Morph-molecular characterization of Meira nicotianae sp. nov., a novel basidiomycetous, anamorphic yeast-like fungus associated with growth improvement in tobacco plant

A novel yeast-like species was isolated from the rhizosphere of tobacco root collected in Guizhou province, China. Sequence analysis of the large subunit (LSU) rDNA gene indicates that the isolate represents a novel species and most closely related to Meira siamensis. The phylogenetic analysis based on the Internal Transcribed Spacer (ITS) rDNA gene also confirmed the placement of the novel species within Meira. However, the novel species differs from M. siamensis by 9.1% bp difference in the LSU rDNA sequence region and also in terms of physiological characteristics as compared to other species. This species is proposed as M. nicotianae sp. nov. Co-incubation of M. nicotianae strain CY-1 and tobacco plantlets demonstrates that this fungal strain can promote growth of tobacco plant. Scanning electron micrographs and other morphological illustrations along with phylogenies are presented in this paper.

Two novel anamorphic yeast strains (S-15LT and 3-C1) were isolated from the inflorescences of plants collected in two different towns in Rajasthan State, India. Sequencing of the D1/D2 domains of the large-subunit (LSU) rDNA and the internal transcribed spacer (ITS) regions suggested they are strains of the same species. Phenotypic characteristics such as the absence of fermentation, the absence of sexual structures and ballistoconidia, the assimilation of myo-inositol and d-glucuronate, and positive Diazonium blue B and urease reactions indicated that these strains belong to the genus Cryptococcus. The novel strains differed from Cryptococcus laurentii in six physiological tests and differed from other related species in more than six tests. A phylogenetic analysis of the sequences of the D1/D2 domains of the LSU rDNA and the ITS regions placed these strains in the Bulleromyces clade within the order Tremellales, with C. laurentii as their closest described relative. The novel strains showed 1.6 and 7.5 % divergence in the D1/D2 domain of the LSU rDNA and ITS regions, respectively, with respect to C. laurentii. The divergence from other species was more than 3 % for the D1/D2 domain and more than 9 % for the ITS region. On the basis of the phenotypic and molecular data, strains S-15LT and 3-C1 represent a novel species within the genus Cryptococcus, for which the name Cryptococcus rajasthanensis sp. nov. is proposed. The type strain is S-15LT (=MTCC 7075T=CBS 10406T).

A novel strain of anamorphic yeast, designated strain NRRL Y-27208T, was isolated from concentrated grape juice in Cape Province, South Africa. Analysis of nuclear large subunit rRNA gene sequences from the D1/D2 domains separated the novel isolate from strains of Starmerella bombicola and Starmerella meliponinorum, as well as from species of the genus Candida that are members of the Starmerella clade. Compared to previously described species, strain NRRL Y-27208T is most closely related to S. bombicola but can be separated from this species by its ability to grow on d-ribose and erythritol. Strain NRRL Y-27208T produced sophorolipids that have an open chain structure similar to Candida batistae, Candida riodocensis and Candida stellata, which is in contrast to the closed chain sophorolipids produced by S. bombicola and Candida apicola. The analyses showed that NRRL Y-27208T ( = CBS 7267T) represents a novel species distinct from previously described species, for which the name Candida kuoi sp. nov. is proposed.

During an investigation of yeast biota in the rhizosphere of lentisk in Sardinian semi-arid areas, a strain was isolated that could not be assigned to any known species. The sequence of the D1/D2 domain of the large subunit rDNA gene revealed that the strain belonged to the genus Kazachstania and was phylogenetically related to a clade including Kazachstania aerobia, Kazachstania servazzii, Kazachstania solicola and Kazachstania unispora. The novel isolate differed from members of this clade in its ability to assimilate d-glucono-1,5-lactone and its very weak fermentation of glucose and sucrose; its assimilation profile was unique within the genus Kazachstania. Monosporal colonies were able to sporulate, indicating that the species is homothallic. It is proposed that the isolate represents a novel species, Kazachstania ichnusensis sp. nov., with LCF 1675T ( = CBS 11859T) as type strain.

