Lactobacillus plantarum N014 was isolated from nham, a traditional Thai fermented pork, and exhibited antimicrobial activity against Listeria monocytogenes. Its bacteriocin had a broad inhibitory spectrum toward both gram-positive and gram-negative bacteria. The bacteriocin activity was sensitive to all proteolytic enzymes used in this study, including papain, pepsin, pronase E, proteinase K, and trypsin, but was resistant to the other enzymes, such as α-amylase, lipase A, and lysozyme. Furthermore, activity was stable over various heat treatments and pH values. The bacteriocin exerted a bacteriolytic mode of action. It was produced during the exponential growth phase and reached its highest level as producer cells entered the stationary phase. Adsorption of the bacteriocin onto producer cells was pH-dependent. No bacteriocin adsorption was detected at pH 1 to 3, whereas 100% bacteriocin adsorption was found at pH 7. Plasmid isolation revealed that L. plantarum N014 contained no plasmids. From Tricine–sodium dodecyl sulfate–polyacrylamide gel electrophoresis and growth inhibition testing against L. monocytogenes, the estimated molecular mass of L. plantarum N014 bacteriocin was 8 kDa.

ABSTRACT The α-proteobacterial strain Sphingobium japonicum UT26 utilizes a highly chlorinated pesticide, γ-hexachlorocyclohexane (γ-HCH), as a sole source of carbon and energy, and haloalkane dehalogenase LinB catalyzes the second step of γ-HCH degradation in UT26. Functional complementation of a linB mutant of UT26, UT26DB, was performed by the exogenous plasmid isolation technique using HCH-contaminated soil, leading to our successful identification of a plasmid, pLB1, carrying the linB gene. Complete sequencing analysis of pLB1, with a size of 65,998 bp, revealed that it carries (i) 50 totally annotated coding sequences, (ii) an IS6100 composite transposon containing two copies of linB, and (iii) potential genes for replication, maintenance, and conjugative transfer with low levels of similarity to other homologues. A minireplicon assay demonstrated that a 2-kb region containing the predicted repA gene and its upstream region of pLB1 functions as an autonomously replicating unit in UT26. Furthermore, pLB1 was conjugally transferred from UT26DB to other α-proteobacterial strains but not to any of the β- or γ-proteobacterial strains examined to date. These results suggest that this exogenously isolated novel plasmid contributes to the dissemination of at least some genes for γ-HCH degradation in the natural environment. To the best of our knowledge, this is the first detailed report of a plasmid involved in γ-HCH degradation.

This learning cycle focused on developing of Biology education student’s performance in bacteria DNA-plasmid isolation as well as using gel electrophroresis. The aim of the study was to improve student’s performance in operating equipment used in plasmid isolation such as: micropipettes, microsentrifuge, vortex, shaker-incubator, gel electrophoresis, and UV Trans- illuminator camera. Beside that students were also trained to DNA-plasmid isolation base on standard procedure, so students could identify the form of plasmid via the use of gel electrophoresis. This study was conducted in 2 cycle learning processes as an enrichment of microbiology activity, which associated with molecular microbes genetics. Using observation sheet did evaluation. The study revealed that biology education students had good performance in isolating the plasmid and all group were succeed to isolate the plasmid. Beside that there were increased in knowledge ability especially associated with molecular microbes genetics subject, which achievment gain average was 18,3% (Class A) and 35,4% (class B). The DNA isolation activity in microbiology learning shared high contribution as well as increasing of knowledge achievment and performance ability.Keyword: DNA-plasmid, gel electrophoresis, isolation, learning cycle
