In vitro regeneration of Moringa oleifera Lam.: A medicinal tree of family Moringaceae

A protocol is developed for somatic embryogenesis and organogenesis from nodal segments and shoot-apices of Moringa oleifera Lam. Callusing and multiple shoots induction occurred in nodal segments (0.5-1.0 cm) after 2 weeks of culture on Woody Plant Medium, 1981 (WPM) containing N6-benzyladenine (BA), alone or in combinations with various concentrations of á-naphthalene acetic acid (NAA). The concentration of BA at 4.44 μM was found optimum for multiple shoots formation, resulting in 3.22 ± 0.17 shoots per explants. WPM was enriched by adding certain additives to minimize yellowing and shedding of leaves in proliferated shoots. Maximum roots induction (86.6%) was observed in enriched WPM containing 2.46 μM IBA. Somatic embryos were developed from the calli obtained from nodal segments on enriched WPM supplemented with IBA and BA. Histological studies were carried out for studying the pattern of somatic embryogenesis. Multiple shoots formed from shoot apices (1.0-1.5 cm) cultured on enriched WPM (with or without IBA) after 2 weeks of culture. Root induction occurred simultaneously. The regenerated plantlets were gradually transferred to half strength and later to quarter strength liquid WPM, for 10 days, before transferring to pots. Approximately, 70% survival rate was recorded from in vitro grown plantlets on transfer to pots. Somatic embryogenesis in M. oleifera from nodal explants is reported for the first time in this communication.

An efficient rapid in vitro regeneration protocol was described from nodal segment, leaf and petiole explants. MS medium supplemented with 1.0 mg/l BAP and 0.5 mg/l IAA was found best for the multiple shoot formation from nodal segments. In this combination 99% explants produced multiple shoots and the average number of shoots per explants was 20.1 ± 1.96. For petiole and leaf explants best response was observed on MS supplemented with 2.0 mg/l BAP, 1 mg/l IAA and 0.5 mg/l Kn. Petiole explants produced highest mean number of shoots/explant (22.9 ± 1.728) among the three explants when the explants were cultured on MS with 2.0 mg/l BAP, 1 mg/l IAA and 0.5 mg/l Kn. The highest frequency of root induction (100%) and mean number of roots/plantlets (11.75) were obtained on MS. An in vitro regeneration protocol of Adhatoda vasica has been developed using excised nodal segments and juvenile leaves for multiple shoots regeneration directly or through callus induction. Explants were cultured on MS medium with different concentrations of IAA, NAA, BAP, GA3 and Kn singly or in combinations. MS medium supplemented with BAP (10.0 mg/l) was found best for multiple shoot formation, in which 93.33% explants produced multiple shoots. After two months, maximum number of multiple shoots were 10.6 ± 1.82, highest length of plantlets was 5.2 ± 2.20 cm. 100% calli formation were observed on MS medium supplemented with IAA (0.05 mg/l) + NAA (0.05 mg/l) + BAP (1.0 mg/l). Callus initiation started after 14 days and gave light green colored callus. Best callus mediated shoot regeneration was found on MS+10.0 mg/l BAP medium. Root induction of in vitro raised shoots was best on ½ MS + IBA (1.0 mg/l). Well rooted plantlets were transferred to plastic pots containing garden soil and compost in a ratio of 2:1 for hardening. To evaluate an efficient protocol for the micropropagation of Tupistra albiflora K. Larsen, the effects of N6-benzylaminopurine (BA) and naphthalene acetic acid (NAA) concentrations on multiple shoot and root induction were examined. In vitro shoots were used as the explant materials which were cultured on Murashige and Skoog (MS) agar medium supplemented with 0, 1, 2, 3 and 4 mg/L BA for 4 weeks to induce multiple shoots. It was found that the MS medium containing 3 mg/L BA induced 100 % shoot formation with the highest number of 3.2 shoots per explant (2.4-fold significantly higher than the control). For root induction, in vitro shoots were cultured on MS agar medium supplemented with 0, 1, 2, 3 and 4 mg/L NAA for 8 weeks. The results showed that the MS medium containing 1 mg/L NAA induced 100 % root formation with the highest number of 6.6 roots per explant (1.8-fold significantly higher than the control).

