Steinernema borjomiense n. sp. (Rhabditida: Steinernematidae), a new entomopathogenic nematode from Georgia

Summary A new species of entomopathogenic nematode, Steinernema borjomiense n. sp., was isolated from the body of the host insect, Oryctes nasicornis (Coleoptera: Scarabaeidae), in Georgia, in the territory of Borjomi-Kharagauli. Morphological characters indicate that the new species is closely related to species of the feltiae-group. The infective juveniles are characterised by the following morphological characters: body length of 879 (777-989) μm, distance between the head and excretory pore = 72 (62-80) μm, pharynx length = 132 (122-142) μm, tail length = 70 (60-80) μm, ratio a = 26.3 (23.0-29.3), H% = 45 (40-51), D% = 54 (47-59), E% = 102 (95-115), and lateral fields consisting of seven ridges (eight incisures) at mid-body. Steinernema borjomiense n. sp. was molecularly characterised by sequencing three ribosomal regions (the ITS, the D2-D3 expansion domains and the 18S rRNA gene) and the mitochondrial COI gene. Phylogenetic analyses revealed that S. borjomiense n. sp. differs from all other known species of Steinernema and is a member of the monticolum-group.

Abstract During a survey for entomopathogenic nematodes in citrus orchards throughout South Africa, a new species of Steinernema was isolated from a citrus orchard on Rietkloof farm, near the town of Piketberg in the Western Cape Province, South Africa. The nematode was isolated from soil using the Galleria-baiting technique. Steinernema citrae n. sp. is characterised by the following morphological characters: third-stage infective juvenile with a body length of 754 (623-849) μm, distance from head to excretory pore of 56 (49-64) μm, tail length of 71 (63-81) μm, and ratio E value of 110 (85-132). The lateral pattern for the new species is 2, 7, 8, 6, 4, 2 and is not typical for the genus. Steinernema citrae n. sp. is closely related the feltiae-group. The body length of the IJ is close to that of S. texanum and S. weiseri, though it differs in body diam., the length of the pharynx and E%. The male of S. citrae n. sp. differs from S. feltiae in the length and shape of the spicule and body diam. Steinernema citrae n. sp. differs from all species in the feltiae-group in the morphology of the vulva, as it has a single flapped, low, epiptygma. It also differs from the most closely related species, S. feltiae, as there is no interbreeding between the two species. In addition, the new nematode differs from other species of the feltiae-group by characteristics of the ITS and D2D3 regions of its rDNA.

A new isolate of the entomopathogenic nematode, Steinernema taiwanensis n. sp., was isolated from soil in Pingtung County, Taiwan. This new species could be characterized and distinguished from other related species by its morphological characters, morphometrics, and phylogenetic analysis. The body length and distance from anterior end to nerve ring of infective juveniles is 1012 (983–1045) µm and 124 (120–127) µm, lateral field formula 2, 6, 7, 8, 2, and the tail length 90 (79–96) µm without dorsal constriction in tail region. The first generation males of S. taiwanensis n. sp. are characterized by spicule shape, smooth blade tip, 23 genital papillae (11 pairs and 1 single papilla), spicule length of 94 (89–99) µm and gubernaculum length of 68 (65–70) µm. Females from the first generation of S. taiwanensis n. sp. have no epiptygmata and a slightly developed post-anal swelling. Phylogenetic analysis of ITS and D2D3 regions of rDNA showed that S. taiwanensis n. sp. belongs to the Longicaudum-clade and comprises a monophyletic group with S. guangdongense and S. longicaudum. The new isolate is described as a novel species according to morphological and phylogenetic analyses.

The derbid genus Oropuna is a small taxon of Neotropical planthoppers in the tribe Cenchreini comprised of three species. Recent survey work on palms for planthoppers in Costa Rica resulted in the discovery of a fourth species, Oropuna halo sp. n. In this study the new species is described and a key to the four species is provided along with sequence data for the cyctochrome c oxidase subunit I (COI) and 18S rRNA gene for the novel taxon.

Two oxytrichid ciliates collected from China, Oxytricha lithofera Foissner, 2016 and Rubrioxytricha haematoplasma (Blatterer and Foissner, 1990) Berger, 1999 were investigated based on living observations, protargol preparations, and molecular analyses. The Chinese population of O. lithofera shares highly distinctive features with the type population, e.g., lithosomes, abutting macronuclear nodules, straight undulating membranes, and long dorsal bristles. The morphology and morphogenesis of our new isolate of R. haematoplasma corresponds well with the type population. The 18S rRNA gene sequences of both species were obtained, and the molecular phylogeny of the genera Oxytricha Bory de Saint-Vincent in Lamouroux et al., 1824 and Rubrioxytricha Berger, 1999 was analyzed. In addition, we found that the Guangzhou population of R. haematoplasma described by Chen et al. (2015) can be distinguished from the type population and our isolate by the reddish-brown cortical granules (vs. lemon yellowish to greenish) and mitochondria-like granules present (vs. absent), as well as the slightly red cytoplasm (vs. slightly orange) and the marine or brackish water habitat (vs. freshwater). In addition, a 20-bp divergence in their 18S rRNA gene sequences indicates that they are not conspecific. Thus, we establish a new species, Rubrioxytricha guangzhouensis, for R. haematoplasma sensu Chen et al. (2015).

