Availability of quality seeds in production of krisan (Chrysanthemum morifolium Ramat cv Dewi ratih) cultivation is still rare, therefore research on seed multiplication through tissue culture is needed. The media used in tissue culture is relatively expensive for home industry. This study aims to determine the respond of leaf and stem explants using foliar fertilizers (Growmore, Gandasil D and Mutiara) as an alternative media for callus inductions. This study used a Completely Randomized Design (CRD) consisted of 4 treatments: P0: ½ MS + 0,25 mg/l BAP, P1 (Growmore + 0,25 mg/l BAP), P2 (Gandasil D + 0,25 mg/l BAP), P3 (Mutiara + 0,25 mg/l BAP). The variables observed in this study included callus appearance time, callus color and callus texture. The result of this study indicated that the use of BAP (6-Benzyl Amino Purine) affected the time of callus formation and callus morphology. Callus was formed on leaf explants 13 days after planting while on stem explants 7 days after planting and compact texture. Growmore + 0,25 mg/l BAP treatment yields the best callus on leaf explant, while Gandasil D + 0,25 mg/l BAP treatment yields the best callus on stem explant.

Plant preservative mixture (PPM) is a new broad-spectrum biocide that may be useful for plant tissue culture. The objective of this study was to determine if PPM interfered with adventitious shoot regeneration on leaf explants from several plant species. Leaf explants from Dendranthema grandiflora `Iridon', Betula pendula, Rhododendron catawbiense var. album and R.c. `America' were made from the top two apical leaves on the microshoots. In the first experiment, 0, 0.5, 1, 2, or 4 mL·L-1 PPM were added to species-appropriate regeneration media. In the second experiment, only mum leaf explants were placed on regeneration media containing 0, 0.1, 0.2, 0.3, or 0.4 mL·L-1 PPM. The percentage of explants forming shoots and the number of shoots per regenerating explant were recorded after 4, 6, and 10 weeks, for mum, birch, and rhododendron leaves, respectively. The percentages of shoot regeneration from birch and rhododendron leaf explants were unaffected by up to 4 mL·L-1 PPM, and the number of shoots formed per R.c. album explant were also unaffected by the tested concentrations of PPM. In contrast, the numbers of shoots formed on birch and `America' explants were reduced 48% and 25%, respectively, when 4 mL·L-1 PPM was used in the media. The percentages of shoot regeneration and number of shoots per explant were drastically reduced on mum explants when only 0.5 mL·L-1 PPM was used in the medium. In fact, 0.3 mL·L-1 PPM or higher reduced shoot formation by more than 5-fold. This study demonstrates that the effects of PPM on shoot regeneration from leaf explants are species specific.

Meningkatnya minat masyarakat terhadap tanaman krisan tidak diimbangi dengan meningkatnya produksi tanaman ini. Salah satu kendala dalam pengembangan produksi benih krisan dari hasil perbanyakan dengan setek adalah perakaran yang lemah. Perbanyakan secara vegetatif melalui setek permasalahan pokoknya adalah pada pembentukan akar. Penelitian dilaksanakan dari bulan November 2018 sampai bulan Januari 2019. Penelitian dilaksanakan di Greenhouse Kebun Percobaan Balai Penelitian Tanaman Hias Jl. Raya Ciherang, Segunung, Pacet, Cianjur, Jawa Barat 43252. Penelitian menggunakan Rancangan Acak Kelompok (RAK) faktorial dengan 2 faktor. Faktor pertama jenis varietas krisan pot yaitu Varietas Avanthe (V1), Varietas Prita (V2) dan Varietas Naura (V3).dan faktor kedua jenis ZPT yaitu Kontrol (Z1), Rootone F (Z2) dan PGPR (Z3). Interakasi antara aplikasi jenis ZPT dan setek 3 varietas krisan pot pada pengamatan 21 HST memberikan pengaruh tidak berbeda nyata terhadap semua parameter, kecuali pada parameter tinggi tanaman dan jumlah daun. Interaksi terbaik ditunjukkan oleh perlakuan penggunaan Rootone F pada Varietas Avanthe.
