A V-shaped ligand Bis(2-benzimidazolymethyl)amine (bba) and its nickel(II) picrate (pic) complex, with composition [Ni(bba)2](pic)2⋅3MeOH, have been synthesized and characterized on the basis of elemental analyses, molar conductivities, IR spectra, and UV/vis measurements. In the complex, the Ni(II) ion is six-coordinated with a N2O4ligand set, resulting in a distorted octahedron coordination geometry. In addition, the DNA-binding properties of the Ni(II) complex have been investigated by electronic absorption, fluorescence, and viscosity measurements. The experimental results suggest that the nickel(II) complex binds to DNA by partial intercalation binding mode.

A tripod ligand tris(N-methylbenzimidazol-2-ylmethyl)amine (Mentb) and its nickel(II) picrate (pic) complex, with composition [Ni(Mentb)(DMF)(H2O)](pic)2, have been synthesized and characterized on the basis of elemental analyses, molar conductivities, IR spectra, and UV/Vis measurements. Single-crystal X-ray diffraction revealed that the Ni atom is six-coordinated in a distorted octahedral geometry. In addition, the DNA-binding properties of the ligand Mentb and its Ni(II) complex have been investigated by electronic absorption, fluorescence and viscosity measurements. The experimental results suggest that the ligand and its Ni(II) complex bind to DNA via an intercalation binding mode, and their binding affinity to DNA follows the order of complex>ligand.

A series of chiral cis-dichloro(pyridyloxazoline)platinum(II) and palladium(II) complexes were synthesized and their reactivity towards a defined sequence of single-stranded and double-stranded DNA was investigated in comparison to cisplatin. The compounds differed in the nature and absolute configuration of the substituent at the C4 position of the oxazoline ring. The DNA-binding ability of these compounds was evaluated by HPLC analysis, post metal exposure, of enzymatic digests of an undecamer duplex containing one putative metallation site. Polyacrylamide gel electrophoresis (PAGE) and thermal denaturation confirmed the results of the HPLC analysis, which showed that the stereochemistry and character of the substituent at the C4 position of the oxazoline ring had little effect on DNA binding, possibly due to the formation of monofunctional adducts.Key words: cisplatin, chiral, pyridyloxazoline, DNA-binding studies, platinum, palladium.

DNA binding studies of selected heterocyclic compounds belonging to the class of quinolinones, substituted quinolinones and thiones were carried out using ct-DNA. The binding nature of the compounds with DNA analyzed using UV-spectroscopy revealed the compounds to be DNA intercalators demonstrating the binding nature of compounds with DNA base pairs. This study is aimed at establishing a facile UV spectroscopic technique to arrive at the binding mode of DNA to ligands.

A ternary cationic Mn(II) complex with the tripodal ligand tris(2-(N-methyl) benzimidazylmethyl)amine (Mentb), salicylate and DMF as ligands and nitrate as counterion, [Mn(Mentb)(salicylate)DMF](NO3), was synthesized and characterized by physico-chemical and spectroscopic methods. The crystal structure of the Mn(II) complex has been determined by single-crystal X-ray diffraction and revealed that the central Mn(II) atom is seven-coordinated. The DNA-binding properties of the Mn(II) complex were investigated by spectrophotometric methods and viscosity measurements, and the results suggest that the Mn(II) complex binds to DNA via an intercalation binding mode. Additionally, the complex exhibited potential hydroxyl radical scavenge properties in in vitro studies

A Schiff base ligand bis(N-salicylidene)-3-oxapentane-1,5-diamine (H2L) and its lutetium(III) complex, with composition Lu2(L)2(NO3)2, were synthesized and characterized by physico-chemical and spectroscopic methods. The crystal structure of the Lu(III) complex has been determined by single-crystal X-ray diffraction. It reveals a centrosymmetric binuclear neutral entity where Lu(III) metal centers are bridged by two phenoxo oxygen atoms. The DNA-binding properties of the Lu(III) complex were investigated by spectrophotometric methods and viscosity measurements, and the results suggest that the Lu(III) complex binds to DNA via a groove binding mode. Additionally, the antioxidant activity of the Lu(III) complex was determined by the superoxide and hydroxyl radical scavenging methods in vitro, which indicate that it is a scavenger for OH· and O-· 2 radicals.

UV-Vis spectroscopy studies, viscosity measurements and competitive binding studies with EB have revealed the ability of the complexes to bind to CT DNA covalently through N7 of guanine residues and non-covalently through intercalation.
