Methanol induced oxidative stress (OS) models in mice were successfully established and evaluated by the electron paramagnetic resonance (EPR) spin trapping technique. The capacity for removal of reactive oxygen species (ROS) free radicals by rhubarb and vitamin C (Vc) as candidate materials was also investigated. EPR was employed to determine the free radicals generated from a spin trapping agent, α-phenyl-N-tert-butylnitrone (PBN), that reacted with the ROS. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), and the level of malondialdehyde (MDA) were also evaluated by enzyme assays. The results indicated that methanol clearly promoted the generation of ROS free radicals in the liver of mice. The activity of SOD and GSH-PX was reduced significantly, although the level of MDA was increased as a result of the harmful effect of methanol. In addition, rhubarb and Vc exhibited a protective effect on the mice liver under acute OS.

N-[tert-13C]Butyl C-phenyl nitrone (PBN-[tert-13C]) has been synthesized for an EPR spin trapping competition study. The newly synthesized PBN-[tert-13C] shows different 13C-hyperfine splitting constants (a13C) when it traps free radicals as compared to another 13C-labeled PBN analogue, N-tert-butyl C-phenyl [nitronyl-13C]nitrone (PBN-[nitronyl-13C]). The PBN- [tert-13C] hydroxyl adduct gives a larger a13C value (5.14 G) as compared to the PBN-[nitronyl-13C] hydroxyl adduct (4.36 G). This gain ofthe a13C value decreases the chance of EPR signal overlap, thus providing a more resolved EPR spectrum when PBN-[tert-13C] is used as an internal standard for EPR spin trapping competition studies of hydroxyl radical formation.

AbstractNTO (5-Nitro-2,4-dihydro-3H-1,2,4-triazol-3-one)is shown to form aphotochemical free radical in alkaline aqueous solution containing nitromethane (CH3NO2). The NTO free radical can also be produced by chemical reduction of NTO with hydroquinone in alkaline solution. Details of radical formation and assignment of the NTO radical to a nitro anion radical, having the NTO ring intact, are presented.
