Sintering of Beta-Tricalcium Phosphate Scaffold Using Polyurethane Template

Porous beta-tricalcium phosphate (β-TCP) bioceramic has been reported as synthetic graft for cancellous bone substitute due to its biocompatibility and biodegradability properties. A highly porous and interconnected porosity architecture of bone scaffold facilitates attachment and in-growth of new bone tissue. β-TCP foam, a porous 3-dimensional scaffold was fabricated by employing polymeric foam replica method in this study. Polyurethane (PU) foam was used as the sacrificial template, in which β-TCP slurry with powder to water ratio of 10g:10ml was coated on PU template and sintered to 1100, 1200, 1250 and 1300°C. Observation on architecture of the foam, macrostructure and microstructure of pores and surface topography of porous strut showed that sintering at 1250°C produced sufficient densification of grains and micropores on the β-TCP strut. The β-TCP foams exhibited high porosity (92 – 97%) and large pore size (200 - 750um) that resemble cancellous bone structure.

Cancellous bone plays an indispensable role in the skeletal system due to its various functions and high porosity. In this work, chitosan and hydroxyapatite nanowires (CS@HAP NWs) hybrid nanostructured scaffolds with suitable mechanical properties, high porosity and a fine porous structure were prepared to simulate the 3-dimensional structure of cancellous bone. The 3D-hybrid scaffolds promote cell adhesion and the migration of human adipose-derived stem cells (hADSCs) inside the scaffolds. The cavities in the scaffolds provide space for the hADSCs proliferation and differentiation. Moreover, the various contents of HAP and the induced mechanical property changes regulate the differentiation of hADSCs toward osteoblasts. Overall, cellular fate regulation of hADSCs via rationally engineered HAP-based hybrid scaffolds is a facile and effective approach for bone tissue engineering.

The aim of this report is to determine the long-term histologic and histometric evaluation of beta-tricalcium phosphate (beta-TCP) and bovine cancellous bone grafted into the left maxillary sinus of a patient in order to augment the sinus floor to prepare it for receiving an implant. Beta-TCP and bovine cancellous bone (BCB) were grafted into the left maxillary sinus at the extracted sites of maxillary left first and second molars of a 62-year-old male patient. Using osteotome technique, 45 months later, core biopsies were taken for histologic and histometric comparison of the newly formed bone. Histometric and histologic evaluation of both specimens revealed a normal maxillary bone structure with 23.08% calcified bone and 76.92% marrow and connective tissue for the beta-TCP grafted site, and 24.81% calcified bone with 75.19% marrow and connective tissue for the BCB grafted site. No residual grafting material was seen in either specimen. Both beta-TCP and BCB were completely resorbed and replaced by new bone 45 months after grafting. Histologic results of the newly formed bone were similar for both grafting materials.
