Immature zygotic embryos of Carica papaya L. var. P-7-9 were inoculated on half-strength MS with different concentrations of 2,4-D and picloram alone and their combinations. The highest induction (80%) of somatic embryogenesis was MS supplemented with 2,4-D (7.0 mg/l) followed by 2,4-D (4.0 mg/l) + picloram (1.0 mg/l) but no embryogenesis was observed in auxin-free medium. The addition of osmoticum such as PEG and ABA significantly increased the maturation of somatic embryos. BAP, TDZ and NAA were used for shoot induction. Combination of 2.0 mg/l BAP and 0.5 mg/l NAA was found significantly better for shoot bud proliferation and 70% multiplication. The percentage of germinating embryos was enhanced significantly when the MS fortified with GA3 (0.5 mg/l). However, the high frequency of embryo germination and plantlets formation were obtained with 2,4-D (4.0 mg/l) + picloram (1.0 mg/l). Plant Tissue Cult. & Biotech. 29(1): 25-32, 2019 (June)

Considering the growing economic importance of tropical flowers and the advantages of techniques applied to the in vitro cultivation of these plants, it is necessary to carry out studies to evaluate growth in species such as Heliconia latispatha. The aim of this study therefore, was to evaluate in vitro shoot induction for different concentrations of BAP and NAA and as a function of the photoperiod. Explants from zygotic embryos were inoculated in MS medium containing different concentrations of BAP (0.0, 0.5, 1.0, 1.5, 2.0 or 2.5 mg L-1), with the cultures kept in a growth room at a temperature of 24.0 ± 2.0° C, under a photoperiod of 12 and 16 hours of light and a light intensity of 30 μmol m-2 s-1. At 21, 28, 35, 42 and 49 days after inoculation, the number of shoots per explant was evaluated. The treatment at the BAP concentration that gave the best multiplication rate (2.5 mg L-1) was set, and was tested in a further trial with different concentrations of NAA (0.0, 0.2, 0.4, 0 6, 0.8 or 1.0 mg L-1) under the same conditions as the previous experiment. The experimental designs were completely randomised, with five replications, and analysed in a 6 x 2 factorial. The data were submitted to analysis of variance and regression. No significant differences were seen in relation to the photoperiod or its interaction with the cytokinin and auxin under test. Multiplication was greater in the presence of 2.5 mg L-1 BAP, which gave a rate of 1.25 shoots/explant at 49 days of in vitro culture. The association of this BAP dosage with 1.0 mg L-1 NAA was even more efficient, producing 1.83 shoots per explant at 30 days of growth. The use of BAP together with NAA is beneficial to the induction of shoots in H. latispatha.

The shoot-tip explant harvested from ethyl methane sulphonate (EMS) and gamma ray (GR) mutagenized seedling was cultured over MS medium fortified with NAA and BAP for five generations to amplify the mutated sector. Mutagens reduced the regeneration efficiency of the explant and affected its plant growth regulator-dependence for multiple shoot induction. While the 12d-old shoot-tip from GR-treated seedling induced shoots with 0.5µM NAA+6.6µM BAP; that from EMS-treated seedling induced shoots with 8.8µM BAP. The present study establishes that the mutagens affect the regeneration process in the explant.

An efficient regeneration/transformation system was developed for `Beurre Bosc' pear. Young leaves were harvested from in vitro shoots proliferated on a medium containing MS basal salts and 5 BAP, 0.5 μM IBA, and 0.6M3. Shoot regeneration was optimized using a modification of the medium of Chevreau and Leblay (1993). Explants were cultured on shoot induction medium contained 10 μM TDZ and 1 μM IBA for 4 weeks in the dark, and then transfered to a similar, but auxinless, regeneration medium until shoots developed, usually after an additional 4 to 8 weeks. Leaf tissues were transformed by co-cultivation for 3 days with Agrobacterium tumefaciens EHA101 carrying a pGA482 plasmid containing NPTII, GUS, and rolC genes, followed by cultivation on SIM containing 300 mg/L timentin. Putative transgenic plants were selected on shoot induction medium containing 80mg/L kanamycin, and multiplied on shoot proliferation medium. Four clones were confirmed as transgenic using the GUS histochemical assay and Southern blots for the NPTII and rolC genes. Plants of each clone have been rooted and successfully transfered to the greenhouse for further analysis of gene expression.

Abstract Background Trachyspermum ammi is one of the key medicinal plant species with many beneficial properties. Thymol is the most important substance in the essential oil of this plant. Thymol is a natural monoterpene phenol with high anti-microbial, anti-bacterial, and anti-oxidant properties. Thymol in the latest research has a significant impact on slowing the progression of cancer cells in human. In this research, embryos were employed as convenient explants for the fast and effectual regeneration and transformation of T. ammi. To regenerate this plant, Murashige and Skoog (MS) and Gamborg's B5 (B5) media were supplemented with diverse concentrations of plant growth regulators, such as 6-benzyladenine (BA), 1-naphthaleneacetic acid (NAA), 2,4-dichlorophenoxyacetic acid (2,4-D), and kinetin (kin). Transgenic Trachyspermum ammi plants were also obtained using Agrobacterium-mediated transformation and zygotic embryos explants. Moreover, two Agrobacterium tumefaciens strains (EHA101 and LBA4404) harboring pBI121-TPS2 were utilized for genetic transformation to Trachyspermum ammi. Results According to the obtained results, the highest plant-regeneration frequency was obtained with B5 medium supplemented with 0.5 mg/l BA and 1 mg/l NAA. The integrated gene was also approved using the PCR reaction and the Southern blot method. Results also showed that the EHA101 strain outperformed another strain in inoculation time (30 s) and co-cultivation period (1 day) (transformation efficiency 19.29%). Furthermore, HPLC method demonstrated that the transformed plants contained a higher thymol level than non-transformed plants. Conclusions In this research, a fast protocol was introduced for the regeneration and transformation of Trachyspermum ammi, using zygotic embryo explants in 25–35 days. Our findings confirmed the increase in the thymol in the aerial part of Trachyspermum ammi. We further presented an efficacious technique for enhancing thymol content in Trachyspermum ammi using Agrobacterium-mediated plant transformation system that can be beneficial in genetic transformation and other plant biotechnology techniques.

An efficient protocol for plant regeneration through multiple shoots induction from shoot tips of Polygonum hydropiper (L.) was established. The highest percentage (96.6) of multiple shoot induction and number of shoots (9.0) per culture were found on MS supplemented with 2.0 mg/l Kn. The induced shoots were excised and inoculated on to MS contains different concentrations of IBA or NAA for rooting. The highest percentage (90.0) of root induction and the highest number of roots per shoot (12.0) was found on MS having 1.0 mg/l IBA.
