Drosophila melanogaster is one of the most famous insects in biological research. It is widely used to analyse functions of different genes. The phosphatidylcholine lysophospholipase gene swiss cheese was initially shown to be important in the fruit fly nervous system. However, the role of this gene in non-nervous cell types has not been elucidated yet, and the evolutional explanation for the conservation of its function remains elusive. In this study, we analyse expression pattern and some aspects of the role of the swiss cheese gene in the fitness of Drosophila melanogaster. We describe the spatiotemporal expression of swiss cheese throughout the fly development and analyse the survival and productivity of swiss cheese mutants. We found swiss cheese to be expressed in salivary glands, midgut, Malpighian tubes, adipocytes, and male reproductive system. Dysfunction of swiss cheese results in severe pupae and imago lethality and decline of fertility, which is impressive in males. The latter is accompanied with abnormalities of male locomotor activity and courtship behaviour, accumulation of lipid droplets in testis cyst cells and decrease in spermatozoa motility. These results suggest that normal swiss cheese is important for Drosophila melanogaster fitness due to its necessity for both specimen survival and their reproductive success.

This research was conducted at the Teaching Farm of the Faculty of Veterinary Medicine, Airlangga University, to know the effect of age, temperature, and humidity on the quality of semen of bulls which are turned into frozen semen. This research uses fresh semen samples of bulls of various ages grouped into 3 groups, young, middle age, and old groups and measures temperature and humidity during the storage process. Fresh semen is collected using an artificial vagina and then examined macroscopic and microscopic tests which is then checked for post thawing motility. Data analysis uses Analysis of Variants (ANOVA) followed by Duncan. The results showed that the percentage of spermatozoa motility of fresh semen and post thawing differed and age affected the quality of semen produced. There was no effect of temperature and humidity when processing of collecting semen. Bull’s semen at Teaching Farm Faculty of Veterinary Medicine Airlangga University has good quality and is suitable for Artificial Insemination.

Infertility occurs in 2 million couples or 17% of couples who are married more than 2 years but are not pregnant or have signs of pregnancy. In couples who do not have children, 50% of male infertility factors are caused by abnormalities in semen. Empirically, parijoto fruits (Medinilla speciosa Blume) are used by the people of Kudus, Central Java, Indonesia to increase fertility. The purpose of this research was to find out the effect of methanol extract, methanol fraction, and n-hexane fraction of parijoto fruit (Medinilla speciosa) on seminal parameters and testicular weight of male Sprague Dawley rats. This research used 36 two-month-old Sprague Dawley rats with 200-300 gram of body weight which were divided into 6 groups. Group 1 (normal); groups 2, 3, and 4, used parijoto fruit methanol extract at 100mg/kgBW, 250mg/kgBW, and 500mg/kgBW doses respectively; groups 5 and 6 used methanol fraction and n-hexane fraction of parijoto fruit at 500 mg/kgBW dose respectively for 14 days. Rats were dissected and had examinations on sperm motility, morphology, abd testicular weight. The data were analyzed using Kruskal Wallis and Mann Whitney. The results found that groups 1, 2, 3, 4, 5 and 6 had mean spermatozoa motility (%) that were 56.5±2.43, 72.5±6.89, 77.6±12.99, 83.3±7.53, 84.7±3.98, and 74.2±11.58, mean spermatozoa morphology (%) that were 95.5±3.67, 95±2.76, 92.6±5.13, 96.5±3.27, 94.3±4.37, and 93.2±6.11, and mean testicular weight (gram) that were 1.08±0.10, 0.90±0.14, 0.98±0.10, 1.18±0.21, 1.28±0.43, and 1.02±0.13. There were significant differences between the normal group and all treatments (P <0.05) on spermatozoa motility. There was no significant difference on spermatozoa morphology and testicular weight. Based on the results, it can be concluded that methanol extract, methanol fraction, and n-hexane fraction of parijoto fruit can increase spermatozoa motility of male Sprague Dawley rats.

