Asymbiotic germination is considered an efficient and viable technique that can increase germination rates. The effect of type and concentration of disinfestants, and the exposure time to disinfestants may differ according to the plant species. Therefore, species-specific standardization of disinfestation agent and procedure is necessary to achieve optimal germination rates. The objective of this study was to determine a disinfestation methodology to increase in vitro germination rates and the early development of seedlings of Dendrobium nobile and Dendrobium phalaenopsis, using different times for seed disinfestation and different culture media. Seeds were disinfected by soaking in a 0.8% sodium hypochlorite solution for 5 or 15 min under aseptic conditions, after which seed suspensions were either washed with water or left unwashed. Next, they were seeded in culture flasks containing four different culture media (MS, ½MS, K, and VW). The flasks were then transferred to a growth room under controlled photoperiod and temperature, where they remained under an irradiance of 20 μmol m-2 s-1. Germination rates of the species were evaluated 45 days after placement in the culture flasks. A higher germination rate was observed when the seeds were triple washed, regardless of the culture medium or soaking time. Seed soaking disinfestation for 5 min is also recommended. MS and ½MS media were the most effective culture media in promoting in vitro germination of the species under study.

The plant Tropaeolum majus, popularly known as nasturtium, presents food, ornamental and medicinal importance. Studies on floral and carpometric characteristics generate basic knowledge for areas of science such as botany, genetics and plant breeding. This work has the objectives of analyzing the effect of combinations of doses of poultry litter on floral structures and the in vitro germination of T. majus pollen grains. From the experimental matrix Plan Puebla III were defined 9 combinations of poultry litter doses incorporated and covered. The design of the experiments was entirely random, varying the replicates number. The number of stamens, petals, sepals and carpels was verified per floral bud in each treatment. The in vitro germination rate of pollen grains was estimated in 5 culture media and 2 germination times, from flower buds randomly collected in the experiment. The overall mean of stamens, sepals, petals and carpels per floral bud was 8.23; 5; 5 and 3, respectively. The combinations of poultry litter doses influenced only the number of stamens. The in vitro germination rate of T. majus pollen grains varied in the culture media. There was no association between the combinations of poultry litter doses and the in vitro germination rate of pollen grains.

ABSTRACT The interspecific oil palm hybrid BRS Manicoré (E. guineensis x E. oleifera) has superior agronomic characteristics. However, the germination rate is low (30%) and the process is slow when the seeds are sown in a conventional form. The purpose of this study was to optimize the in vitro germination of zygotic embryos of this hybrid comparing seed lots. The viability of zygotic embryos was evaluated by the tetrazolium test (0.075%) for 4 h. The embryos were cultured on MS and Y3 culture media, with and without the addition of NaH2PO4, as well as on MS, MS1/2 and N6 medium. In MS medium containing NaH2PO4, the germination rate was increased from 40 to 70% in comparison with the medium without sodium phosphate. The comparison between the culture media MS, MS 1/2, N6 and Y3 showed that 75% of zygotic embryos cultured in the Y3 medium formed whole plants (with roots and shoots defined), a higher percentage than embryos cultured on MS, MS 1/2 and N6 media (46, 35 and 17% respectively). In the same Y3 culture medium, the embryos were larger (36% ≥ 2 cm and 30% ≥ 5 cm) than in the other media. Results obtained by the tetrazolium test were similar to those of germination, showing the effect of the genotype of each seed lot. For the germination and development of plantlets it is essential to add NaH2PO4 to a culture medium containing no phosphate or with a low phosphate concentration.

In vitro embryo culturing has arisen as a powerful tool for embryo germination of low-viability seeds. This tool has been used to germinate seeds of early-maturing or hybrid Prunus species. ‘Myrobalan’ (Prunus cerasifera Ehrh.) is a widely used rootstock for plum and apricot cultivars and its interspecific hybrids have a clear potential for breeding purposes. However, early seed abortion is often a problem in interspecific crosses because no protocol has been established yet for ‘Myrobalan’ seeds. In this work, we developed a procedure for in vitro germination of embryos of different sizes. Various factors affecting embryo germination such as the culture media, the presence of cotyledons, the stratification temperature, and the embryo size were tested in three different ‘Myrobalan’ clones. The developed protocol includes the use of full embryos that were stratified at 4 °C and cultured in C2d culture medium. The germination rate was strongly affected by the embryo size and reached 90% germination with intermediate- to large-sized embryos (6.5 to 10 mm). However, smaller embryos could also be germinated, and up to 30% germination was achieved with 0.5- to 2-mm long embryos. The results obtained here provide a protocol for in vitro germination of ‘Myrobalan’ embryos that will likely be helpful in breeding programs.

