Liquid chromatographic procedures employing two derivatisation reagents, dansylchloride and o-phthaldialdehyde (OPA), were compared and applied for the determination of biogenic amines in meat products during fermentation and long-term storage. Both methods give similar results in terms of the detection limits, repeatability, recovery, and accuracy. Secondary amines (spermine and spermidine) do not react with o-phthaldialdehyde thus only their LC determination after derivatisation with dansylchloride is possible. Their contents during fermentation and/or long-term storage are nearly constant or slightly decrease, thus their determination in both cases is not necessary. LC procedure employing OPA derivatisation is faster, much simpler in terms of the pre-treatment of samples, and it can be fully automated using an intelligent auto sampler.

Abstract Using high-performance liquid chromatography with electrochemical detection, we determined serotonin in plasma from parkinsonian patients being treated with L-3,4-dihydroxyphenylalanine or N-(DL-seryl)-N'(2,3,4-trihydroxybenzyl)hydrochloride plus L-3,4-dihydroxyphenylalanine ("Sinemet") and in serum from a blood bank, from "normal" persons, and a pooled specimen from a hospital clinical laboratory. The values obtained for the two groups of Parkinson's disease patients showed no significant difference. Long-term storage on solid CO2 was xhown to be an adequate technique for preserving samples. The mean (+/-SEAM) normal value obtained for serotonin in serum was 146 +/- 46 microgram/liter (n = 23), a result in harmony with that previously obtained [Clin. Chem. 20, 812 (1974)] by fluorometry. In comparison to other methods for measurement of serotonin in serum or plasma, we believe that the present scheme offers greater selectivity, sensitivity, and precision.

The aim of the article is to describe the issue of determining the characteristics and parameters of raw timber natural losses due to shrinkage at long-term storage, defining the theoretical basis for creating standards, and verify its finding by means of a case study in raw timber storing. This issue is very topical in forestry practice in the Czech Republic as well as in other countries. The lower and upper limits of the standards were calculated, and the proposed mean value was grouped according to woody plants that reflected the most frequent commercial usage with respect to subsequent processing. Subsequently, experimental verification was carried out on a representative sample of 2 209.99 m3 of raw timber of Norway spruce (Picea abies /L./ H. Karst.) at selected forest administrations. Furthermore, the article addresses the related accounting and tax issues of the standards of natural losses of raw timber. Currently, no legislation mandates an entity to establish standards.

Abstract The normal phase and reverse phase liquid chromatographic properties of seven 8-quinolinesulfonyl derivatives of primary and secondary amines are examined using dual wavelength ultraviolet detection. The amines are further identified by infrared spectrophotometry with emphasis on structural elucidation based on absorption bands at characteristic wavelengths.