Research on the evaluation of the spermatozoa quality of Brahma, Cochin and Bangkok chickens as the basis for the application of artificial insemination technology in native chickens has been carried out. The purpose of this study was to evaluate the spermatozoa quality of Brahma, Cochin and Bangkok rooster as the basis for determining the spermatozoa dose for artificial insemination in local chickens. method research was used a completely randomized design (CRD) with three treatments, that is: Brahma, Cochin and Bangkok rooster. Each treatment was repeated five times. Cement storage is carried out by inserting an artificial vagina into the cloaca of the rooster and stimulating it to climb the hen. The collected sperm were analyzed at the Animal Reproduction Laboratory, Faculty of Animal Science, Mataram University. The variables observed included motility, viability and morphology of spermatozoa. The results of studied that the spermatozoa motility of Brahma, Chicin and Bangkok rooster were 72 ± 2.74%. 74 ± 2.24% and 73 ± 2.74% (P> 0.05) respectively; The spermatozoan viability of Brahma rooster and Bangkok rooster was 99.2 ± 1.30%, 99.4 ± 0.55% and 99.2 ± 1.10% (P>0.05)respectively. The spermatozoan morphology of brahma rooster, chicin rooster and Bangkok rooster was 94.6 ± 2.30%, 94.4 ± 2.70% and 97.4 ± 2.07% (P> 0.05), respectively. Cochin rooster and Bangkok rooster respectively, 62.6 ± 7.92 x 107, 57 ± 5.83 x107 , and 65.2 ± 12.28 x107, respectively. The results of the study concluded that Brahma, Cochin and Bangkok rooster had good sperm quality and could be able to used in artificial insemination of Kampung hens.

Abstract This study compared the semen quality traits between Etawah grade (EGB) and Senduro bucks (SB) raised in Singosari National Artificial Insemination Center, Indonesia. A total of 96 ejaculated semen were collected from three EGB and three SB with an average age of three years. The ejaculates were evaluated for semen volume (SV), spermatozoa concentration (SC), total spermatozoa (TS), spermatozoa motility (SM), post-thawing spermatozoa motility (PTSM), the recovery rate of spermatozoa motility (RRSM), and frozen semen production (FSP). Data of SV, SM, PTSM, and RRSM were compared using the Mann-Whitney U test. While the data of SC, TS, and FSP were analyzed using an independent t-test. The results showed that SV, SM, PTSM, and RRSM were not significantly differed (P>0.05) between breeds. Whereas, SC, TS, and FSP of EGB were significantly higher (P<0.05) than SB. Therefore, it can be concluded that Etawah grade bucks can produce higher total spermatozoa and as a result higher frozen semen production for artificial insemination as compared to Senduro bucks.

Despite available information on the roles of osmotic pressure, potassium (K+), and calcium (Ca2+) in activation of Eurasian burbot spermatozoon motility, the changes in milt biochemical composition and mechanisms underlying their activation at temperatures above optimal spawning temperature is still unclear. We explored spermatozoon swelling, lipid composition and proteome in relation to osmolality and temperature of swimming medium. The result revealed that temperature increment from 4 to 30°C increases burbot spermatozoa vulnerability to osmotic pressure, decreasing motility in extremely hypotonic media, and the presence of Ca2+ decreases swelling of spermatozoa. No spermatozoon swelling was observed in non-ionic isotonic media at all studied temperatures. A role of swelling in activation of spermatozoa motility at 30°C was rejected. No differences were found in protein profile and lipid composition with respect to activation of burbot spermatozoa at 30°C. Burbot spermatozoon activation occurs at 30°C without modification of the spermatozoon membrane. Elucidation of the intrinsic signaling pathway of burbot spermatozoon spontaneous activation requires further study.

