IntroductionEpithelial ovarian cancer (EOC) is the deadliest gynecologic malignancy worldwide. Reliable predictive biomarkers are urgently needed to estimate the risk of relapse and to improve treatment management. Soluble immune-checkpoints in EOC are promising molecules serving as prognostic biomarkers accessible via liquid biopsy. We thus, aimed at elucidating the role of sB7-H4 in EOC.Material and MethodsWe analyzed soluble serum B7-H4 (sB7-H4) using ELISA and circulating tumor cells (CTCs) in blood applying the AdnaTest OvarianCancer in 85 patients suffering from advanced EOC. Findings were correlated with clinical parameters as well as survival data.ResultssB7-H4 was detectable in 14.1% patients, CTCs in 32.9% patients and simultaneous presence of CTCs and sB7-H4 was found in 7% patients, respectively. Although no association between sB7-H4 and CTC could be documented, each of them served as independent predictive factors for overall survival (OS).ConclusionsB7-H4 and CTCs are independent prognostic biomarkers for impaired survival in EOC. As they are easily accessible via liquid biopsy, they may be of potential benefit for the prediction of therapy response and survival for EOC patients.

AbstractLiquid biopsy is noninvasive and convenient to detect cancer-derived materials in blood or other body fluids. The aim of this study was to identify tRNA-derived small RNAs (tsRNAs) in plasma that could distinguish patients with breast cancer (BC) from healthy controls. Basing on high-throughput sequencing, 15 significantly upregulated tsRNAs were selected and assessed in cell supernatants and cell lines. 6 tsRNAs were identified and verified in a large cohort of 120 patients with BC and 112 healthy controls. tRF-Arg-CCT-017, tRF-Gly-CCC-001, and tiRNA-Phe-GAA-003 could serve as novel diagnostic biomarkers. Meanwhile, tRF-Arg-CCT-017 and tiRNA-Phe-GAA-003 could also act as prognostic biomarkers. Target genes of these tsRNAs were related to the development of cancers. These results suggested that specific tsRNAs in plasma might serve as diagnostic and prognostic biomarkers of BC.

EGFR-Tyrosinkinase-Inhibitoren (EGFR-TKI) haben zu einem Paradigmenwechsel in der Behandlungsstrategie für nicht-kleinzellige Bronchialkarzinome (NSCLC) geführt und gehören inzwischen zum Standard. Dabei ist es auch wichtig, primäre und erworbene Resistenzen zu überwinden. Eine aktuelle Studie mit 116 Patienten ergab unterschiedliche Resistenz-Mechanismen für TKI der 1./2. und der 3. Generation. Die Liquid Biopsy ist nach Ansicht der Autoren eine Option für ein nicht-invasives Monitoring der molekularen Veränderungen unter der EGFR-Therapie. Ein Review beschäftigte sich mit den Resistenzmechanismen und der Liquid Biopsy als Instrument für die Therapieplanung und -monitoring.