The novel genus Holtermanniella is proposed here to accommodate four Cryptococcus species closely related to Holtermannia corniformis that are included in the Holtermannia clade (Basidiomycota, Agaricomycotina). Thus, four novel combinations are proposed: Holtermanniella nyarrowii comb. nov., Holtermanniella festucosa comb. nov., Holtermanniella mycelialis comb. nov. and Holtermanniella wattica comb. nov. In addition, a novel anamorphic yeast species was studied with 15 isolates obtained from different habitats around the world. Analysis of the sequences of the D1/D2 region of their large subunit rDNA showed that the novel species is placed phylogenetically within the Holtermannia clade of the Tremellomycetes (Agaricomycotina, Basidiomycota). PCR fingerprinting and sequencing of ITS1–5.8S–ITS2 showed genetic intraspecific variability among the strains: three groups were formed, which did not correlate with geographical origin or substrate. This novel species, designated the type species of Holtermanniella gen. nov., is described as Holtermanniella takashimae sp. nov.; the type strain is CBS 11174T (=HB 982T =DBVPG 8012T). The order Holtermanniales ord. nov. is proposed here to include Holtermannia (the type genus) and Holtermanniella.

Four strains of a novel yeast species were isolated from laboratory nests of the leaf-cutting ant Atta sexdens in Brazil. Three strains were found in older sponges and one was in a waste deposit in the ant nests. Sequencing of the D1/D2 region of the large-subunit rRNA gene showed that the novel species, named Sympodiomyces attinorum sp. nov., is phylogenetically related to Sympodiomyces parvus. Unlike Sympodiomyces parvus, Sympodiomyces attinorum can ferment glucose, assimilate methyl α-d-glucoside, salicin and citrate, and grow at 37 °C, thus enabling these two species to be distinguished. Differentiation from other related species is possible on the basis of other growth characteristics. The type strain of Sympodiomyces attinorum is UNESP-S156T (=CBS 9734T=NRRL Y-27639T).

Five strains of a novel methanol-assimilating yeast species were isolated from mango (Mangifera indica) leaves collected at the campus of the Federal University of Minas Gerais in Brazil. The sequences of the internal transcribed spacer (ITS) region and the D1/D2 domains of the large subunit of the rRNA gene showed that this species belongs to the Ogataea clade and is related to O. allantospora, O. chonburiensis, O. dorogensis, O. kodamae, O. paradorogensis and Candida xyloterini (Ogataea clade). The novel species differs in the D1/D2 domains of the large subunit of the rRNA gene by 12 to 40 substitutions from these Ogataea species. The name Ogataea mangiferae sp. nov. is proposed for this novel species. The type strain of Ogataea mangiferae sp. nov. is UFMG-CM-Y253T ( = CBS 13492T). The Mycobank number is MB 811646.

Five strains of a novel yeast species were isolated from rotting wood samples collected in an Amazonian forest site in the state of Roraima, northern Brazil. The sequences of the D1/D2 domains of the large subunit of the rRNA gene showed that this species belongs to the Scheffersomyces clade and is related to Candida coipomoensis, Candida lignicola and Candida queiroziae. The novel species Candida amazonensis sp. nov. is proposed to accommodate these isolates. The type strain of C. amazonensis sp. nov. is UFMG-HMD-26.3T ( = CBS 12363T = NRRL Y-48762T).

A novel anamorphic yeast species is described to accommodate three isolates recovered from the guts of three different wood-boring insect larvae collected in Henan, central China. On the basis of sequence analyses of the D1/D2 domains of the large-subunit rRNA gene and the internal transcribed spacer regions, the three strains are assigned to a novel species of the genus Wickerhamomyces, although the formation of ascospores was not observed. These strains also exhibited a number of distinct morphological and physiological characteristics that clearly differentiated them from Wickerhamomyces mucosus, Candida odintsovae and Wickerhamomyces rabaulensis, the most closely related species. In view of the phenotypic differences and unique rRNA gene sequences, we consider that these three isolates represent a novel species of the genus Wickerhamomyces, Wickerhamomyces mori sp. nov. The type strain is NYNU 1216T ( = CICC 1983T  = CBS 12678T).

Six strains of a novel yeast species were isolated from tree bark collected in the Atlantic Forest and the Amazon Rainforest in Brazil. Analyses of the sequences of D1/D2 domains of the large subunit rRNA gene showed that the strains belong to a species in the genus Zygotorulaspora. The species differed by 5.54 % sequence divergence (25 substitutions and five indels out of 542 bp) in the D1/D2 sequences from Zygotorulaspora mrakii, its closest relative. The ITS sequence of the type strain of the novel species differs by 27–69 nucleotide substitutions/indels from the other Zygotorulaspora species. The novel species is able to grow on trehalose, maltose, l-sorbose, inulin and at 37 °C, which are negative in Z. mrakii. The name Zygotorulaspora cariocana sp. nov. is proposed. The holotype of Z. cariocana sp. nov. is CBS 16118T. The MycoBank number is MB 833702.