This study was based on the determination of post-thaw semen quality, oxidant–antioxidant status and DNA-damage status of frozen Mesopotamian catfish semen. To this end, the sperm was frozen in diluent containing different inositol concentrations (5, 10, 20 and 40 mg). Increasing levels of inositol linearly increased the spermatozoa motility rate and duration significantly (p < 0.05). High doses of inositol increased antioxidants and decreased oxidants. In addition, lower intracellular DNA damage and percentage of apoptotic spermatozoa occurred in high doses of inositol (p < 0.05).Abstract: In this study, the effects of supplemented inositol on sperm extenders were examined on the spermatozoa motility rate and duration, total antioxidant and oxidant status, apoptotic spermatozoa and DNA damage, during the sperm post-thaw process of Mesopotamian Catfish (Silurus triostegus, H. 1843). The semen was frozen in diluents containing different inositol concentrations (5, 10, 20 and 40 mg). Increasing levels of inositol linearly improved the spermatozoa motility rate and duration significantly (p < 0.05). MDA and TOS were linearly decreased, however, TAS and GSH linearly increased (p < 0.05). The increasing inositol levels resulted in a linear and quadratic decrease in DNA damage in the comet assay, 8-hydroxydeoxyguanosine and the determined percentage of apoptotic spermatozoa (p < 0.05). These results suggest that there are many positive effects of the use of supplemental inositol on enhancing sperm cryopreservation efficiency in Silurus triostegus.

According to global data, there is a male reproductive potential decrease. Pathogenesis of male infertility is often associated with autoimmunity towards sperm antigens essential for fertilization. Antisperm autoantibodies (ASAs) have immobilizing and cytotoxic properties, impairing spermatogenesis, causing sperm agglutination, altering spermatozoa motility and acrosomal reaction, and thus preventing ovum fertilization. Infertility diagnosis requires a mandatory check for the ASAs. The concept of the blood–testis barrier is currently re-formulated, with an emphasis on informational paracrine and juxtacrine effects, rather than simple anatomical separation. The etiology of male infertility includes both autoimmune and non-autoimmune diseases but equally develops through autoimmune links of pathogenesis. Varicocele commonly leads to infertility due to testicular ischemic damage, venous stasis, local hyperthermia, and hypoandrogenism. However, varicocelectomy can alter the blood–testis barrier, facilitating ASAs production as well. There are contradictory data on the role of ASAs in the pathogenesis of varicocele-related infertility. Infection and inflammation both promote ASAs production due to “danger concept” mechanisms and because of antigen mimicry. Systemic pro-autoimmune influences like hyperprolactinemia, hypoandrogenism, and hypothyroidism also facilitate ASAs production. The diagnostic value of various ASAs has not yet been clearly attributed, and their cut-levels have not been determined in sera nor in ejaculate. The assessment of the autoimmunity role in the pathogenesis of male infertility is ambiguous, so the purpose of this review is to show the effects of ASAs on the pathogenesis of male infertility.

The process of semen freezing causes an increase in free radicals concentration which can damage spermatozoa. The addition of natural ingredients in semen diluent is expected to solve this challenges. One of the natural ingredients that can be used is jamblang (Syzygium cumini) leaves. The objective of the current study was to investigate the quality of spermatozoa in Aceh cattle which was added with jamblang leaves extract in skim milk- egg yolk extender during pre-freezing and post-thawing. This study applied Completely Randomized Design (CRD) with 5 treatments and 5 replications. The treatments consisted of J0 = skim milk-egg yolk; J1 = skim milk-egg yolk + jamblang leaves extract 0.2%; J2 = skim milk-egg yolk + jamblang leaves extract 0.4%; J3 = skim milk-egg yolk + jamblang leaves extract 0.6%; and J4 = skim milk-egg yolk + jamblang leaves extract 0.8%. The parameters observed in this study were the percentage of motility and viability of frozen semen of Aceh cattle. The data obtained were analyzed using Analysis of Variance (ANOVA) and if differences were found, then it would be continued with Duncan's Multiple Distance test. The results showed that the addition of jamblang leaves extract in egg yolk skim milk significantly affected the percentage of motility during pre-freezing and post-thawing, significantly affected spermatozoa viability during pre-freezing and significantly affected the spermatozoa viability during post-thawing. J3 treatment (jamblang leaves extract 0.6 gram/100 ml) it should be higher than the other treatment, where the percentage of motility at pre-freezing and post-thawing were 55.48% and 52.71%, respectively, and the percentage of viability during pre-freezing and post-thawing were 56.59% and 53.94%, respectively. It was concluded that the addition of jamblang leaves extract in the skim milk-egg yolk extender affected the percentage of spermatozoa motility and viability of Aceh cattle during pre-freezing and post-thawing.
