Sphingobacterium psychroaquaticum sp. nov., a psychrophilic bacterium isolated from Lake Michigan water

A psychrophilic, Gram-negative bacterium, designated MOL-1T, was isolated from water of Lake Michigan. 16S rRNA gene sequence analysis revealed that the sequence of strain MOL-1T has sequence similarity of 95.6, 94.8, 94.3, 94.3, 94.2 and 93.9 %, respectively, to the 16S rRNA gene sequences of Sphingobacterium shayense HS39T, S. lactis WCC 4512T, S. composti T5-12T, S. daejeonense TR6-04T, S. bambusae IBFC2009T and S. alimentarium WCC 4521T. The major cellular fatty acids were iso-C15 : 0, iso-C17 : 0 3-OH and summed feature 3 (iso-C15 : 0 2-OH and/or C16 : 1ω7c). Menaquinone MK-7 is the predominant respiratory quinone, while sym-homospermidine is the predominant polyamine. The polar lipid profile is composed of the predominant lipids phosphatidylethanolamine and unidentified polar lipid L2, with moderate amounts of unidentified polar lipids L1, L5 and L6 and unidentified aminophospholipids APL1 and APL2 and minor to trace amounts of unidentified polar lipids L3, L4, L7, L8, L9 and L10, unidentified phospholipid PL4 and unidentified aminophospholipid APL3. After molecular and phenotypic studies, including chemotaxonomic analyses, it was concluded that strain MOL-1T represents a novel Sphingobacterium species, for which the name Sphingobacterium psychroaquaticum sp. nov. is proposed. The type strain is MOL-1T ( = NRRL B-59232T  = DSM 22418T).

A novel Gram-stain-positive actinobacterium, designated strain SCSIO 11529T, was isolated from tissues of the stony coral Galaxea fascicularis, and characterized by using a polyphasic approach. The temperature range for growth was 22–50 °C (optimum 28–45 °C), the pH range for growth was 6.0–8.0 (optimum pH 7.0), and the NaCl concentration range for growth was 0–7 % (w/v) NaCl. The polar lipid profile contained diphosphatidylglycerol, phosphatidylcholine, phosphatidylglycerol, phosphatidylmethylethanolamine, phosphatidylethanolamine and an unknown polar lipid. The predominant menaquinone was MK-9(H4). The major fatty acids (>10 %) were iso-C16 : 0, iso-C17 : 1ω6c, iso-C16 : 1 H and C16 : 1ω7c/iso-C15 : 0 2-OH. The DNA G+C content of strain SCSIO 11529T was 70.2 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain SCSIO 11529T belongs to the genus Prauserella , with the closest neighbours being Prauserella marina MS498T (97.0 % 16S rRNA gene sequence similarity), Prauserella rugosa DSM 43194T (96.4 %) and Prauserella flava YIM 90630T (95.9 %). Based on the evidence of the present study, strain SCSIO 11529T is considered to represent a novel species of the genus Prauserella , for which the name Prauserella coralliicola sp. nov. is proposed. The type strain is SCSIO 11529T ( = DSM 45821T = NBRC 109418T).

A novel Gram-stain-negative, rod-shaped, yellow bacterium, designated as LB1R16T, was isolated from the Laigu glacier on the Tibetan Plateau, PR China. Strain LB1R16T was catalase-positive, oxidase-negative and grew at 0–28 °C, pH 6.0–8.0 and in the absence of NaCl. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain LB1R16T belongs to the family Sphingosinicellaceae but formed an independent lineage. The highest level of 16S rRNA gene sequence similarities were found to Polymorphobacter arshaanensis DJ1R-1T (95.24 %), Sphingoaurantiacus capsulatus YLT33T (94.78 %) and Sandarakinorhabdus limnophila DSM 17366T (94.67 %). The genomic DNA G+C content was 68.8 mol%. The main cellular fatty acids were summed feature 8 (C18 : 1  ω7c/C18 : 1  ω6c), summed feature 3 (C16 : 1  ω7c/C16 : 1  ω6c), C16 : 0 and C12 : 0-OH. The respiratory quinone was ubiquinone-10. The polar lipids were phosphatidylethanolamine, phosphatidylglycerol, one sphingoglycolipid, one unidentified aminolipid, one unidentified phospholipid and two unidentified polar lipids, which were different from the type strains of Polymorphobacter arshaanensis , Sphingoaurantiacus capsulatus and Sandarakinorhabdus limnophila . Based on a polyphasic approach, a novel species of a new genus, Glacieibacterium frigidum gen. nov., sp. nov., within the family Sphingosinicellaceae is proposed. The type strain is LB1R16T (=CGMCC 1.11941T=NBRC 113873T).

