In India 14 seagrass species can be found with monospecific genera (Enhalus, ThalassiaandSyringodium),Cymodoceawith two species andHalophilaandHalodulerepresented by more than two taxonomically complex species. Considering this, the present study was made to understand the level and pattern of genetic variability among these species collected from Tamilnadu coast, India. Random amplified polymorphic DNA (RAPD) analysis was used to evaluate the level of polymorphism existing between the species. Out of the 12 primers tested, 10 primers amplified 415 DNA fragments with an average of 41.5 fragments per primer. Of the total 415 amplified fragments only 123 (29.7%) were monomorphic and the remaining 292 (70.3%) were polymorphic for Indian seagrass species. Among the 10 primers used four are identified as the key primers capable of distinguishing all the Indian seagrasses with a high degree of polymorphism and bringing representative polymorphic alleles in all the tested seagrasses. From the present investigation, this study shows that the RAPD marker technique can be used not only as a tool to analyse genetic diversity but also to resolve the taxonomic uncertainties existing in the Indian seagrasses. The efficiency of these primers in bringing out the genetic polymorphism or homogeneity among different populations of theHalophilaandHalodulecomplex still has to be tested before recommending these primers as an identification tool for Indian seagrasses.

In this study, sulfated polysaccharide-rich extracts were isolated from 22 tropical seaweeds (4 red, 11 brown, and 7 green) found in northeastern Brazil, and evaluated for the role of anticoagulant agents. Fifteen of the extracts showed anticoagulant activity, including all the extracts from green seaweeds. Udotea flabellum (a green seaweed) extract was the most potent, requiring an amount of only 3 µg to double the plasma coagulation time in the activated partial thromboplastin time test. A similar result was obtained with 1 µg of heparin. Two sulfated homogalactans with anticoagulant activity, F-I (130 kDa) and F-II (75 kDa), were isolated from this extract using several bio-guided purification steps. Their anticoagulant activity, as well as properties related to antitumor activity (anti-proliferative, anti-adhesive, and anti-migratory), were accessed. Their anticoagulant activities were close to that of heparin. We found that F-I and F-II (0.5–10 μg/mL) were not able to directly inhibit thrombin. In the presence of anti-thrombin, F-I (0.5 μg/mL) was more effective than heparin (0.5 μg/mL) in inhibiting thrombin, while F-II showed similar effects as heparin. F-I and F-II also inhibited B16-F10 (murine melanoma cells) adhesion, migration, and proliferation on a fibronectin-coated surface, but not on laminin- or collagen I-coated surfaces. Except for the antiproliferative activity, the other effects of F-I and F-II were eliminated upon their desulfation (~50%), indicating that the degree of sulfation is not as important for F-I and F-II anti-proliferative activity as the sulfation position. Taken together, the results provide strong evidence for the potential utility of sulfated galactans from U. flabellum, making these compounds an interesting option for future investigations that aim to design new anticoagulant/antitumor agents.

The bryozoan Electra posidoniae Gautier is found solely on the leaves of the Neptune grass Posidonia oceanica (L.) Delile, dominating the leaf epifauna of this seagrass. Epiphytes of marine angiosperms (or seagrasses) often play an important role in ecosystem functioning, for example as food web suppliers. As dysfunction of the epiphytic component is often implied in human-induced seagrass decline, it is important to understand the dynamics and life traits of this community in pristine areas. This study involved the monthly assessment of colonization dynamics, biomass seasonality, and diet composition through measurements of stable isotopes, in E. posidoniae at a depth of 10 m in the Revellata Bay (Corsica, Mediterranean Sea). Ancestrulae (i.e. colony founders) appeared towards the end of winter and were very selective in their settlement position along the leaves of P. oceanica. A maximum of 100,000 colonies per square meter was recorded. Colonies of E. posidoniae dominated the epiphytic community biomass in early spring, but were overtaken by epiphytic algae in June. Food shortage could be involved in this reduction in dominance. Although stable isotope ratios of C, N and S showed that this suspension feeder mainly relies on the water column for its food, other food sources such as re-suspended epiphytic diatoms could be important in late spring (i.e. after the phytoplanktonic bloom). Additionally, a contribution of seagrass phytodetritus to the diet of this species cannot be excluded. The species was almost absent in winter, raising the question of its recruitment in spring. This study confirms the quantitative importance of this species in the seagrass meadow and explores its role in the relationship between the water column and this seagrass ecosystem.
