Genome characterization and analysis is an imperative step in identifying and selectively breeding for improved traits of agriculturally important species. Expressed sequence tags (ESTs) represent a transcribed portion of the genome and are an effective way to identify genes within a species. Downstream applications of EST projects include DNA microarray construction and interspecies comparisons. In this study, 694 ESTs were sequenced and analyzed from a library derived from a 24-day-old turkey embryo. The 437 unique sequences identified were divided into 76 assembled contigs and 361 singletons. The majority of significant comparative matches occurred between the turkey sequences and sequences reported from the chicken. Whole genome sequence from the chicken was used to identify potential exon–intron boundaries for selected turkey clones and intron-amplifying primers were developed for sequence analysis and single nucleotide polymorphism (SNP) discovery. Identified SNPs were genotyped for linkage analysis on two turkey reference populations. This study significantly increases the number of EST sequences available for the turkey.Key words: turkey, cDNA, expressed sequence tag, single nucleotide polymorphism.

Context In an era of rapid environmental change, many species are shifting their distributions. As temperate-zone species’ expand their ranges north, different and potentially severe limiting factors may begin to affect their ability to survive and reproduce. The wild turkey (Meleagris gallopavo) is one example of a species undergoing rapid northern expansion. Aims An improved understanding of wild turkey demography at the species’ northern range periphery would facilitate effective management of this important game species. Therefore, we undertook a study to evaluate survival, causes of mortality, and behavioural strategies that may govern survival of female wild turkeys. Methods We captured 53 female wild turkeys, and used backpack transmitters to monitor their individual fates during 2012–13. Key results The annual survival estimate was 0.37 (95% CI: 0.25–0.55), with the lowest seasonal survival in the winter. The most frequent cause of mortality was mammalian predation, predominately by coyotes (Canis latrans). Age, proximity to supplemental food, and habitat use did not affect risk of mortality. Conclusions Northern wild turkeys in our study exhibited lower survival and suffered higher predation than did populations in the species’ historic range. Despite our findings, the wild turkey has expanded its range northwards and continues to exist in these peripheral areas. This may be due to high productivity or a source–sink dynamic, whereby high mortality is offset by immigration from the south. Implications The low survival and high predation of wild turkeys in the north must be considered when developing management strategies, particularly if interest exists in translocating turkeys farther north. Further research is needed to better understand whether northern turkey populations persist as sinks.

The effect of taurine on the turkey spermatozoa motility and viability during the in vitro incubation was assessed. Experimental samples were prepared by diluting the raw semen in nine different concentrations of taurine – from 10 mg/ml to 0.078125 mg/ml. The motility parameters were evaluated by the CASA system (Computer Assisted Semen Analyser) using the program Sperm Vision<sup>®</sup> and for spermatozoa viability assessment the eosin-nigrosin staining was performed. Selected parameters were evaluated at six time periods: 0, 1, 2, 3, 4, and 5 h at 5°C and 41°C. At 5°C, a significantly lower percentage of motility and progressive motility was detected only in the samples with the highest concentration of taurine (10 mg/ml) at time 0 and 1. After 2 h of incubation a significant preventive effect of taurine on spermatozoa parameters was observed. The tendency of the taurine effect on motility parameters was different during the in vitro incubation at 41°C. Significantly lower values of motility parameters were detected in all experimental samples in comparison to the control after 5 h. The analysed concentrations of taurine did not significantly affect viability of turkey spermatozoa during all time periods. A higher percentage of dead spermatozoa were observed at 41°C (4.87–9.90%) if compared to 5°C (2.12–4.88%). The results indicated that the addition of taurine (from 2.5 to 7.5 mg/ml) to turkey spermatozoa positively affected the monitored spermatozoa parameters incubated at 5°C.
