Objective. To establish a quantitative analysis of multicomponents by single marker (QAMS) method for the simultaneous determination of 4 active components such as protocatechuic acid, catechin, quercetin, and luteolin in Plantago depressa. Method. 4 active components in Plantago depressa were studied. Quercetin was used as an internal reference to establish the relative correction factors among protocatechuic acid, catechin, and luteolin and calculate the contents of each component; the results were compared with those measured by the external standard method. Results. 4 components showed a good linear relationship in their respective concentration ranges (r > 0.9995). The relative correction factors (fs/k) of protocatechuic acid, catechin, and luteolin were 1.1992, 0.8613, and 1.6069, respectively. The method had good durability. The contents of protocatechuic acid, catechin, and luteolin calculated by QAMS were not significantly different from those measured by the external standard method. Conclusion. QAMS can be used to determine the content of 4 components in Plantago depressa at the same time, and the method is simple, accurate, and can be used for quality control.

Background: Desheng pills (DSP) consist of six traditional Chinese medicine. This preparation is used fornourishing blood, eliminating stasis, soothing liver and regulating menstruation, and can also be used to treat menoxenia and dysmenorrhea caus ed by qi stagnation and blood stasis. Objective: In this paper, an accurate and sensitive high-performance liquid chromatography-diode array detector (HPLC-DAD) method was developed and validated for simultaneous determination of seven active components (gallic acid, paeoniflorin, costunolide, dehydrocostuslactone, rutin, leonurine hydrochloride and ferulic acid) in the traditional Chinese formula-Desheng pills. Methods: The seven analytes were separated on Agilent ZORBAX SB-C18 column (250mm× 4.6mm, 5μm) maintained at the temperature of 30.. Gradient elution was performed with the mobile phase of methanol (A)-0.1% phosphoric acid solution (B) at the flow rate of 1.0mL·min-1. The analysis was carried out at the wavelength of 225 nm, 256 nm, 277 nm and 320 nm with an injection volume of 10 μL. Results: The measured seven components showed good linear relationships within their own concentration ranges along with coefficients of determination ≥0.9996. The limits of detection and quantitation of all analytes were in the range of 0.19-13.51 μg/mL and 0.59-40.93 μg/mL, respectively. Average recoveries ranged from 98.82% to 102.01% with RSDs of 1.47%-1.99%. The content of tested components was in the range of 0.053-0.421 mg/g. Conclusion: The proposed method was found to be sensitive, accurate and reproducible, which provided an effective quantitative analytical method for quality control of Desheng pills.

Huang-Lian Jie-Du decoction (HLJDD) has been used to treat cardiovascular and cerebrovascular disease for many years in China. Currently, the determination of effect components in HLJDD is focusing either on the formula or on the extract, while quantification of that in biological samples is scarce, especially simultaneous determination of multicomponent. In this paper, a rapid, specific, and sensitive ultra-high performance liquid chromatography-tandem mass spectrometry method was developed and fully validated for the simultaneous determination of seven main active constituents, i.e., baicalin, baicalein, wogonoside, wogonin, berberine, palmatine, jatrorrhizine in rat plasma. The method was also successfully applied to a quantitative study after oral administration of HLJDD at different doses of 1.5, 3, and 6 g/kg body weight to high fat-induced atherosclerosis rats. The analytes were detected by ESI source and multiple reactions monitoring (MRM) using positive scanning mode. The blood was collected from the abdominal aorta of rats at predetermined time and preprepared with icariin and tetrahydropalmatine as internal standards (IS). Sample preparation was achieved by protein precipitation (PPT). The validation parameters (linearity, sensitivity, intra-/interday precision and accuracy, extraction recovery, and matrix effect) were within acceptable ranges, and biological extracts were stable during the entire storing and preparing process. And the result of determination of HLJDD-containing plasma, baicalin, baicalein, wogonoside, and wogonin could be highly detected in a dose-dependent manner while berberine, jatrorrhizine, and palmatine were determined in a very low level and in a dose-independent mode. Thus, the established method was sensitive enough and successfully applied to the determination of seven effective components in plasma taken from 24 high fat-induced atherosclerosis rats after oral administration of three dosages of HLJDD.
