Abstract Acetamiprid, that is known as the commercial formulation Mospilan® 20SP is the part of the neonicotinoid insecticide group and is widely used against various pests. In our study we assessed the potential clastogenic effects of Mospilan® in human peripheral blood lymphocytes in vitro using a chromosome aberration test. The lymphocytes were treated with acetamiprid in the concentration range of 5, 10, 25 and 50 µg.ml−1 for 24 and 48 h. After 24 h exposure, the insecticide induced statistically significant higher levels of chromosome aberrations from the concentration of 10 µg.ml−1 (P < 0.05 and P < 0.001) and a significant decrease in mitotic index (MI) at the concentrations of 25 and 50 µg.ml−1 (P < 0.05 and P < 0.01), respectively. After a 48 h exposure, we found a dose dependent increase in the percentage of chromosome aberrations at all concentrations (P < 0.05; P < 0.01 and P < 0.001) and a decrease in MI at concentrations of 25 and 50 µg.ml−1 (P < 0.05 and P < 0.01). Our results indicated that neonicotinoid insecticide formulations containing acetamiprid may have potential cytotoxic and genotoxic effects.

Aim. Research objective is to establish the possibility of modifying the astaxanthin (a carotenoid from a xanthophyll group) radiation-induced cytogenetic effects in human peripheral blood lymphocytes (PBLs) in vitro. Methods. The cultivation of PBLs from four conventionally healthy volunteers, the preparation and analysis of uniformly stained slides of metaphase chromosomes. Astaxanthin in final concentrations of 2, 10 and 20 µg/ml was added into the culture of PBL prior to the incubation before irradiation with γ-quanta in a dose of 1 Gy. Results. Astaxanthin did not affect the level and spectrum of chromosome damage in non-irradiated PBLs both in individual persons, and along the group on average (P>0.05), indicating a lack of mutagenic activity. The effect of astaxanthin at a concentration of 20 μg/ml on irradiated PBLs resulted in a significant reduction of radioinduced cytogenetic effect in all donors. Medium-group level of chromosome aberrations decreased almost 3 times and was characterized by statistically significant (P<0.001) decrease in frequency of chromosomal type aberrations due to the classical unstable cytogenetic markers of radiation effect, dicentrics and ring chromosomes. Conclusions. Astaxanthin at a concentration of 20 µg/ml was found to reduce the mutagenic effect of ionizing radiation, thus suggesting its powerful radioprotective potential. Trimethyltin chloride was found to induce cytotoxic damage in vitro in human peripheral blood lymphocytes. Two concentrations (0.5 μg and 1.0 μg) were added to lymphocytes from male and female subjects in mitogen stimulated and serum supplemented culture medium for 72 h. A dose-related increase of inhibition of replication index (P < 0.01) and cell division (P < 0.001) was observed. The frequencies of abnormal cells and chromosomal aberrations such as chromatid and chromosome breaks, dicentrics, triradial and quadriradial configurations were increased significantly (P < 0.001), as were micronucleus counts (P < 0.001) and sister chromatid exchanges (P < 0.001). Endoreduplication, an extremely rare spontaneous event in human lymphocytes, was observed in a few cases at all dose levels. The effects were more pronounced in lymphocytes obtained from habitual smokers.
