Objective of the study was to perform a comparative analysis of molecular-genetic properties in non-toxigenic Vibrio cholerae O1 strains biovar El Tor, isolated in the Republic of Kalmykia and on cholera endemic territories and to reveal their phylogenetic relations to toxigenic isolates.Materials and methods. We have carried out bio-information analysis of whole genome sequences of 60 cholera vibrio strains from endemic as regards cholera regions and from Kalmykia. The presence of pathogenicity and endemicity islands in their genomes has been determined. Specifed have been the sequence-types of the examined strains and whole genome SNP-analysis conducted.Results and discussion. Non-toxigenic El Tor vibrios circulating in Kalmykia are clustered into two major genotypes: ctxA–tcpA+VPI-2+VSP– and ctxA–tcpA–VPI-2Δ+VSP–, where VPI-2 Δ+ signifes the presence of deletions of varying length in the genome of this pathogenicity island. Only the latter one is found in regions endemic for cholera. In addition, ctxA– tcpA+VPI-2+VSP+ populations circulate in cholera endemic foci, not found in Kalmykia. 17 sequence-types were identifed among the studied strains (by seven housekeeping gene loci). Phylogenetic analysis performed using SNP-typing demonstrated the absence of close genetic relation between the ctxA–tcpA+VPI-2+VSP– vibrios from Kalmykia and both toxigenic and non-toxigenic vibrios with different composition of pathogenicity and pandemicity islands in the genome. At the same time, genetic proximity of ctxA–tcpA–VPI-2Δ+VSP– cholera vibrios from endemic cholera foci with those isolated in Kalmykia was detected, which may indicate the possibility of their recurrent importation into the territory of Russia. Non-toxigenic V. cholerae strains found in the territory of Kalmykia are characterized by a high genetic diversity. Circulation of the strains with unique sequence-types suggests their potential for long-term persistence on this territory. At the same time, phylogenetic closeness and identity of certain strains with strains from endemic territories can be an evidence of repeated importation.

ABSTRACT Here, we present the draft whole-genome sequence of Vibrio cholerae O1 El Tor strains 76 and M3265/80, isolated in Mariupol, Ukraine, and Moscow, Russia. The presence of various mutations detected in virulence-associated mobile elements indicates high genetic similarity of the strains reported here with new highly virulent variants of the cholera agent V. cholerae.

Summary. Introduction: The formation of highly pathogenic strains of Vibrio cholerae resistant to antibiotics determines the pathogenesis, clinical picture, epidemiology, and laboratory diagnosis of cholera. It is necessary to study the structure of the genome of V. cholerae of any serological groups isolated during routine monitoring of environmental objects or patients with acute intestinal infections in order to detect toxigenic genes inherent to epidemic strains of V. cholerae O1. Our objective was to study changes in phenotypic properties and molecular genetic bases of persistence, epidemic and pathogenetic potential of El Tor variant of Vibrio cholerae O1 isolated from patients and environmental objects in the Caucasus in 1970–1998. Materials and methods: We studied phenotypic and molecular genetic characteristics of 32 V. cholerae O1 strains isolated from patients and environmental objects in the Caucasus (1970–1998) using a microbiological assay and PCR test, respectively. Results: We established that the following biotypes of V. cholerae O1 were found in the Caucasus: a) El Tor, ctxA–, and Hly+ nontoxigenic strains of V. cholerae O1, typical in their phenotypic and molecular genetic properties. Their toxigenic potential is represented by exo- and endotoxins different from the enterotoxin of toxigenic V. cholerae; b) toxigenic El Tor, ctxABEL+, rtхC+, and Hly– variant biotypes of V. cholerae O1, typical in their phenotypic, molecular and genetic properties during outbreaks of invasive cholera, producing a CT-2 enterotoxin and containing genes responsible for persistent, pathogenic and pandemic potential, and c) typical in terms of their main phenotypic characteristics but molecularly genetically altered variants (hybrids) of El Tor variant of Vibrio cholerae O1 including El Tor ctxABCL+, rtxC+, Hly–. Conclusions: The main manifestation of evolutionary changes in a typical toxigenic vibrio of the El Tor variant biotype was the replacement of the ctxBEl gene in its genome with the ctxBCL gene encoding the classical type of enterotoxin (CT-1), which significantly increased the persistent, toxic, epidemic, and pandemic potential of genetically modified variants of El Tor biotype Vibrio cholerae. The genome of these strains contains an integrative and conjugative element SXT with genes for antibiotic multiresistance.

No período de 1992 a 1994, foram analisadas 2.585 amostras de águas de diferentes ecossistemas, acrescidas de 91 espécimens de alimentos visando ao monitoramento de Vibrio cholerae O1 no Estado de Pernambuco. Nas 2.676 amostras foram detectadas 193 cepas de Vibrio cholerae O1 (7,21%) com predominância do sorovar Inaba (183-94,8%) sobre Ogawa (10-5,1%), todas classificadas no biotipo El Tor e sensíveis à tetraciclina. Numa parcela de setenta amostras selecionadas ao acaso, mas incluindo todas do sorovar Ogawa, foi evidenciada a produção de toxina colérica. A maior incidência do vibrião colérico em águas de rios, canais e de esgoto, representando 86% dos isolados, indicou a contaminação fecal por excretores como a causa preponderante na disseminação da bactéria nos sistemas aquáticos. Assinala-se a discreta ocorrrência de V. cholerae O1 nos alimentos processados (2,1%).
