order to conserve diverse species of citrus, an experiment on in vitro micropropagation and production of synthetic seeds from in vitro regenerated plant propagules of the species; Citrus aurantifolia (Lime) was carried out in which shoot tips were found to be suitable for excapsulation of artificial seeds. Highest rate of germination was obtained from the shoot tips when MS was supplemented with 1 mg/l BAP. Beaded shoot tips produced maximum germination (81.43%). Germinated synthetic seeds with well established roots and shoots were were taken out from the culture bottles and transferred in plastic cups containing a mixture of sterile soil: sand and farmyard manure at a ratio of 1:1:1. Seedlings were further shifted in earthen pots and kept in a partial shed net house for 7 days. Those seedlings were finally transferred under the field conditions for acclimatization.
