Indica rice variety Ciherang is the most planted variety in Indonesia, but the micropropagation technique is restricted because it is known has low regeneration frequency and included as recalcitrant cultivar for tissue culture and tranformation activities. One of solution to resolve that problem is developing a technique of somatic embryogenesis in callus of ciherang rice cultivar. The aims of study were to determine medium composition for inducting embryogenic callus in Ciherang rice and to know the effectivity of rice regeneration using callus as explant. The methods were included induction of embryogenic callus in callus induction media (CIM) containing MS basal, Proline 600 mg/l, Casein Hidrolisat 300 mg/l, phytagel 2,5 g/l, BAP 0.25 mg/l, sukrosa 30 %, and 2,4-D in different concentration, from 2,4-D 0 mg/l as control (CIM 1), 2,4-D 2 mg/l (CIM 2), 2,4-D 3 mg/l (CIM 3), and 2,4-D 4 mg/l (CIM 4). It then be continued to regenerate the calli in RM 1 medium containing MS basal, NAA 0,2 mg/l, Kinetin 2 mg/l, Agarose 10 g/l, and sukrosa 30 %, pH 5,8 for six days in dark and RM2 medium containing MS basal , NAA 0,2 mg/l, Kinetin 2 mg/l, Agarose 8 g/l, sukrosa 30 %, pH 5,8 in light room. Results showed CIM 4 medium using 2,4-D 4 mg/l gave optimum result in calli induction with procentage 57,63% and CIM 3 using 2,4-D 3 mg/l gave optimum result in embryonic calli induction with procentage 53,63%. Result of embryonic calli gave regeneration frequency procentage is 9,6%. The total planlet obtained after five weeks old in regeneration medum were 11 planlets ready for aclimatization. The application of biotechnology in upland rice improvement programs depends on the availability of efficient regeneration protocols.  Although protocols for shoot regeneration of upland rice are available, none has been reported for pigmented cultivars.  This study reports on a protocol for callus induction and regeneration of Tadong, a pigmented upland rice cultivar from Sabah.  For callus induction, immature embryos were cultured on media containing 2,4-Dichlorophenoxyacetic (2,4-D) at various concentrations (0 – 2.5 mg/L) and on different types of media (MS; MSB5; N6B5; N6).  To induce shoot regeneration, callus explants were cultured on MS medium supplemented with combinations of 6-Benzylaminopurine (BAP) at various concentrations (0 – 3.0 mg/L) and 1-Naphthaleneacetic acid (NAA) at 1.0 mg/L.  To induce shoot development, callus explants were pre-treated with Thidiazuron (TDZ) at various concentrations (0-1.0 mg/l) and exposed to different desiccation periods (0 – 72 hours).  2,4-Dichlorophenoxyacetic at 2.5 mg/L and N6B5 medium resulted in the highest percentages of explant forming callus which were 60.3 ± 17.0 % and 58.7 ± 9.8 % respectively.   The regeneration media failed to induce shoot on callus explants, instead, green spots were formed on the surface of the callus.  The green spots were stimulated to develop into shoots when the callus explants were pre-treated with 0.5 mg/L TDZ or exposed to partial desiccation for 24 h, the percentages of explant forming shoot were 35.7 ± 4.8 % and 47.7 ± 6.8 % respectively.   Shoots developed into complete plants on hormone-free MS medium and acclimatized.

Thirty-nine Thai rice cultivars, including 17 non-colored rice cultivars and 22 colored rice cultivars (black rice cultivars and red rice cultivars), were investigated for γ-oryzanol content. The γ-oryzanol content of non-colored rice cultivars and colored rice cultivars was found to be 226.40 ± 17.50 μg/g to 411.80 ± 19.80 μg/g and 295.80 ± 15.40 μg/g to 459.80 ± 6.60 μg/g, respectively. Black rice cultivars seem to have higher γ-oryzanol content than do red rice and non-colored rice cultivars. The highest γ-oryzanol levels were identified in Khao Jao Dam Sa-Nit, a black rice cultivar, which contained 1.55 and 2.03 times more γ-oryzanol than Khao Man Bpoo, a red rice cultivar with lowest γ-oryzanol levels, and Khao Gor Kor 57, a non-colored rice cultivar with lowest γ-oryzanol levels, respectively. For further investigation, the effects of gamma irradiation on the γ-oryzanol content of four germinated Thai market rice cultivars, Khao Pathum Thani 1, Khao Gor Kor 31, Khao Gor Kor 41, and Khao Gor Kor 57, was investigated. The highest increase in γ-oryzanol content among the gamma irradiated rice cultivars was identified in Khao Pathum Thani 1 and Khao Gor Kor 31, which had γ-oryzanol levels approximately 1.27 times higher than that of the control rice (non-gamma irradiated rice). Our report contains basic knowledge about creating new rice lines that are rich in highly beneficial nutrients.
