Somatic embryos from Eastern redbud show a high degree of malformation during development and a low conversion rate to seedlings. This problem is common with somatic embryo systems, especially with legume species. A procedure for multiple shoot formation from somatic embryo explants of Eastern redbud was developed that bypasses the need for germination to recover plantlets. Somatic embryo explants cultured on DKW medium containing benzyladenine (BA) and thidiazuron (TDZ) produced more shoots than either treatment alone. The highest number of shoots (3.3 to 3.4 shoots per explant) was obtained from partially desiccated and wounded explants treated with a combination of 5 or 10 M BA and 0.5 or 1.0 M TDZ for 20 days before being transferred to the same medium without TDZ. The number of shoots formed was increased from 1.5 to 3.2 shoots per explant by cutting through the cotyledonary node prior to culture. In addition, the frequency of explants forming shoots was increased by desiccation of somatic embryo explants to ≈50% moisture and by using somatic embryos with two well-formed cotyledons as explants.

Multiple shoot formation and in vitro flowering was found in Brassica campestris (L.) var. Bhavani. Maximum numbers of shoots were produced in both cotyledonary node and shoot apex explants on MS-media supplemented with BA (2.5 mg/l) + IAA (1.0 mg/l) + Kn (0.5 mg/l). Maximum flowering (50%) was noted at IBA (1.5 mg/l) + IAA (1.0 mg/l) + Kn (0.5 mg/l) in the shoots from cotyledonary nodes. In vitro flowering may contribute in many ways to Brassica Improvement Programs. The shoots rooted well in the half and full strength media each with IBA (1.0 mg/l) and NAA (1.0 mg/l) and the plantlets have been maintained. Effects of different hormonal concentration on multiple shoot formation of RABI Strawberry-3 were studied. Where the explants were runner tips that cultured on MS basal medium supplemented with 6-Benzyl Adenine and Kinetin. The maximum percentage 80% of shoot was obtained from Benzyl Adenine concentrations of 0.5 mg/l and 1.0 mg/l whereas Kinetin concentrations were 1.5 mg/l and 2.0 mg/l separately. More promising result was found by combining two impressive concentrations of each hormone. When MS basal media was supplemented with Kinetin 2.0mg/l + Benzyl Adenin A 0.5 mg/l, Kinetin 1.5 mg/l + Benzyl Adenine 0.5 mg/l and Kinetin 1.5 mg/l + Benzyl Adenine 1.0 mg/l respectively then 100% shoots were obtained.