We describe Tanaopsis japonica sp. nov. from intertidal and nearshore habitats around Hokkaido Island, northern Japan. This species closely resembles T. chotkarakde Bird & Bamber, 2000 and T. rawhitia Bird, 2011 in having the uropod with the endopod biarticulate and the exopod uniarticulate, but differs from them in the following characters: pereonite 4 as long as pereonite 5; mandibles lacking the molar; the number of simple setae on the antenna, propodal palm, and dactylus of pereopods 1–3; the number of spiniform setae on the cutting surface of the chelipedal dactylus; and the number of setae on the pleopods. We also determined a nearly complete nucleotide sequence for the 18S rRNA gene in T. japonica. In both sexes, Tanaopsis japonica sp. nov. bears serial ridges and grooves on the chela, antennule, and antenna. Parallel, fringed hatch marks on the chelipedal dactylus may function in stridulation. A series of denticulate ridges on the dorsal surface of antennal article 2 faces the ventral side of article 1 of the antennule, where there is a ventral series of slight grooves. Although the ranges of motion of the antennule and antenna were unclear, the ridges and grooves on the antenna and antennule may also function in producing sound. Two outer series of ridges on the chela, together with an outer furrow on the chela, may function in digging, by allowing bottom-sediments to be efficiently pushed aside.

A new species of the Asian leaf litter toad genus Leptobrachella from Guizhou Province, China is described based on molecular phylogenetic analyses, morphological comparisons, and bioacoustics data. Phylogenetic analyses based on the mitochondrial 16S rRNA gene sequences supported the new species as an independent clade nested into the Leptobrachella clade and sister to L. bijie. The new species could be distinguished from its congeners by a combination of the following characters: small body size (SVL 30.8–33.4 mm in seven adult males, and 34.2 mm in one adult female); dorsal skin shagreened, some of the granules forming longitudinal short skin ridges; tympanum distinctly discernible, slightly concave; internasal distance longer than interorbital distance; supra-axillary, femoral, pectoral and ventrolateral glands distinctly visible; absence of webbing and lateral fringes on fingers; toes with rudimentary webbing and shallow lateral fringes, relative finger lengths II < IV < I < III; heels overlapped when thighs are positioned at right angles to the body; and tibia-tarsal articulation reaches the tympanum.

A new species of the Asian leaf litter toad genus Leptobrachella is described from Guizhou Province, China. The new species is distinguished from its congeners by a combination of the following morphological characters: body size medium (SVL 29.7–31.2 mm in five adult males); dorsal skin shagreened, some of the granules forming longitudinal short skin ridges; tympanum distinctly discernible, slightly concave; supra-axillary, femoral, pectoral and ventrolateral glands distinctly visible; absence of webbing and lateral fringes on fingers; toes with narrow lateral fringes but without webbing; heels overlapping when thighs are positioned at right angles to the body; tibia-tarsal articulation reaching the middle of eye when leg stretched forward. The discovery highlighted the underestimated species diversity in the Leptobrachella toads in southwestern China.

We describe a new species of Megophryidae frog, Megophrys shunhuangensis sp. nov., from Hunan Province, Southern China. It can be distinguished from other known species in the subgenus Panophrys by morphological characters, bioacoustic data and a molecular divergence in the mitochondrial 16S rRNA gene similar to that found among other species of Panophrys. M. shunhuangensis sp. nov. is characterized by a relatively small body size, with adult females measuring 37.6 mm and adult males measuring 30.3–33.6 mm in snout to vent length; maxillary teeth present, vomerine teeth absent; tongue smooth, not notched behind; hindlimb slender, heels overlapping, tibio-tarsal articulation reaches forward between the nasal and tip of snout. Molecular phylogenetic analyses also show that M. sp. 6 from (Mao’er Shan, China) from Chen et al. (2017) and M. sp. 24 (Anjiangping and Mao’er Shan, China) from Liu et al. (2018) may be the same species as M. shunhuangensis sp. nov., we consequently speculate that the new species also distributed in Anjiangping and Mao’er Shan, China. At present, the genus Megophrys contains 85 species of which 48 species are distributed in China, and 30 belong to the subgenus Panophrys.

A new species of the genus Nothotylenchus, N. persicus n. sp. was collected around the roots of grapevine and is described and illustrated herein based on morphological and molecular studies. The new species is characterised by a body length of 776-900 μm, delicate stylet 5-6 μm long, six lines in the lateral field, post-vulval uterine sac short, 10-18 μm long, female tail elongate-conoid with pointed terminus, and bursa covering 40-45% of tail length. Morphologically, N. persicus n. sp. appears closer to four known species of the genus, namely: N. hexaglyphus, N. affinis, N. medians and N. taylori. The results of phylogenetic analyses based on sequences of D2-D3 expansion region of 28S rRNA gene confirmed the close molecular relationship between N. persicus n. sp. and other anguinids, but Nothotylenchus claded separately from Ditylenchus species.

SUMMARY On 9 November 2015, a juvenile male fin whale of 11·60 m length was observed on the bulb of a merchant vessel in the Channel Terneuzen – Ghent (The Netherlands – Belgium). A severe parasitosis was present in the right heart ventricle and caudal caval vein. Parasites were identified as Crassicauda boopis based on macroscopic and microscopic observations. The sequence of the 18S rRNA gene obtained from the parasite samples was 100% similar to the sequence of the 18S rRNA gene from Crassicauda magna available on GenBank. While adults of C. boopis and C. magna are morphologically distinct and found at different locations in the body, the molecular analysis of the 18S rRNA gene seems insufficient for reliable species identification. Although numerous C. boopis were found, the cause of death was identified as due to the collision with the ship, as suggested by the presence of a large haematoma, and the absence of evidence of renal failure. The young age of this whale and the absence of severe chronic reaction may suggest that the infestation was not yet at an advanced chronic stage.