A novel Gram-staining-negative, strictly aerobic bacterium, designated BS14T, was isolated from a marine tidal flat of the South Sea in Korea. Colonies were opaque, white, smooth and circular on marine agar. Cells were moderately halophilic, non-motile rods showing catalase- and oxidase-positive reactions. Growth of strain BS14T was observed at 5–40 °C (optimum: 30 °C), pH 6.5–9.5 (optimum: 7.0–7.5) and 0–10 % (w/v) NaCl (optimum: 1–1.5 %). The G+C content of the genomic DNA was 61.6 mol%. Strain BS14T contained ubiquinone-10 (Q-10) as the sole respiratory quinone and summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c), C18 : 0 3-OH, C10 : 0 3-OH and C18 : 0 as the major fatty acids. The polar lipid pattern comprised phosphatidylethanolamine, diphosphatidylglycerol, an unidentified aminolipid, an unidentified phospholipid and an unidentified polar lipid. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain BS14T formed a tight phylogenetic lineage with Defluviimonas denitrificans D9-3T with a bootstrap value of 100 %. The 16S rRNA gene sequence similarity between strain BS14T and D. denitrificans D9-3T was 97.4 % and their DNA–DNA relatedness was 19.1±3.6 %. Based on the phenotypic and genotypic studies, strain BS14T represents a novel species of the genus Defluviimonas , for which the name Defluviimonas aestuarii sp. nov. is proposed. The type strain is BS14T ( = KACC 16442T = JCM 18630T). An emended description of the genus Defluviimonas Foesel et al. 2011 is also proposed.

A Gram-positive, spore-forming, aerobic, rod-shaped, xylanolytic bacterium designated strain CC-Alfalfa-35T was isolated from the rhizosphere of Medicago sativa L. in Taiwan. Phylogenetic analysis based on 16S rRNA gene sequence showed that strain CC-Alfalfa-35T was affiliated to the genus Cohnella . Strain CC-Alfalfa-35T shared 95.3 % pairwise 16S rRNA gene sequence similarity to the type strain of the type species of the genus Cohnella ( Cohnella thermotolerans DSM 17683T) besides showing a similarity of 97.4–93.6 % with other recognized species of the genus Cohnella . The DNA–DNA hybridization value between CC-Alfalfa-35T and Cohnella thailandensis KCTC 22296T was 37.7 %±1.7 % (reciprocal value, 55.7 %±3.0 %). Predominant cellular fatty acids were iso-C16 : 0 and anteiso-C15 : 0. The polar lipid profile constituted diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, lysyl-phosphatidylglycerol, three unidentified phospholipids and three unidentified aminophospholipids. The major respiratory quinone was MK-7 and the DNA G+C content was 58.3 mol%. Strain CC-Alfalfa-35T contained meso-diaminopimelic acid as the major diamino acid in the cell-wall peptidoglycan. Based on the polar lipid and fatty acid profiles, which were in line with those of C. thermotolerans DSM 17683T, coupled with additional distinguishing genotypic, phenotypic and chemotaxonomic features, strain CC-Alfalfa-35T is proposed to represent a novel species within the genus Cohnella , for which the name Cohnella formosensis sp. nov. is proposed. The type strain is CC-Alfalfa-35T ( = JCM 18405T = BCRC 80428T).