Meizotropis pellita is a rare, endangered and threatened plant species found in Patwadanger forest. In nature this species grows from rootstock in precise and sensitive habitats. However, phenolic compounds provoke browning reaction which is responsible for lethal browning in plant tissue culture and hindering in vitro germination. The aim of this study was to minimize the effect of phenolic compound during seed germination. We evaluate different antibrowning treatments in Meizotropis pellita seeds. The seeds were pretreated in Polyvinylpyrrolidone and then treated seeds were cultured on MS media and sterilized seeds were directly inoculated on PVP containing MS media. These culture media incubate under dark and light conditions at different durations respectively. Our results showed that addition of antibrowning agent, PVP inhibit onset of browning. Dark and light condition also played a crucial role on onset of browning. Delayed and low intensity of browning were observed in case of seeds that were inoculated on MS media containing PVP and were kept in dark. The present research offers a positive in vitro seed germination protocol for Meizotropis pellita conservation.

The barueiro (Dipteryx alata Vog.) is a native fruit species of the Cerrado ecoregion that has multiple uses. It is a wild species, and its cultivation is difficult. Furthermore, it is threatened with extinction. Plant tissue culture is a major tool for the conservation of germplasm, as well as a means of propagating high-quality seedlings on a large scale. However, this technique has not been used with barueiro, although it might provide valuable contributions to the process of barueiro domestication. The most popular method of cultivation is the use of the Murashige and Skoog medium (MS), which is considered one of the most nutritionally complete media. Woody plant medium (WPM) is indicated for the propagation of woody species, but there are no reports of its use for barueiro cultivation. Woody plants tend to have problems with rust in vitro during the establishment phase. Activated charcoal acts as an adjuvant for the adsorption of phenolic compounds, mitigating its effects in the medium. Thus, the objective of this study was to test four activated charcoal doses (0, 2, 4 and 6 g L-1) and three culture media: MS, WPM, and AA (over water agar) in the in vitro establishment of barueiro. The experimental design was a completely randomised (DIC), 4 × 3 factorial design with three replications. At 60 days after inoculation, the explants were evaluated for dry matter, fresh weight, stem diameter, shoot length, number of leaves, longest root length, germination rate, and chlorophyll contents. The MS medium supplemented with 3,0 g L-1 activated charcoal appeared to be the best for in vitro establishment of barueiro.

This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.

Zingiber spectabile is a tropical ornamental species with difficulties to obtain efficient propagation system. Thus, this study aimed to assess the in vitro propagation of Zingiber spectabile. Seed characterization was determined by measuring length, width and thickness, the weight of 1000 seeds and imbibition curve. In vitro germination of seeds was at constant (25 °C) or alternating temperatures (20-30 ºC). For optimization of in vitro multiplication, different concentrations of activated charcoal (0.0, 0.1 and 0.3%) and sucrose (0.0, 0.1, 0.3, 0.5 and 0.7 M) were evaluated. Plantlets were inoculated in flasks with different sealing systems (PVC covers with or without filters at the center) and culture media (MS or WPM). The plants were acclimatized in Plantmax® substrate. Seeds were of 6.06 mm length, 3.22 mm wide and 2.83 mm thick. The weight of 1,000 seeds corresponded to 46.4 g. The seed imbibition curve approaches to a tree phase pattern. Alternating temperatures induced high germination rates (68%). The addition of 0.3% activated charcoal provided higher root growth and plants with smaller number of senescent leaves. The best plant growth was obtained by the use of 0.1 M sucrose. All acclimatized plants survived (100%). The results demonstrate that Z. spectabile respond well to in vitro propagation.

Many golf courses and turfgrass managers use recycled water, which contains high salts, as part or a sole irrigation source to lower costs and comply with governmental restrictions on water use. High salinity negatively affects turfgrass performance. Using salt-tolerant species or cultivars is one the most effective methods to address salinity problems. Twenty-six commercially available creeping bentgrass (Agrostis stolonifera) cultivars were evaluated for salt tolerance during in vitro germination on 1% agar media supplemented with NaCl at 0, 5, 10, 15, or 20 g·L−1 at 25/15 °C (day/night) under fluorescent light (36 μmol·s−1·m−2) with an 8- to16-h photoperiod. Significant variations in salinity tolerance were observed among the cultivars. Final germination rate (FGR, %) and daily germination rate (DGR, %/d) decreased linearly or quadratically as salinity levels increased. ‘Declaration’, ‘Seaside II’, ‘T-1’, and ‘Bengal’ were the most salt-tolerant, requiring salt levels at or greater than 16.0 and 10.0 g·L−1, respectively, to reduce FGR and DGR by 50%. In contrast, ‘Tyee’, ‘Kingpin’, and ‘SR1150’ required average salinity levels of 11.6 and 6.5 g·L−1 to cause 50% reduction in FGR and DGR, respectively, showing that they were the least salt-tolerant cultivars. The largest difference between FGR (1.9%) and DGR (26.2%) reduction under saline conditions was observed at 5 g·L−1, indicating that DGR was more sensitive to salinity changes than FGR. Therefore, DGR might be a more reliable method to be used for salt selection.
