As part of a five years monitoring project “Water Quality Monitoring of the Strait of Istanbul”, February-December 1999 nutrient dynamics of the Black Sea-the Sea of Marmara transect are studied to evaluate the effect of discharges given by deep disposals. Through a one-year study, upper layer nutrient concentrations were generally under the effect of northwestern-shelf Black Sea originated waters. This effect was strictly observed in July, when the upper layer flow was the thickest. On the other hand, partly in November but especially in December the northwestern-shelf Black Sea originated water flow was a minimum resulting in similar concentrations in both layers. Nutrient fluctuations also affected the chlorophyll a and POC concentrations as parameters of productivity. The nutrient concentrations decreased with the effect of spring bloom and highest chlorophyll a values were detected in November at Strait stations that did not match to the Sea of Marmara values. This fact represents the time-scale difference between the Black Sea and the Sea of Marmara. On the contrary, high nutrient concentrations in the lower layer (especially inorganic phosphate), and therefore low N:P ratios reflect the effect of deep discharge. Vertical mixing caused by meteorological conditions of the shallow station (M3) under the effect of surface discharges resulted in homogenous distribution of nutrients. Nutrient concentrations of the stations affected by deep discharge showed that the two-layer stratification of the system did not permit the discharge mix to the upper layer.

Abstract Carbon and nitrogen stable isotope ratios are increasingly used to study long-term change in food web structure and nutrient cycling. Whether isotope composition in primary producers and consumers (so-called isotope baselines) reflect environmental changes in a similar manner is largely unknown. We take advantage of long-term oceanographic monitoring data and archived biological samples for the well-studied Baltic Sea to retrospectively analyse elemental composition (C, N and P) and stable isotopes (δ13C, δ15N) in the filamentous ephemeral macroalgae Cladophora spp. and in blue mussel Mytilus edulis trossulus from three contrasting regions (coastal Bothnian Sea and Baltic proper, open sea central Baltic) with the aim to statistically link the observed spatial and interannual (8–24 years’ time-series) variability in elemental and isotope baselines with environmental changes. We find clear differences in isotope baselines between the two major Baltic Sea basins. However, the temporal development in Mytilus δ13C was remarkably similar among regions and, at the open sea station, mussels and algae δ13C also correlated over time, likely reflecting a global Baltic Sea or Northern Hemisphere pattern. In contrast, δ15N of both taxa responded to regional and local drivers (water nutrient concentrations). δ15N in source amino acids allowed detection of diazotrophic N signal in Mytilus, which was masked in bulk δ15N. Finally, Cladophora N:P reflected regional nutrient levels in the water while P %, which differed for both taxa, was linked to food quality for Mytilus. This study highlights the potential of a multi-taxa and multi-stable isotope approach to understand nutrient dynamics and monitor long-term environmental changes.

Cell culture systems have become increasingly popular as a means of screening toxic agents and studying toxic mechanisms of drugs and other chemicals at the cellular and subcellular levels. These in vitro tests can be conducted rapidly in a broad range of relevant mammalian culture systems; a variety of biological and biochemical cytotoxicity endpoints can be examined. The following study utilized human keratinocytes to evaluate the relative cytotoxicities of nitrofurazone (NF) and silver sulfadiazine (SS), the active ingredients of FURACIN(R) Topical Cream and SILVADENE(R) Cream, respectively. These compounds are anti-infectives used in the treatment of burn patients. Cell ultrastructure and elemental composition were utilized as cytotoxicity endpoints.Normal Human Epidermal Keratinocytes (HK) were prepared from the EpiPackTM culture system (Clonetics Corporation, Boulder, CO). For scanning electron microscopy (SEM) and transmission electron microscopy (TEM), cells were seeded on sterile 35 mm Falcon plastic dishes; for elemental microanalysis, cells were plated on polished pyrolytic carbon discs (E. Fullam, Latham, NY) placed in the culture dishes.

The elemental composition of amino acids is similar to that of the major structural components of the epithelial cells of the small intestine and other tissues. Therefore, their subcellular localization and concentration measurements are not possible by x-ray microanalysis. Radioactive isotope labeling: I131-tyrosine, Se75-methionine and S35-methionine have been successfully employed in numerous absorption and transport studies. The latter two have been utilized both in vitro and vivo, with similar results in the hamster and human small intestine. Non-radioactive Selenomethionine, since its absorption/transport behavior is assumed to be the same as that of Se75- methionine and S75-methionine could serve as a compound tracer for this amino acid.