A novel halophilic, Gram-reaction-negative, strictly aerobic, non-motile, rod-shaped and oxidase- and catalase-positive bacterial strain, designated WCC 4520T, was isolated from a semi-hard, Raclette-type cheese. The colonies were yellow–orange; flexirubin-type pigments were not found. The strain hydrolysed gelatin, hippurate, tyrosine and Tweens 20 and 80. Optimal growth was observed with 6–8 % (w/v) NaCl, at pH 7–8 and at 27–30 °C. The genomic DNA G+C content was 33.6 mol%. In phylogenetic analysis based on 16S rRNA gene sequences, strain WCC 4520T appeared to be a member of the family Flavobacteriaceae and the closest phylogenetic neighbours were identified as Psychroflexus gondwanensis DSM 5423T (94.0 % 16S rRNA gene sequence similarity) and Psychroflexus salinarum CCUG 56752T (94.0 %). The predominant cellular fatty acids were iso-C15 : 0, anteiso-C15 : 0, iso-C15 : 1 G and iso-C17 : 0 3-OH. The only detected quinone was MK-6 and the major polar lipids were phosphatidylethanolamine, an unidentified aminolipid and an unidentified polar lipid. Minor polar lipids and traces of polyamines were also detected. On the basis of the data presented, strain WCC 4520T represents a novel species of the genus Psychroflexus , for which the name Psychroflexus halocasei sp. nov. is proposed. The type strain is WCC 4520T ( = LMG 25857T = CCUG 59705T).

Two actinomycete strains, designated TRM 49117T and TRM 49136T, were isolated from a hypersaline habitat in Xinjiang Province, north-west China and were characterized taxonomically by using a polyphasic study. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain TRM 49117T had 93.93 % similarity with the type strain Glycomyces halotolerans TRM 40137T (GenBank accession no. HQ651156) and TRM 49136T had 94.32 % similarity with G. halotolerans TRM 40137T. The 16S rRNA gene sequence similarity between the two new isolates was 93 %. The isolates contained meso-diaminopimelic acid as the diagnostic diamino acid and anteiso-C15 : 0, iso-C16 : 0 and anteiso-C17 : 0 as major cellular fatty acids. The predominant menaquinones of the isolates were MK-9(H4) and MK-9(H6). The whole-cell sugar patterns of these strains contained xylose and ribose, and strain TRM 49136T also contained arabinose. The polar lipid pattern of strain TRM 49117T comprised phosphatidylglycerol, diphosphatidylglycerol, phosphatidylinositol mannosides, phosphatidylcholine, phosphatidylinositol and three additional unknown phospholipids. The polar lipid pattern of strain TRM 49136T comprised phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylinositol, glycolipids and two phosphoglycolipids of unknown composition. Genotypic and phenotypic data confirmed that strains TRM 49117T and TRM 49136T represent two novel species, clearly different from related species of the genus Glycomyces , for which the names Glycomyces fuscus sp. nov. (type strain TRM 49117T = CCTCC AA 2013003T = NRRL B-59998T = KACC 17682T) and Glycomyces albus sp. nov. (type strain TRM 49136T = CCTCC AA 2013004T = NRRL B-24927T = KACC 17681T) are proposed.

Two Haemophilus -like isolates with similar biochemical characteristics, designated strains SZY H1T and SZY H2, were isolated from human semen specimens. Cells were Gram-negative, non-motile, non-acid-fast, pleomorphic rods or coccobacilli. The major fatty acids (>10 %) were C16 : 0, C14 : 0, iso-C16 : 0 and/or C14 : 0 3-OH and C16 : 1 ω6c and/or C16 : 1 ω7c. The polar lipids were determined to be phosphatidylethanolamine, phosphatidylglycerol, an unidentified phospholipid, an unidentified aminophospholipid, two unidentified polar lipids and four unidentified aminolipids. The major polyamine was found to be cadaverine. The near-full-length (1462 nt) 16S rRNA gene sequences analysis showed the two isolates were nearly identical (>99.8 %), and closely matched Haemophilus haemolyticus ATCC 33390T with 98.9–99.1 % sequence similarities. Phylogenetic analysis based on 16S rRNA gene sequences and concatenation of 30 protein markers also revealed that the isolates clustered together with H. haemolyticus ATCC 33390T, and formed a distinct lineage well separated from the other members of the genus Haemophilus . Further, the average nucleotide identity values between the two isolates and their related species were below the established cut-off values for species delineation (95 %). Based on these findings, the two isolates are considered to represent a new species of the genus Haemophilus , for which name Haemophilus seminalis sp. nov. is proposed. The type strain is SZY H1T (=NBRC 113782T=CGMCC 1.17137T).

