In vitro regeneration is a pre-requisite for developing transgenic plants through tissue culture-based genetic engineering approaches. Huge variations among different genotypes of the genus Brassica necessitate the identification of a set of regeneration conditions for a genotype, which can be reliably used in transformation experiments. In this study, we evaluated the morphogenesis potential of four commercial cultivars (Faisal canola, Punjab canola, Aari canola, Nifa Gold) and one model, Westar, from four different explants namely cotyledons, hypocotyls, petioles and roots on three different Brassica regeneration protocols, BRP-I, -II and -III. The regeneration efficiency was observed in the range of 6–73%, 4–79.3%, 0–50.6%, and 0–42.6% from cotyledons, petioles, hypocotyls and roots, respectively, whereas, the regeneration response in terms of average shoots per explant was found to be 0.76–10.9, 0.2–3.2, 0–3.4 and 0–2.7 from these explants. Of the commercial varieties tested, almost all varieties showed poorer regeneration than Westar except Aari canola. In comparison to Westar, its regeneration frequency from cotyledons was up to 7.5-fold higher on BRP-I, while it produced up to 21.9-fold more shoots per explant. Our data show that the explant has strong influence on the regeneration response, ranging from 24% to 92%. While the growth of commercial cultivars was least affected by the regeneration conditions provided, the effect on Westar was twice that of the commercial cultivars. After determining the optimal explant type and regeneration conditions, we also determined the minimum kanamycin concentration levels required to selectively inhibit the growth of untransformed cells for these cultivars. Regenerated shoots of Aari canola could be successfully grown to maturity within 16–18 weeks, with no altered phenotype noted and normal seed yields obtained. Therefore, the commercial variety, Aari canola, could be a good candidate for future genetic transformation studies.

An efficient and reproducible in vitro regeneration protocol was established for two varieties of ginger (Z. officinale Roscoe) namely, BARI Ada-1 and Chinese ginger accession number SG876). In case of BARI Ada-1 best result was obtained on MS supplemented with 2.0 mg/l BAP, 0.5 mg/l Kn and 0.5 mg/l NAA. In this combination, 95% rhizome bud explants responded within 6 - 8 days and mean number of shoots per explant was 8.79 ± 0.42. On the other hand, Chinese ginger showed best (90%) shoot regeneration response from the same explants on the same medium and hormonal combinations but in exchange of 0.25 mg/l NAA. In this hormonal composition shoot initiation started within 7 - 8 days of culture and mean number of shoots/explant was 6.83 ± 0.71 after 24 - 27 days of culture. Maximum root induction (90 and 80%) was found on MS supplemented with 0.5 mg/l IBA and 0.5 mg/l NAA in case of BARI Ada-1 and Chinese ginger, respectively. The in vitro regenerated plantlets were successfully transplanted into the soil after acclimatization.

An efficient regeneration protocol was established for two varieties (BARI tomato-9 and BARI tomato-15) of tomato (Lycopersicon esculentum Mill.) using three explants namely cotyledonary node, cotyledonary leaf and hypocotyls. Among the three explants, maximum number of shoots was produced from cotyledonary leaf explants of BARI tomato-15 on MS with 2.0 mg/l BAP and 0.5 mg/l IAA. In this combination of BAP and IAA 86%, on an average, cotyledonary leaf explants showed regeneration response 14.12 shoots/explants. Explants from hypocotyl showed best results in MS medium with 2.0 mg/l BAP and 0.2 mg/l IAA in both the varieties. In case of cotyledonary node, BARI tomato-15 showed 6.0 shoot/explant on MS with 2.0 mg/l BAP and 1.0 mg/l IAA. Molecular characterization of total ten varieties of tomato in Bangladesh was done by using six arbitrary oligonucleotide RAPD primers. A total of 140 bands were produced where the highest genetic distance (0.6769) was found between BARI tomato-3 and Mintoo tomato and lowest distance (0.1035) was observed between BARI tomato-7 and BARI tomato-8. Abstract Transverse sections (2 mm thickness) of stem-nodes from in vitro raised seedlings had morphogenic potential on semisolid and liquid Murashige and Skoog medium supplemented with cytokinins N6-benzyladenine (BA 4.44 μM), furfurylaminopurine (KIN 4.65 μM) and auxin α-naphthalene acetic acid (NAA 5.37 μM) individually and in combinations. The regeneration response was influenced by both the type of growth regulator and physical state of the medium. The explants produced either shoot buds on cytokinincontaining media or protocorm-like bodies (PLBs) on NAA containing media both solid and liquid. More neo-formations were produced on liquid media, especially those containing only NAA. They were formed at nodal and inter-nodal regions. The secondary buds were produced on the surface of primary PLBs. The plantlets were developed on MS medium containing banana homogenate 50 g·dm-3. The current study is the first ever report on successful regeneration of Dendrobium chrysotoxum from stem-node segments.
