Histology of organogenesis and somatic embryogenesis in excised root cultures of an endangered species Tylophora indica (Asclepiadaceae)

This paper reports an efficient regeneration protocol through parallel organogenic and embryogenic pathways from green root segments (GRSs) of Tylophora indica (Burm.f) Merrill. GRSs explants from one year old in vitro cultures were cultured on Murashige and Skoog (MS) medium containing various cytokinins. Five µmol/L of 6-benzyladenine (BA) was most responsive for organogenesis in 1.5 cm long GRSs. Repeated subculture on medium containing both BA (5 µmol/L) and 1-naphthleneacetic acid (NAA) (0.1 µmol/L) promoted multiplication and proliferation of direct shoot buds (46.80 ± 0.96) and callus mediated somatic embryogenesis (18.07 ± 0.33). Germinated embryos isolated from callus were transferred onto maturation medium consisting of half-strength MS medium either devoid of plant growth regulators (PGRs) or with various concentrations of gibberellic acid (GA). Microshoots were excised during subculture and transferred onto root induction medium, thus ensuring a continuous supply of germplasm. Morphogenic variations were noticed in types of roots induced on various auxins. Regenerated plantlets and emblings hardened best on vermiculite with a survival rate of 90% and 70% respectively. However, the emblings were healthier in comparison to the regenerated plants. Histological analysis showed the origin and development of organogenesis.

Clonal multiplication of Aegle marmelos was achieved through in vitro culture. Cotyledonary nodes from one month old in-vitro grown seedlings of Aegle marmelos were cultured on MS medium supplemented with BAP, Kn, and IBA either alone or in combination. The highest regenerative response was observed on medium containing 2.5 mg/L BAP and 0.5 mg/L Kn from node and shoot tip within 8-10 days. When regenerated shoots were subcultured on MS medium supplemented with 0.2 mg/L BAP and 0.02 mg/L Kn, maximum number of multiple shoots were observed after third phase of subculture. Poor response was found using MS medium supplemented with Kn only. In vitro induced shoots were transferred into root induction medium consisting of half-strength MS supplemented with auxins, IAA, IBA or NAA. Rooting was best in medium supplemented with 1.0 mg/L IBA. Rooted plantlets were acclimatized and transferred to the soil with 96% survival rate. Nodal segments from naturally grown Eclipta alba (L.) Hassk.were used as explants for organogenesis. Multiple shoots were obtained from the explants cultured on MS medium supplemented with various concentrations of BAP and Kn alone or in combination with NAA and IAA. Maximum number of multiple shoots (18.40±0.67) were induced on MS medium supplemented with 1.0 mg/l BAP and 0.1mg/NAA. In vitro raised shoots were cultured onto half and full strength MS medium supplemented with different concentration of IBA, IAA and NAA. The best root induction medium was found to be half strength MS containing 0.1 mg/l IBA where 96% shoots rooted. Somatic embryos were obtained from cotyledon and midrib explants of Solanum melongena L., cultivar Loda. For callus induction, medium was supplemented with different concentrations of auxin singly or in combination with BAP. The best callusing 83-85% was obtained from both of the explants cultured on MS medium containing 2.0 mgl-1NAA + 0.05 mgl-1BAP. Somatic embryogenesis and shoot regeneration was achieved after transferring the calli to MS medium supplemented with BAP, GA3, NAA and Zeatin. Cotyledon derived calli showed better performance (87%) for regeneration than that of midrib (82%) when sub cultured on MS medium having 2.0 mgl-1 Zeatin + 1.0 mgl-1 BAP. For root induction, MS + 3.0 mgl-1 IBA was proved to be better treatment for average number (14-15) and mean length (12 cm) of roots than those of other treatments. Anther of five varieties of Brassica species, namely BARI Shariaha-7, Tori-7, Agrani, Daulat and Safal were cultured in vitro to observe their regeneration potentiality. Different concentrations and combinations of growth regulators were supplemented in MS medium. The range of callus induction was 12.50-87.50 %. Maximum callus induction (75.00%) was observed on MS +4 mg/L 2, 4-D + 1.0 mg/L BAP. Among the genotypes, BARI Sharisha-7 showed the highest percentage of callus induction (60.42%). Among the treatments, highest percentage of shoot regeneration (75.00%) was observed on MS + 4 mg/L BAP + 1.0 mg/L NAA. BARI Sharisha-7 also showed the highest rate of plant regeneration (66.67%). Root induction was highest (75%) on half strength MS medium supplemented with 1.0 mg/L IBA and 0.5 mg/L NAA. The plantlets with sufficient roots thus obtained were transferred successfully to plastic pots and subsequently to the field. BARI Sharisha-7 and Tori-7 survived easily in the pots as well as in the field but Safal was very poor in survivability both in the pots and in the field. In vitro-proliferated shoots of `Delicious' and `Starkspur Red' apples (Malus domestica Borkh.) were submitted for 1 week to root induction in the dark with their basal parts in liquid medium containing 1.5 μm IBA and 43.4 mm sucrose. The shoots were transferred to root initiation and root elongation medium made up of plugs wetted with half-strength Lepoivre salts. When antioxidants, such as PVP, GH, 2-Me, and DIECA, were added to the liquid induction medium, the percentages of rooting usually decreased. In contrast, if the antioxidants PVP and citric acid were added in the initiation-elongation medium, the percentages of rooting increased. The results agree with the finding of other researchers who proposed a rapid increase in peroxidase activity during root induction followed by a fast decline during rooting initiation as a marker for good rooting responses. Moreover, the use of antioxidants both in induction and in initiation-elongation had no effect on number and length of roots. Chemical names used: polyvinylpyrrolidone-40 (PVP); glutathione reduced (GH); 2-mercaptoethanol (2-Me); diethyldithiocarbamic acid, sodium salt trihydrate (DIECA); N-(phenylmethyl) 1 H-purin-6-amine (BA); 1 H-indole-3-butyric acid (IBA); gibberellic acid (GA3).