An efficient in vitro regeneration system was developed for Rauvolfia serpentina L. through direct and indirect organogenesis from nodal and leaf explants. Among the different growth regulators, MS medium supplemented with 2.0 mg/l BAP, 0.5mg/l IAA and 0.02mg/l NAA found best for the multiple shoot formation from nodal segments. In this combination 98% explants produced multiple shoots and the average number of shoots per explants is 13∙4. The frequency of callus induction and multiple shoot induction from leaves was highest 88% in MS medium supplemented with 2.0 mg/l BAP, where mean number of shoots/explants was 12.5. The highest frequency of root induction (80%) and mean number of roots/plantlets (10) were obtained on half strength of MS medium containing 0.2 mg/l IBA. Agrobacterium-mediated genetic transformation system has been developed for two tomato (Lycopersicon esculentum Mill.) varieties, namely Pusa Ruby (PR) and BARI Tomato-3 (BT-3). Prior to the establishment of transformation protocol cotyledonary leaf explants from the two varieties were cultured to obtain genotype independent in vitro regeneration. Healthy multiple shoot regeneration was obtained from the cut ends of cotyledonary leaf segments for both the varieties on MS containing 1.0 mg/l BAP and 0.1 mg/l IAA. The maximum root induction from the regenerated shoots was achieved on half the strength of MS medium supplemented with 0.2 mg/l IAA. The in vitro grown plantlets were successfully transplanted into soil where they flowered and produced fruits identical to those developed by control plants. Transformation ability of cotyledonary leaf explants was tested with Agrobacterium tumefaciens strain LBA4404 harboring binary plasmid pBI121, containing GUS and npt II genes. Transformed cotyledonary leaf explants were found to produce multiple shoots on MS containing 1.0 mg/l BAP and 0.1 mg/l IAA. Selection of the transformed shoots was carried out by gradually increasing the concentration of kanamycin to 200 mg/l since kanamycin resistant gene was used for transformation experiments. Shoots that survived under selection pressure were subjected to rooting. Transformed rooted plantlets were transferred to soil. Growth regulators especially auxins and cytokinins are critical for plant in-vitro regeneration. The effect of these plant growth regulators on in-vitro propagation of Saccharum officinarum L (Sugarcane) was investigated. In vitro response of two different varieties of sugarcane (NCS 005 and NCS 008) to Plant Growth Regulators was obtained in this study. Formation of buds was obtained on shoot apical meristem when cultured on MS (Murashige and Skoog) medium supplemented with 0.1mg/l BAP (6-Benzylaminopurine). After two weeks of initiation, regenerated meristem was inoculated into MS (Murashige and Skoog) fortified with different concentrations and combination of cytokinins. Shoot multiplication was optimal on 0.5mg/l BAP + 0.25 mg/l Kin(Kinetin) for NCS 005 variety while for NCS 008 variety, no significant (P≥0.05) difference was observed between 1.5mg/l BAP and 1.5mg/l BAP +0.5mg/l Kin. The best root induction for in vitro derived shoots was obtained on 1.0 mg/l NAA (Naphthalene acetic acid) and 2.0 mg/l IBA( Indole butyric acid) for both varieties of sugarcane within ten days of culture transfer. Successfully established plantlets showed excellent growth response when weaned under regulated green house conditions.

Multiple shoots were obtained from both shoot tips and nodal segments cultured on MS fortified with different concentrations of BAP and Kn singly or in combination with low concentration of NAA. Maximum number of shoots (9.28 ± 0.61) were found on MS supplemented with 1.5 mg/l BAP and 05 mg/l NAA. In vitro regenerated shoots were separated and transferred onto half and full-strength MS supplemented with different concentration of IBA, IAA and NAA for root induction. Full strength MS containing 0.2 mg/l IBA was found to be best for root induction where 93.33% shoots were rooted. In vitro regenerated plants grew normally without showing any morphological variation and flowered after 40 days of transplantation.

An efficient in vitro plant regeneration protocol was developed for the medicinally potent plant species Phyllanthus amarus Schum. and Thonn. (Euphorbiaceae) using nodal segment as explant. Maximum multiplication of shoots (15.275±0.96) was achieved on Murashige and Skoog’s medium supplemented with BAP (0.5 mg/l) after 3-4 weeks of inoculation. The shoots were separated from cluster and subcultured for their elongation on the same medium. In vitro flowering was also observed on the elongated shoots after 3–4 weeks of sub culturing on the shoot elongation medium. In vitro regeneration protocol for grass pea (Lathyrus sativus L.) was optimized using different concentrations and combinations of growth regulators. Direct shoot regeneration obtained through shoot organogenesis from different explants of grass pea cultured on MS medium supplemented with Gamborg B5 vitamin containing 6-benzylaminopurine (BAP), Thidiazuron (TDZ) and ?-naphthalene acetic acid (NAA). Highest percentage of shoots were obtained at 4.0 mg/l of BAP on nodal explants. Stunted multiple shoots were developed from nodal explants while 1.5 mg/l TDZ was used. About 56% of direct shoots were also obtained, while the combination of BAP (4.0 mg/l) and NAA (0.5 mg/l) were used. Regenerated plantlets were rooted most effectively (40%) in rooting medium containing half strength of MS basal medium containing 1.0 mg/l NAA. Well rooted plantlets were further successfully acclimatized to ambient humidity level and grown in controlled environment until hardening.Jahangirnagar University J. Biol. The present research was carried out in Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-e-Bangla Nagar, Dhaka-1207 from the period of September 2017 to June 2018. This research aims to study the effect of Benzyladenine (BA), Kinetin (KIN) and Naphthalene acetic acid (NAA) either in combination or alone on In vitro propagation of papaya (Carica papaya). The shoot tips of young shoots were used as explant, which was sterilized using freshly prepared 0.1% HgCl2 mixing with few drops of Tween-20, were inoculated in MS media supplemented with 0.1% activated charcoal. The minimum days to shoot induction (10.25) were recorded on MS medium containing 0.5 mg/L BA. The highest shoots (4.5) and length of shoot (5.75 cm) observed in 1.0 mg/L BA. The combined treatment 1.0 mg/L BA+0.75 mg/LKIN gave the highest number of shoots (5.25) and length of shoot (5.78 cm).The minimum days (8.5) to root induction was reported in 2.0 mg/L NAA along with maximum 8.25 roots per plantlet. The highest length of root (6.92 cm) was observed in 2.0 mg/L NAA. In regenerated plantlets, 80% survival rates were observed in growth chamber conditions and 75% in the open atmosphere were achieved. Finally, the in vitro regeneration protocol described herein can potentially be used as a tool in molecular breeding programs for the improvement of different cultivars and genotypes of papaya.
