The likelihood that uric acid was the only compound capable of triggering germination of Bacillus fastidiosus spores was reinforced by the finding that ureidoglycollic acid, urea, NH4Cl, 2,8-dihydroxypurine and a combination of L-alanine and O-carbamoyl-D-serine were ineffective as germinants. Uric acid-triggered germination of B. fastidiosus was prevented by a range of inhibitors that also inhibited uricase activity in dormant spore extracts. O2 uptake during germination started immediately after addition of uric acid, possibly as a consequence of the oxidation of uric acid by the enzyme uricase. Germination showed a dependence on uric acid concentration, with a relatively high Km (4-5 mM). During the first 10 min of germination of heat-activated spores there was no detectable change in the number of spore-cortex reducing groups, indicating that selective cortex hydrolysis is not involved in the trigger mechanism of germination of B. fastidiosus. On the basis of the results, a model is proposed in which re-initiation of uricase activity is the mechanism by which B. fastidiosus spores are triggered to emerge from the dormant state.

In the present study some predominant bacteria from textile effluent were isolated and their decolorizing activity was evaluated. Heavy load of bacteria (1.3 × 103 to 3.07 × 107 cfu/ml) were found in eight effluent samples collected from two textile industries. Six dye decolorizing bacteria were isolated and identified as Bacillus fastidiosus, Bacillus polymyxa, Bacillus licheniformis, Bacillus megaterium, Staphylococcus aureus and Micrococcus luteus. The isolates were tested for their ability to decolorize different concentrations (0.01, 0.05 and 0.1%) of six different textile dyes. Among the dyes Terasil red W-FS and Terasil black W-NS were found to be completely decolorized by the all isolates at different concentrations. Cibacron yellow F-4G was decolorized very easily by B. fastidiosus while the other dyes were only partially decolorized by the test organisms. Cibacron blue FN-R, Cibacron navy FN-B and Cibacron orange FN-R were only weakly decolorized by the most of the isolates. The results of the present study show that B. fastidiosus, B. polymyxa, M. luteus and S. aureus were the potential isolates in the treatment of dyeing industry effluents and their potentiality can be exploited to clean up the environment. In the present investigation five cheese samples were collected and their pH and moisture contents were determined which was found to vary from 6.0 to 6.5 and 35.0-40.0% respectively. Major nutritional factors protein (1N NaOH soluble) and fat (chloroform soluble) of the samples were also analyzed and found 57.9 to 100.0 mg/g protein and 15-25% fat. The samples showed higher number of total bacterial count (5.3×104 - 2.9×107cfu/g) than the load of Lactobacilli (1.9x105 cfu/g) and Streptococci (2.1x105cfu/g). All the isolates were grouped into six on the basis of their morphological and cultural features. Six isolates were selected on the basis of their morphological, cultural, physiological and biochemical characteristics. An attempt was made to identify the isolates. They were found to belong to the three genera viz. Bacillus (S1X3, S1X8, S2Y1, and S2Y2), Staphylococcus (S1X7) and Planococcus (S1X11). They were provisionally identified as Bacillus fastidiosus (S1X3); B. sphaericus (S1X8); B. brevis (S2Y1); B. aminovorans (S2Y2); Staphylococcus saprophyticus (S1X7) and Planococcus citreus (S1X11).The selected isolates were screened for their efficacy in terms of curd and cheese production.
