AbstractDuring the double fertilization process, pollen tubes deliver two sperm cells to an ovule containing the female gametes. In the pollen tube, the vegetative nucleus and sperm cells move together to the apical region where the vegetative nucleus is thought to play a crucial role in controlling the direction and growth of the pollen tube. Here, we report the generation of pollen tubes in Arabidopsis thaliana whose vegetative nucleus and sperm cells are isolated and sealed by callose plugs in the basal region due to apical transport defects induced by mutations in the WPP domain-interacting tail-anchored proteins (WITs) and sperm cell-specific expression of a dominant mutant of the CALLOSE SYNTHASE 3 protein. Through pollen-tube guidance assays, we show that the physiologically anuclear mutant pollen tubes maintain the ability to grow and enter ovules. Our findings provide insight into the sperm cell delivery mechanism and illustrate the independence of the tip-localized vegetative nucleus from directional growth control of the pollen tube.

This article presents the results of the development of a methodology for describing the main morphological and cultural traits of the Pyricularia oryzae Cav. strains widespread in the south of Russia. At the same time, the types of traits are identified and listed, which make it possible to unambiguously determine the uniqueness and variety of the pathogen. The relationships and patterns established using cluster and statistical analysis make it possible to identify the conditions for the development of a pathogen that determine its predominant forms. Thus, research shows that leaf forms of P. oryzae strains isolated from rice plants with leaf form of blast disease have an equally directional growth pattern of a colony with a felt structure, and strains isolated from neck-affected plant form often produce a zone of a colony with a clumpy structure. The classification of cultural traits will make it possible to obtain scientifically grounded and comparable data that can be used in the analysis of the interaction of P. oryzae strains with rice plants on various varieties and in various agro-technological conditions in order to improve and rationalize agricultural activities. The study opens up the possibility of using data in breeding, making it possible to identify forms of a pathogen that infect certain varieties.

ABSTRACT Directional growth is a function of polarized cells such as neurites, pollen tubes, and fungal hyphae. Correct orientation of the extending cell tip depends on signaling pathways and effectors that mediate asymmetric responses to specific environmental cues. In the hyphal form of the eukaryotic fungal pathogen Candida albicans, these responses include thigmotropism and galvanotropism (hyphal turning in response to changes in substrate topography and imposed electrical fields, respectively) and penetration into semisolid substrates. During vegetative growth in C. albicans, as in the model yeast Saccharomyces cerevisiae, the Ras-like GTPase Rsr1 mediates internal cellular cues to position new buds in a prespecified pattern on the mother cell cortex. Here, we demonstrate that Rsr1 is also important for hyphal tip orientation in response to the external environmental cues that induce thigmotropic and galvanotropic growth. In addition, Rsr1 is involved in hyphal interactions with epithelial cells in vitro and its deletion diminishes the hyphal invasion of kidney tissue during systemic infection. Thus, Rsr1, an internal polarity landmark in yeast, is also involved in polarized growth responses to asymmetric environmental signals, a paradigm that is different from that described for the homologous protein in S. cerevisiae. Rsr1 may thereby contribute to the pathogenesis of C. albicans infections by influencing hyphal tip responses triggered by interaction with host tissues.