An aerobic, Gram-negative-staining, motile, psychrophilic bacterium, designated strain S8-3T, was isolated from alpine soil. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain S8-3T was related to the genus Sphingomonas and had highest 16S rRNA gene sequence similarity with Sphingomonas oligophenolica S213T (98.0 %). 16S RNA gene sequence similarity between strain S8-3T and Sphingomonas paucimobilis ATCC 29837T (the type species of the genus Sphingomonas ) was 93.0 %. Strain S8-3T contained Q-10 as the ubiquinone and C18 : 1ω7c (65.0 %) and C14 : 0 2-OH (13.4 %) as the dominant fatty acids (>10 %). The major polyamines were the triamine sym-homospermidine and spermidine. The polar lipid profile contained sphingoglycolipid, phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine and phosphatidylmonomethylethanolamine. The DNA G+C content was 64.1 mol%. Combined data from phenotypic, phylogenetic and DNA–DNA relatedness studies demonstrated that strain S8-3T is a representative of a novel species of the genus Sphingomonas , for which the name Sphingomonas alpina sp. nov. is proposed. The type strain is S8-3T ( = DSM 22537T = LMG 26055T).

A Gram-stain-negative, strictly aerobic, yellow-pigmented, rod-shaped and N-acylhomoserine lactone-degrading bacterium, designated strain Th78T, was isolated from the intestine of a cultured flounder (Paralichthys olivaceus). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain Th78T belonged to the genus Flaviramulus (family Flavobacteriaceae ) and showed the highest 16S rRNA gene sequence similarity to Flaviramulus basaltis H35T (96.70 %). Optimal growth occurred in the presence of 2–3 % (w/v) NaCl, at pH 7.0–8.0 and at 28 °C. The major fatty acids were iso-C15 : 0 3-OH, iso-C15 : 1 G and iso-C17 : 0 3-OH. The major polar lipids were phosphatidylethanolamine, one unidentified aminolipid and three unidentified polar lipids. Menaquinone 6 (MK-6) was the only respiratory quinone. The DNA G+C content of strain Th78T was 31.5 mol%. On the basis of polyphasic analysis, strain Th78T is considered to represent a novel species of the genus Flaviramulus , for which the name Flaviramulus ichthyoenteri sp. nov. is proposed. The type strain is Th78T ( = JCM 18634T = KCTC 32142T = DSM 26285T). Emended descriptions of the genus Flaviramulus and Flaviramulus basaltis are also proposed.

A Gram-stain-negative, facultatively anaerobic, rod-shaped, motile bacterium with a subpolar or lateral flagellum, designated strain XH089T, was isolated from deep-sea sediment sample collected from the South Pacific Gyre (41°51′ S 153°06′ W) during the Integrated Ocean Drilling Program (IODP) Expedition 329. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain XH089T belonged to the genus Achromobacter and showed the highest 16S rRNA gene sequence similarities with Achromobacter ruhlandii ATCC 15749T (96.95 %), Achromobacter denitrificans DSM 30026T (96.70 %) and Achromobacter marplatensis B2T (96.66 %). The DNA G+C content of strain XH089T was 66.5 mol%. The major fatty acids were C16 : 0 and C17 : 0 cyclo. The major polar lipids were phosphatidylethanolamine, phosphatidylmonomethylethanolamine, diphosphatidylglycerol, three unknown phospholipids and four unknown polar lipids. On the basis of data from the polyphasic analysis, strain XH089T is considered to represent a novel species of the genus Achromobacter , for which the name Achromobacter sediminum sp. nov. is proposed. The type strain is XH089T ( = DSM 27279T = JCM 19223T).
