Lurking in the water: testing eDNA metabarcoding as a tool for ecosystem-wide parasite detection

Raw sewage disposal in marine waters is a common practice in many countries. This practice raises health risk concerns of possible transmission of Giardia and Cryptosporidium. Both of these protozoa have been shown to be transmitted by recreational swimming. To date no studies have determined the efficiency of their detection and concentration in marine waters. This study evaluated the efficiency of their detection in tap water and from marine waters in Hawaii with two different filter types. This study compared a polypropylene fiber cartridge filter, DPPPY (1.0 μm nominal porosity) (Cuno, Meriden, CT) which is typically used for parasite detection and the Filterite negatively charged filter (0.45μm) (Filtemp Sales, Inc., Phoenix, AZ). The latter would allow for both viruses and parasites to be concentrated simultaneously. The organisms were removed from the filter by passing the eluent through the filters in the opposite direction of collection and detected by indirect immunofluorescence antibody staining specific for Giardia and Cryptosporidium. Processing was simpler and faster with the Filterite filter and the overall efficiency for both Giardia and Cryptosporidium detection was greater. These methods are currently being used for the detection of the oocysts and cysts at bathing beaches in Hawaii impacted by marine sewage discharge.

ABSTRACT The environment is the likely source of most nontuberculous mycobacteria (NTM) involved in human infections, especially pulmonary, skin, and soft tissue infections. In order to measure the prevalence of NTM in different aquatic ecosystems, we tried to standardize the culture methods used for surface water testing since many procedures have been described previously. Cultivation of mycobacteria requires long-term incubation in rich media and inactivation of rapidly growing microorganisms whose growth impedes observation of mycobacterial colonies. Consequently, the two criteria used for evaluation of the methods examined were (i) the rate of inhibition of nontarget microorganisms and (ii) the efficiency of recovery of mycobacteria. We compared the competitive growth of Mycobacterium chelonae and M. avium with nontarget microorganisms on rich Middlebrook 7H11-mycobactin medium after treatment by several chemical decontamination methods that included acids, bases, detergent, or cetylpyridinium chloride (CPC) with and without an antibiotic cocktail, either PANTA (40 U/ml polymyxin, 4 μg/ml amphotericin B, 16 μg/ml nalidixic acid, 4 μg/ml trimethoprim, and 4 μg/ml azlocillin) or PANTAV (PANTA plus 10 μg/ml vancomycin). Our results showed that treatment for 30 min with CPC (final concentration, 0.05%) of water concentrated by centrifugation, followed by culture on a rich medium supplemented with PANTA, significantly decreased the growth of nontarget microorganisms (the concentrations were 6.2 � 0.4 log10 CFU/liter on Middlebrook 7H11j medium and 4.2 � 0.2 log10 CFU/liter on Middlebrook 7H11j medium containing PANTA [P < 0.001]), while the effect of this procedure on NTM was not as great (the concentrations of M. chelonae on the two media were 7.0 � 0.0 log10 CFU/liter and 6.9 � 0.0 log10 CFU/liter, respectively, and the concentrations of M. avium were 9.1 � 0.0 log10 CFU/liter and 8.9 � 0.0 log10 CFU/liter, respectively). We propose that this standardized culture procedure could be used for detection of NTM in aquatic samples.

Though recent May 2020 standoff that continued between India and China in Galwan valley was not taken by the Indian leadership as routine water testing by the Chinese. India took it seriously and very well managed to handle the aggression on its own and halted the China’s backdoor thinking of projecting themselves a superpower at least in Asia. Now it’s going to be a normal routine along the LAC as India is now determined to change the ground strategic infrastructure reality along the LAC. By tackling Chinese on its own, India has bolstered its position in the world’s strategic power pie chart. Further India need to work on many fronts to meet the challenges and convert these sour relations in a Diplomatic, Economic and Strategic opportunities for India. The aim of this paper is to analyze in detail the India’s stand against the PLA's incursion, internal assessment to meet this “going to be routine” challenge on LAC and further considering the India’s population, market size and china’s sour relations with neighbors, it’s time for India to encash the opportunities out of such attempts from China.

Background: Feline otitis externa is a dermatological disorder with a multifactorial complex etiology.Aim: This study aimed to investigate the prevalence of different etiological agents, particularly the parasitic and bacterial, responsible for the cases of feline otitis externa in Tripoli, Libya, and to assess the antimicrobial susceptibility of the bacterial isolates from those cases.Methods: Cerumen and otic discharges of the suspected cats were collected for parasite detection and bacterial culture. Kirby–Bauer’s disk diffusion method was used for antimicrobial susceptibility testing.Results: The results showed that otodectic mites and bacterial causes were equally the most prevalent in those cases, with a prevalence of 47.1% each. Otodectes cynotis infestation was more frequently bilateral and severe. Staphylococcus spp. were the most prevalent among bacterial causes (75%), followed by Proteus spp. (16.6%) and Pseudomonas spp. (8.4%). Norfloxacin and gentamicin were the most effective antimicrobials against bacterial isolates, as they were effective against 83.3% and 70.8% of isolates, respectively.Conclusion: Otodectes cynotis infestation and staphylococcal infections constituted the most common etiology of feline otitis externa in Tripoli, Libya, and norfloxacin represented a cogent antibacterial for the treatment of otitis externa.
