A New and Reproducible Surface-Enhanced Resonance Raman Scattering Method for the Determination of Trace Au(III) in Solution Using 4-Mercaptopyridine as Molecular Probe

In pH 6.5 Na2HPO4-NaH2PO4 buffer solution and in the presence of stable reagent PEG10000 and aggregation reagent NaCl, nanosilvers were aggregated to form stable aggregated-nanosilvers (ANS). The molecular probe of 4-mercaptopyridine (MP) adsorbed on the surface of ANS to form MP-ANS conjugates that produce strongest surface-enhanced resonance Raman scattering (SERRS) peak at 1097 cm-1. When Au(III) was added, the SERRS intensity at 1097 cm-1 decreased. The decreased SERRS intensity was linear to Au(III) concentration in the range of 5-50 ×10-10 mol/L, with a detection limit of 2.0×10-10 mol/L Au(III).

In pH 6.6 Na2HPO4-citric acid buffer solution and in the presence of KCl, the nanosilver-labeled rabbit anti-hCG (Ag-RAhCG) was aggregated un-specifically to the aggregations. Upon addition of rhodamine 6G (RhG) molecular probe, it adsorb on the surface of Ag-RAhCG aggregations that exhibited the strongest surface-enhnaced resonance Raman scattering (SERRS) peak at 613 cm-1. In the optimal condition, the decreased SERRS intensity responds linearly with the concentration of hCG over 0.05-1.75 µg/mL. Based on this, a new and simple SERRS method has been proposed for the determination of hCG in serum samples, with satisfactory results.

A stable nanosilver solution was prepared, using PEG10000 as stabilizer and NaBH4 as reducer. In pH 6.6 Na2HPO4-NaH2PO4 buffer solution containing PEG10000 and NaCl, nanosilvers were aggregated to form the stable aggregated-nanosilvers (ANS) that could conjugate with 4-mercaptopyridine (MP) to produce an ANS-MP surface-enhanced resonance Raman scattering (SERRS) probe, which exhibited a strong SERS peak at 1097 cm-1. When cystine (Cy) concentration increased, the SERS intensity at 1097 cm-1 decreased linearly. In the optimal condition, the decreased SERRS intensity was linear to Cy concentration in the range of 5.0×10-9-1.4×10-7 mol/L, with a detection limit of 2.0×10-9 mol/L Cy.

Colloidal Au nanoparticles as active substrate for surface-enhanced resonance Raman scattering(SERRS) were prepared by the trisodium citrate-reduced procedure. In pH 6.6 Na2HPO4-NaH2PO4 buffer solution and in the presence of aggregation reagent NaCl, nanogolds were aggregated to form stable aggregated- nanogolds (ANG). The crystal violet (CV) adsorbed on the surface of ANG to form CV-ANG conjugates that produce strongest surface-enhanced resonance Raman scattering peak at 1616 cm-1. In the optimal condition, the SERRS intensity at 1616 cm-1 was linear to the CV concentration in the range of 2.5×10-8 -1.75×10-7 mol/L, a surface-enhanced resonance Raman scattering assay was set up for detection of trace CV, with good selectivity.

In this paper, a facile large-scale preparation of surface-enhanced resonance Raman scattering (SERRS) substrates for the determination of Rhodamine B (RhB) based on silver nanowires (Ag NWs) has been developed. The morphology, structure, and properties of as-prepared Ag NWs are characterized using ultraviolet–visible (UV-Vis) spectroscopy, field emission scanning electron microscopy (FE-SEM), and X-ray diffraction (XRD), respectively. Ag NWs were assembled onto glass slides through a self-assembly method. Moreover, in our experiment, as-prepared Ag NWs@glass were used as a SERRS substrate to detect RhB at the excitation wavelength of 532 nm. Experimental conditions such as pH value and soaking time on SERRS performance were studied and optimized. Under the optimized conditions, the SERRS intensity at 1648 cm−1 exhibited a linear relationship with the concentration of RhB in the range of 1.0 × 10−9–1.0 × 10−5 mol L−1 and detection limit (signal-to-noise ratio [S/N] = 3) is as low as 0.3 nmol L−1. The corresponding correlation coefficient of the linear equation was 0.996. This method based on Ag NWs@glass for the detection of RhB in three kinds of condiment was investigated. The limits of detection (LODs) for RhB were 0.35 µg/g in chili powder, 0.14 µg/g in chili sauce, and 0.02 µg/g in Chinese prickly ash. The relative standard deviations (RSD) were between 2.18% and 4.56% (n = 3) and recoveries at three levels were in the range of 80.0–98.7% for different spiked food products. Moreover, the results showed that the proposed method was sensitive, convenient, and feasible for the determination of RhB in condiments.