 Technique of tissue culture for Chia (Salvia hispanica) micropropagation was achieved, this study investigated the impact of various concentrations of plant growth regulators on shoot multiplication and root induction with the Chia’s mature seed as a source explant. The highest percentage of shoot formation (80%), shoots number per explant(3.20) and shoot length(3.26 cm), were recorded on MS medium enriched with BAP(1.0 mgl-1) after eight weeks of seed culture. The optimal medium for the rhizogenesis was achieved on half strength MS medium fortified with 1.0 mgl-1 IBA after four weeks of culture, which had the highest rooting percentage (100%) with highest mean of roots number (5.6 roots per shoot) with (3.40 cm root length). The rooted plants were successfully adapted ex vitro with a survival rate of 85%.

The study involved two auxins viz., 2,4-D (2,4-Diclorophenoxyacetic acid) and picloram at three different concentrations (1,2, 3 mg/L) in full strength MS media to study their comparative influence on induction of somatic embryogenesis from immature zygotic embryos of two gynodioecious varieties of papaya ‘CO.7’ and ‘Red Lady’. In papaya cultivar ‘CO.7’, 2,4-D at 2 mg/L gave the highest callus induction frequency of 90.93%, whereas comparatively higher concentration of 3 mg/L 2,4-D was found suitable for ‘Red Lady’ (87.26%). Although there was profuse callus formation, 2 mg/L 2,4-D recorded comparatively higher frequency of embryogenic calli in ‘Red Lady’ (51.67%) when compared to ‘CO.7’ (30.00%). Somatic embryo maturation was achieved upon transfer of embryogenic calli exhibiting globular stage embryos on to maturation medium (MS medium + ABA (Abscisic acid) and BAP (Benzyl amino purine) in different concentrations + glutamine 400 mg/L). In the maturation medium, the combination of 1.5 mg/L ABA and 0.4 mg/L BAP registered better conversion of the globular embryo to cotyledonary embryos than other levels. The frequency of somatic embryo germination was higher in ‘Red Lady’ (50.00%) as compared to ‘CO.7’ (31.67%) on half-strength MS medium devoid of growth regulators.

