Some 5, 7-diaryl-1,5-dihydro (or 1, 2, 3, 5-tetrahydro)- pyrano[2, 3-d] pyrimidin-2, 4- diones (or 2-thioxo-4-ones) (3a-g) has been synthesized in one-step by the cyclocondensation of barbituric acid or thiobarbituric acid (1) with arylideneacetophenones (2a-d), in glacial acetic acid in the presence of phosphorous pentoxide. The structures of the compounds 3a-g were determined by their UV, IR, 1H NMR, 13C NMR, mass spectral data and elemental analyses. Abstract An improved colorimetric method for furfural in citrus juices, which is based on the well known aniline-acetic acid reaction with furfural (Stenhouse reaction), is described. Addition of SnCl2 and HCl yielded higher intensity and improved color stability. The reagent giving the best results contained 1% SnCl2, 0.6N HCl, and 10% aniline in glacial acetic acid.

Abstract A modification of the Zak cholesterol reaction is described in which ethanol is substituted for glacial acetic acid. This modification avoids the hazard of purifying glacial acetic acid and also increases the specificity of the reaction by eliminating interference from bromide or iodide salts. The ethanolic reaction has the absorbance spectrum, sensitivity, and stability of the usual acetic acid reaction. A rapid ultramicro method has been developed based on this ethanolic reaction. A 0.01- to 0.05-ml. sample of serum is extracted with 2.5 ml. of ethanol, and a 2.0-ml. aliquot of this extract is reacted directly with 2.0 ml. of a Zak color reagent. Cholesterol concentrations as determined by this procedure and by the Sperry-Webb method showed satisfactory agreement for monkey and human sera, including normal, hemolyzed, high cholesterol, and severely lipemic or jaundiced sera..

A simple and highly sensitive spectrophotometric method was developed for the determination of thiobarbituric acid reactive substances (TBARS) as a marker for lipid peroxidation in fried fast foods. The method uses the reaction of malondialdehyde (MDA) and TBA in the glacial acetic acid medium. The method was precise, sensitive, and highly reproducible for quantitative determination of TBARS. The precision of extractions and analytical procedure was very high as compared to the reported methods. The method was used to determine the TBARS contents in the fried fast foods such as Shami kebab, samosa, fried bread, and potato chips. Shami kebab, samosa, and potato chips have higher amount of TBARS in glacial acetic acid-water extraction system than their corresponding pure glacial acetic acid and vice versa in fried bread samples. The method can successfully be used for the determination of TBARS in other food matrices, especially in quality control of food industries.

SummaryTo develop a simple method for estimation of platelet lipoxygenase (PLO) and cyclo-oxygenase (PCO) pathways, the arachidonic acid (AA) metabolism of human platelet was investigated under various experimental conditions by the use of the thiobarbituric acid (TBA) reaction and a radioisotope technique. A TBA-reactive substance different from malondialdehyde (MDA) via PCO pathway was detected and shown to be derived from the PLO pathway. Since the optimal pH and time course of its formation were different from those of MDA formation via PCO pathway, PLO and PCO pathways were estimated by quantitating the TBA-reactive substances produced by the incubation of AA either with aspirin-treated platelets or with untreated ones, respectively, each under optimal conditions. Normal values expressed in terms of nmol MDA/108 platelets were 1.17±0.34 (M±SD, n = 31) and 0.79±0.15 (n = 31) for PLO and PCO pathways, respectively.
