Effects of different hormonal concentration on multiple shoot formation of RABI Strawberry-3 were studied. Where the explants were runner tips that cultured on MS basal medium supplemented with 6-Benzyl Adenine and Kinetin. The maximum percentage 80% of shoot was obtained from Benzyl Adenine concentrations of 0.5 mg/l and 1.0 mg/l whereas Kinetin concentrations were 1.5 mg/l and 2.0 mg/l separately. More promising result was found by combining two impressive concentrations of each hormone. When MS basal media was supplemented with Kinetin 2.0mg/l + Benzyl Adenin A 0.5 mg/l, Kinetin 1.5 mg/l + Benzyl Adenine 0.5 mg/l and Kinetin 1.5 mg/l + Benzyl Adenine 1.0 mg/l respectively then 100% shoots were obtained. In vitro propagation systems by means of areole activation were developed for Echinocereus knippelianus, Echinocereus schmollii, Mammillaria carmenae, M. carmenae fo. rubrisprina, M. herrerae, M. theresae, Melocactus curvispinus, Escontria chiotilla and Polaskia chichipe. In vitro germinated seedlings were used as source of explants. Multiple shoot formation from areoles was achieved on MS basal medium supplemented with 3% sucrose, 10 g L-1 agar and 6-benzylaminopurine (BA) or 6-(, -dimethylallylamino)purine (2iP). Efficiencies ranged from 6.0 shoots per explant in M. carmenae fo. rubrisprina to 13.5 shoots per explant in Echinocereus schmollii. Rooting of the in vitro generated shoots was achieved in MS basal medium, or MS basal medium supplemented with indoleacetic acid, indolebutyric acid or activated charcoal. Finally, 49-98% of these plants survived.

The effects of different hormonal concentration on shoot formation and callus induction were studied on BARI Malta-1 (Citrus sinensis). Seeds containing seed coat and without seed coat was treated by Murashige and Skoog (MS) media supplemented with 6-benzyl adenine (BA) and Kinetin (KIN). Removal of seed coat showed an early response for shoot formation. The highest (70%) shoot formation was obtained from seeds without seed coat treated with MS basal media + BA 1.0 mg/l while KIN showed no response for shoot formation in any supplemented concentration. However, in case of callus induction internodes and apical shoot tips were used as explants and 2, 4-dichlorophenoxy acetic acid (2, 4-D) was used as callus inducing hormone. The morphogenetic potential of parval or pointed gourd (Trichosanthes dioica Roxb.) shoot-tip explants was investigated to establish this species as a model tissue culture system. An effective multiple-shoot propagation method is described. Ten-millimeter shoot tips from young branches of greehouse-grown plants served as explants. They were initiated on a MS basal medium. Multiple shoots were encouraged by transferring established explants to a proliferation medium consisting of MSB + 1 mg BAP/liter, because lower concentrations of BAP (0.1 to 0.5 mg–liter–1) inhibited multiple shoot formation; however, the same concentrations promoted rooting in explants. Medium supplemented with 1 mg BAP/liter and 100 mg PVP/liter caused the best proliferation of shoot tips. Upon transferring to fresh medium of the same composition, these shoot tips elongated 24 cm with three to five nodes in 4 weeks of culturing. Shoot multiplication cultures were maintained by transferring segments of multiple-shoot clusters to medium containing 1 mg BAP/liter and 0.5 mg GA3/liter. Medium supplemented with TDZ inhibited the number of regenerating explants but enhanced the number of shoot buds. Eighty percent of these plantlets were successfully rooted on MS medium supplemented with 1 mg NAA/liter. Plantlets survived in potting soil and exhibited normal growth under mist in the greenhouse.

A micropropagation protocol for an endangered slipper orchid species, Cypripedium formosanum Hayata, through axillary buds from adult plants has been developed. The season of explant collection is crucial for the initial success of an aseptic culture. Explants collected in the middle of January gave the highest percentage of explant survival (54.2%) and shoot-forming percentage (41.7%). Of the two cytokinins tested, N6-benzyladenine (BA) was found to be superior to thidiazuron for normal shoot formation. The optimum result was obtained in quarter-strength Murashige and Skoog medium containing 22.2 or 44.4 μM BA in which the cultures produced 6.3 and 7.1 shoots per explant with 10.6 to 11.7 mm average length after 90 d of culture. Regenerated shoots rooted for 60 d in the basal medium with 1 g·L−1 activated charcoal and 20 g·L−1 potato homogenate were ready for growth in pots. This is the first report on shoot multiplication in vitro from mature plants of Cypripedium that provides a reliable method for propagating the selected elites.

In vitro culture is a promising technique for mass propagation of high-value species. Study of propagation for Acacia hybrid (A. mangium x A. auriculiformis) through this technique has been conducted using single node stem from seedlings as explants. Growth medium used was modified Murashige and Skoog (MS), basal medium Woody Plant Medium (WPM), and Gamborg (B5) supplemented. The study was conducted in two stages, namely shoot induction and shoot multiplication. The treatment tested was the Benzyl Adenine (BA) supplementation at the concentration of 0.3; 0.7; and 1.0 mgL-1 of. Observation was conducted on the frequency of shoot induction, number of shoot, shoot length and visual performance of the culture. The result showed that treatment of BA 0.7 mgL-1 on modified MS medium is the best for shoot induction, shoot multiplication and visual performace of the culture. The average of number of shoot was 2.6; 5.0 and 7.7 shoots on the first three consecutive subcultures. Changing to different basal medium on the fourth subculture showed that the treatment of BA 0.7 mgl-1 is the best condition for shoot regeneration (12.60 shoots) and shoot length (6.97 cm). The culture from this treatment showed the best visual morphological performance.Keywords:Acacia hybrid; multiplication; subculture; in vitro; BA. ABSTRAKKultur in vitro merupakan suatu teknik yang menjanjikan untuk perbanyakan massal spesies-tanaman bernilai tinggi. Penelitian perbanyakan akasia hibrid (A. mangium x A. auriculiformis) melalui kulturin vitro telah dilakukan dengan menggunakan eksplan berupa batang satu buku yang berasal dari anakan. Media tumbuh yang digunakan adalah media dasar Murashige dan Skoog (MS) yang sudah dimodifikasi, media dasar Woody Plant Medium (WPM), dan Gamborg (B5). Penelitian dilakukan dalam dua tahap yaitu induksi tunas dan perbanyakan tunas. Perlakuan yang diuji adalah penggunaan Benzyl Adenine (BA) dengan konsentrasi 0,3; 0,7 dan 1,0 mg L-1. Pengamatan dilakukan terhadap waktu induksi tunas, jumlah tunas, tinggi tunas dan penampilan biakan secara visual. Hasil penelitian menunjukkan bahwa penggunaan BA 0,7 mg L-1 pada media MS modifikasi merupakan perlakuan terbaik untuk induksi tunas, perbanyakan tunas, tinggi tunas, dan kondisi biakan secara visual. Jumlah rata-rata tunas yang dihasilkan dari perlakuan ini adalah 2,6; 5,0 dan 7,7 tunas pada subkultur pertama, kedua dan ketiga. Pada penggunaan media dasar berbeda pada subkultur keempat menunjukkan bahwa perlakuan BA 0,7 mg L-1 merupakan perlakuan terbaik dengan jumlah tunas sebanyak 12,60 tunas dan rata-rata tinggi tunas 6,97 cm. Biakan yang dihasilkan dari perlakuan tersebut mempunyai penampilan yang baik dan normal.