Current Status of Saussurea heteromalla was investigated in Galiyat areas of District Abbottabad, viz., Jahaffar, Seri, Beeran Gali, Banj, Haryala, Daryala Gali, Sarbhanna, Barriyan, Akhreela and Broangiala. Saussurea heteromalla was found common in Seri, Sarbhanna and Barriyan; endangered in Haryala, Jahaffar, Banj, Daryala Gali and Beeran Gali and absent in Akhreela and Broangiala. In vitro regeneration of Saussurea heteromalla (D. Don.) Hand-Mazz on MS media was conducted and shoots were developed on full strength MS medium supplemented with 1 mgL-1 GA3. The developed shoots were transferred for root induction to half strength MS medium fortified with various concentrations of Indole butyric acid (IBA) and α-Naphthalene acetic acid (NAA) i.e. T1 (control), T2 (2 mgL-1 IBA), T3 (3 mgL-1 IBA), T4 (4 mgL-1 IBA), T5 (1 mgL-1 NAA), T6 (2 mgL-1 NAA) and T7 (3 mgL-1 NAA). Maximum mean shoot length (6.3 cm), mean number of leaves (7), mean number of nodes (5.25); highest root emergence percentage (71%), means root length (1.5 cm), mean number of roots (3) and highest survival rate (100%) was recorded in treatment T4. However, treatments T5 and T6 also seem to be effective for in vitro rooting of Saussurea heteromalla. Treatments T2, T3 and T7 showed minimum root growth. It was concluded that IBA at higher concentration is more effective for in vitro rooting and better shoot growth of Saussurea heteromalla whereas NAA also initiate rooting but at lower concentration.

The optimum range of temperature for germination (96-100%) of Calamintha cretica, an herb with potential pharmaceutical and horticultural uses, was 15 to 20 °C, with 10 and 30 °C cardinal temperatures. Storage up to one year did not affect germination. The effect of zeatin (ZEA), 6-benzyladenine (BA), kinetin, and 6-γ-γ-(dimethylallylamino)-purine added in MS medium at concentrations from 0.0 to 8.0 mg L-1 was tested for shoot proliferation of both adult- and seedling-origin nodal explants at first- and sub-culture. Both explant types responded similarly during in vitro culture. At cytokinin concentrations up to 1 mg L-1 explant response was high (over 85%) but shoot number per explant was low (1.2-2.2). Increasing cytokinin from 2.0 to 8.0 mg L-1 resulted to an analogous decrease of explant response and shoot length, and an increase of shoot number, particularly when ZEA or BA was used (5.0-6.6 shoots per explant, 0.5-1.0 cm long) with simultaneous though increase of hyperhydricity (up to 50%). The addition of 0.1 mg L-1 naphthaleneacetic acid into the 8.0 mg L-1 BA medium almost eliminated hyperhydricity and increased explant response, while the increase of agar concentration from 8.0 to 12.0 g L-1 eliminated hyperhidricity and induced the highest shoot proliferation (93-95% explant response, 11.2-12.3 shoots per explant, 0.8-1.0 cm long). Microshoots and microshoot clusters rooted (88-96%) on half-strength MS medium either hormone free or supplemented with 1 to 4 mg L-1 indole-3-butyric acid. Plantlets survived at 80% to 100% after ex vitro acclimatization in peat: perlite 1:1 (v/v).

The present study describes a protocol for high frequency plant regeneration of Physalis minima. Shoots were induced by culturing nodal segments and shoot tips from 15 day old seedlings. About 29 and 32 shoots were found to be induced from nodal segment and shoot tip explants, respectively, cultured on MS medium supplemented with 1.0 mg/l BAP. When shoots were subcultured on the fresh medium with same component as mentioned above, the shoots were elongated. Shoots rooted well when they were excised individually and implanted on half-strength MS medium with 0.3 mg/l NAA, where 98% shoots rooted within 12-15 days. In vitro grown plantlets with strong root system were successfully established in normal room temperature for seven days before transplanting in pots where they were reared for three weeks through successive acclimatization. The regenerated plants were successfully transferred to the soil with 90% survival rate.
