Occurrence of Viroids in Commercial Hop (Humulus lupulus L.) Production Areas of Washington State

Viroid status was investigated in commercial hop (Humulus lupulus) plantings in three regions of intensive hop production in Washington State. Hop stunt viroid (HSVd), the causal agent of hop stunt disease, was detected for the first time in hop plants in North America. HSVd was detected in samples from 10 of the 33 hop gardens sampled and in 19 of 126 plant samples. Infection with HSVd was associated with chlorosis and reduced plant vigor. Reverse transcription-polymerase chain reaction amplification, sequence identity, and pathogenicity of hop extracts on cucumber seedlings confirmed the presence of HSVd. Apple fruit crinkle viroid (AFCVd), recently reported in hops in Japan, was not detected by RT-PCR in any samples. Hop latent viroid (HLVd), which is frequently associated with hop germplasm world-wide, was detected with high frequency (98 of 126 hop plants surveyed). Accepted for publication 23 September 2007. Objective.Interferon (IFN) regulatory factors (IRF) are transcriptional mediators of IFN-induced signaling pathways and are involved in immune response. We have analyzed for the first time the association of 2 IRF5 gene variants in the susceptibility to giant cell arteritis (GCA).Methods.Two hundred twenty patients with biopsy-proven GCA and 520 matched controls were assessed. DNA from patients and controls was obtained from peripheral blood. Samples were genotyped for the IRF5 rs2004640 and for the IRF5 CGGGG insertion/deletion polymorphism using a predesigned TaqMan allele discrimination assay and by polymerase chain reaction amplification, followed by an ABI3100 sequencer, respectively.Results.A genotyping rate of 96% was achieved in this series of GCA patients. No significant differences were found in the genotype distribution between GCA patients and controls for both IRF5 gene variants. In this regard, similar genotype frequencies were found in GCA patients and controls. No significant differences were observed when GCA patients were stratified according to the presence of specific clinical features of the disease such as polymyalgia rheumatica or severe ischemic complications.Conclusion.Our results showed no association of IRF5 rs2004640 and CGGGG insertion/deletion polymorphisms in the susceptibility to and clinical expression of GCA.

Abstract The structure of chromosomal inversions mediated by hobo transposable elements in the Uc-1 X chromosome was investigated using cytogenetic and molecular methods. Uc-1 contains a phenotypically silent hobo element inserted in an intron of the Notch locus. Cytological screening identified six independent Notch mutations resulting from chromosomal inversions with one breakpoint at cytological position 3C7, the location of Notch. In situ hybridization to salivary gland polytene chromosomes determined that both ends of each inversion contained hobo and Notch sequences. Southern blot analyses showed that both breakpoints in each inversion had hobo-Notch junction fragments indistinguishable in structure from those present in the Uc-1 X chromosome prior to the rearrangements. Polymerase chain reaction amplification of the 12 hobo-Notch junction fragments in the six inversions, followed by DNA sequence analysis, determined that each was identical to one of the two hobo-Notch junctions present in Uc-1. These results are consistent with a model in which hobo-mediated inversions result from homologous pairing and recombination between a pair of hobo elements in reverse orientation.
