Biodiversitas 21: 4671-4676.  Cultivation of Citrus reticulata Blanco cv. Tawangmangu, one of leading mandarin cultivars in Indonesia, remains challenging due to the limitations of suitable rootstocks. The use of suitable rootstocks will greatly determine the success of grafting and affect the performance of the scion. This study aimed to evaluate the morphological, anatomical, and metabolic profiles of grafts between Tawangmangu and rootstocks from various species using shoot tip grafting. Observations of morphological, anatomical, and metabolite variation were carried out eight weeks after the beginning of the growth of the scions. The morphological characters were observed on leaves and stems of the scions. Anatomical data were obtained from the examination of longitudinal sections of grafts prepared using paraffin-embedded method. Analysis of metabolite profiles was performed using LC-MS. The results showed that for two of the three rootstocks used, C. ichangensis × C. reticulata cv. Austera (Japansche Citroen (JC)) and Citrus (Fortunella) japonica (Fortunella) scion growth was vigorous and resulted in a large number of leaves and long stems. Morphological variation was found in the leaf shape, as indicated by wider and wavy leaves of grafts with Fortunella, while narrower and straighter leaves were found in grafts with JC. The grafting onto the third rootstock, C. hystrix, induced dwarfing of the scions. Variation in the anatomical profiles was found in vascular vessels, and in exodermic and endodermic cell sizes. Differences in metabolic profiles of the grafts were found compared to scion and rootstocks. Results of this study provide an important contribution to the successful improvement of Tawangmangu.

Representatives of the genus Citrus L., which includes C. limon, C. sinensis, C. reticulata, C. paradisi, C. maxima, C. medica, C. aurantifolia and other species are the most important fruit crops in countries with subtropical and tropical climates. Cross-pollination, variability of sexual offspring fixed by apomixis, preserved during vegetative propagation contributed to the appearance of intergeneric and interspecific hybrids combining characters and properties of different species. The aim of this work was to carry out interspecific crosses to create biodiversity of Citrus reticulata Blanco var. unchiu Tan. and create new forms with breeding valuable traits. Six interspecific combinations of crossings with the involvement of different varieties were carried out, and a large variety of seedlings was obtained. In sexual offspring from interspecific crosses, a high degree of phenotypic variability is observed on the leaf blade, in shape, size, color of the fruit, thickness and separability of the peel, the presence and absence of seeds, as well as organoleptic properties. All these characteristics represent important criteria for the variety. According to the totality of morphological characters, 24 promising forms were selected. The combination of C. reticulata × C. maxima crossing is the most productive, which allowed us to identify the largest number of forms with breeding valuable traits. Forms Yub - P Nmik; Yub – P Nmik - 1; MV - P Azah; MV - P Azah - 1; MV - P Azah - 2; Kr - P Azah; Cr - P Azah – 1 are promising and are important for the creation of new large-fruited varieties of mandarin. From combination of crossbreeding with C. sinensis such forms as KV - ApV, KV-ApV-2, MV-ApV-1were identified, characterized by large-fruited, fruits with high organoleptic properties. Hybrids bred with C. medica, C. aurantifolia, C. limon meyer are important for creating citrus biodiversity with valuable traits.

Considering the huge economic loss caused by postharvest diseases, the identification and prevention of citrus postharvest diseases is vital to the citrus industry. In 2018, 16 decayed citrus fruit from four citrus varieties—Satsuma mandarin (Citrus unshiu), Ponkan (Citrus reticulata Blanco cv. Ponkan), Nanfeng mandarin (Citrus reticulata cv. nanfengmiju), and Sugar orange (Citrus reticulata Blanco)—showing soft rot and sogginess on their surfaces and covered with white mycelia were collected from storage rooms in seven provinces. The pathogens were isolated and the pathogenicity of the isolates was tested. The fungal strains were identified as Lasiodiplodia pseudotheobromae based on their morphological characteristics and phylogenetic analyses using the internal transcribed spacer regions (ITS), translation elongation factor 1-α gene (TEF), and beta-tubulin (TUB) gene sequences. The strains could infect wounded citrus fruit and cause decay within two days post inoculation, but could not infect unwounded fruit. To our knowledge, this is the first report of citrus fruit decay caused by L. pseudotheobromae in China.

RAPD analysis of 19 Ponkan mandarin accessions was performed using 25 random primers. Of 112 amplification products selected, only 32 were polymorphic across five accessions. The absence of genetic variability among the other 14 accessions suggested that they were either clonal propagations with different local names, or that they had undetectable genetic variability, such as point mutations which cannot be detected by RAPD.

O trabalho foi desenvolvido com o objetivo de avaliar o efeito do acúmulo de temperaturas abaixo de 20° C, sobre a floração de tangerineiras 'Ponkan' (Citrus reticulata Blanco). Foi adotado o delineamento inteiramente casualizado, com seis tratamentos e quatro repetições. Os tratamentos foram formados por sete datas de transferência das mudas, mantidas a céu aberto, para a câmara de crescimento. Foram utilizadas plantas de tangerineira 'Ponkan' enxertadas sobre limoeiro 'Cravo', cultivadas em recipientes, que inicialmente foram mantidas a céu aberto. O experimento foi iniciado no dia 16-05-2007 (início do período de redução da temperatura na região) e conduzido até 17-08-2005. Quinzenalmente, quatro plantas que estavam a céu aberto foram transferidas para câmara de crescimento, com temperaturas de 27°C/20°C (dia/noite), umidade relativa de 75%, fotoperíodo de 12 horas e radiação fotossinteticamente ativa de 170 µmol m-2 s-1. Foram avaliados a data do florescimento e o número total de flores de todas as plantas. Quanto maior foi o tempo de permanência das plantas a céu aberto em temperaturas inferiores a 20° C, maiores foram a antecipação do florescimento e o número de flores produzidas.

Os marcadores moleculares apresentam várias aplicações no melhoramento de plantas, permitindo uma série de análises genéticas. Este trabalho foi realizado com o objetivo de estabelecer marcadores RAPD para serem utilizados em estudos de mapeamento genético e na seleção de híbridos entre tangerina-'Cravo' (Citrus reticulata Blanco) e laranja-'Pêra' (C. sinensis (L.) Osbeck). Extraiu-se DNA de folhas dos parentais e de seis híbridos F1. As reações de amplificação foram preparadas em 13 uL de solução, constituída por tampão 1x GIBCO BRL; soluções 1,54 mM de MgCl2 e 0,2 mM de cada dNTP; 15 ng de cada 'primer'; 1,5 unidade de 'Taq DNA Polymerase' e 15 ng de DNA genômico. As reações foram realizadas em termocicladores programados para 36 ciclos de 1 min a 92ºC, 1 min a 36ºC, 2 min a 72ºC e 10 min de extensão a 72ºC. Foram testados 'primers' decâmeros arbitrários dos 'kits' A, AB, AT, AV, B, C, D, E, G, H, M, N, P, Q, R e U da Operon, sendo selecionados 113 por apresentarem polimorfismo, com número de marcadores variando de 1 a 6 por 'primer'. Esses 'primers' amplificaram 201 (23,13%) bandas polimórficas, aplicáveis no mapeamento genético e seleção de híbridos. A freqüência de 'primers' com 1; 2; 3; 4; 5 e 6 bandas polimórficas foi de 49,5%, 33,6%, 9,7%, 4,4%, 1,8% e 1,0%, respectivamente.
