In vitro adventitious shoot organogenesis and plant regeneration from seedling explants of Albizia odoratissima L.f. (Benth.)

Dissected white ash seeds were placed on an agar-solidified MS medium with 10 μM TDZ and 1 μM 2,4-D (shoot initiation medium). After 4 weeks, explants were transferred to shoot elongation medium (3 μM TDZ, 1 μM BA, and 1 μM IBA) solidified with 0.7% Sigma agar, 0.525% agar + 0.05% gelrite, 0.35% agar + 0.1% gelrite, 0.175% agar + 0.15% gelrite, 0.2% gelrite, or no gelling agent (liquid medium). After 12 weeks in vitro, shoot growth and number were suppressed in cultures containing 0.2% gelrite (9.3 mm and 0.7 shoots) and in cultures containing no gelling agent (6.9 mm and 0.7 shoots). There were no differences in shoot growth and number in cultures containing 0.7% Sigma agar (2.2 mm and 16.5 shoots), 0.525% agar + 0.05% gelrite (2.6 mm and 18.7 shoots), 0.35% agar + 0.1% gelrite (1.6 mm and 17.4 shoots), and 0.175% agar + 0.15% gelrite (2.0 mm and 20.4 shoots). The most vitrification occurred in cultures on medium with the lowest amount of agar, gelrite only, and liquid medium.

ABSTRACT: A plant regeneration protocol via adventitious shoot organogenesis from internode explants of Paederia foetida (Skunk vine) is reported here for the first time. Three explants (leaf, mature internode and internode derived from axenic shoot cultures) were tested for shoot organogenesis. Leaf explants failed to induce adventitious shoots whereas axenic internode explant was found to be superior to mature internode explants for the induction of adventitious shoots. Axenic internode explants cultured on MS medium supplemented with 3.0 mg/l BAP showed maximum (86.7 %; 10.4 shoots per explant) adventitious shoot organogenesis. The regenerated shoots were best rooted (90 %; 14 roots per shoot) on half-strength MS medium. Eighty percent of the rooted shoots were successfully acclimatized in soil: sand (1:1) mixture. All these acclimatized plants were successfully transferred to larger pots containing garden soil and subsequently established in the field.

Lychnis cognate Maxim and Lychnis fulgens Fish. Ex Spreng are two valued ornamental plants in Korea. Soaking of seeds in GA3 solution remarkably promoted germination up to 60%, but the control (0 mg/l) was not effective (> 5%). To select an adequate temperature for seed germination, seeds, previously soaked in a 1000 mg/l GA3 for 24 hrs, were incubated at 15, 20, 25, and 30°C. Seed germination of over 20% was obtained at 15, 20, and 25°C, but only 10% at 30°C. These results indicate that the seeds of L. cognate and L. fulgens are in a such dormant state that they hardly germinate even by dormancy breaker (GA3) and low (15 ? 25°C) temperature treatment. The highest callus induction was observed in the leaf explants of the seedlings on MS containing specific concentrations of 3.0 mg/l BA and 1.0 mg/l NAA. The adventitious shoot was formed < 90% of calli on 1/2 WPM medium. The height of in vitro propagated plantlet was no different media used for regeneration. This in vitro propagation protocol should be useful for conservation of endangered and ornamental plant.Plant Tissue Cult. & Biotech. 25(1): 1-12, 2015 (June)
