ABSTRACT Sinorhizobium meliloti strains lacking BacA function are impaired in symbiosis with alfalfa host plants and display altered sensitivities to a number of compounds relative to wild-type strains. With the goal of finding clues to the currently unknown biological function(s) of BacA, we carried out a genetic analysis to determine which amino acids are critical for protein function and to attempt to ascertain whether the multiple phenotypes that result from abacA-null allele were the result of a common cause or whether BacA has multiple functions. We have created a set of 20 site-directed mutants in which selected individual amino acids inbacA were replaced with glycine residues. The resulting mutants were characterized to determine how the various amino acid changes affected a number of phenotypes associated with loss of BacA function. Mutants H165G, W182G, D198G, and R284G had null phenotypes for all functions assayed, while mutants W57G, S83G, S231G, and K350G were indistinguishable from wild-type strains. The remaining 12 site-directed mutants demonstrate mixed phenotypic characteristics and fall into a number of distinctly different groups. These observations may be consistent with a role for BacA in multiple, nonoverlapping functions.

SummaryAccording to the classical genetic analysis in Paramecium caudatum by Tsukii & Hiwatashi (1983), the E mating type of each syngen is expressed when the cell bears alleles specific for syngen at the Mt locus. The O mating type is expressed when cells are homozygous for the null allele, mt, at the Mt locus. In such mt/mt cells the O syngen specificity is determined by alleles at two other loci called MA and MB. Inthe study reported here, macronucleoplasmic transplantation technique was used to test the above hypothesis. When macronucleoplasm of type E3 (mating type E of syngen 3) was injected into a macronucleus of type O12 (mating type O of syngen 12), some recipients changed to type E of the donor syngen but some others changed to type E of the recipient syngen. Thus, syngen specificity of donor macronucleoplasm controlling type E was converted into that of the recipients, even though the latter has no gene that controls type E. When this transformant expressing type E of the recipiexnt syngen was re-injected back into E of the other syngen, the expression of the converted mating type in some way continued in the recipient. This suggests that syngen specificity of gene Mt of the donor was changed to that of the recipients by intersyngenic transplantation of macronucleoplasm. We also obtained results suggesting that the gene dosage ratio of Mt to mt or Mt to MA and MB may be important for syngen specific expression of type E.

AbstractBeing an economical and endangered species, microsatellite markers of Taxus chinensis var. mairei were very limited. We have developed a set of microsatellite markers, which was benefit for future genetic analysis of this rare species. Polymorphic loci were developed from congeneric species by cross-species amplification methods, and new primers were redesigned to test for potential null alleles. 15 loci showed polymorphism. The number of alleles per locus varied from 2 to 23 tested in 48 individuals. The observed heterozygosity (Ho) and expected heterozygosity (He) values ranged form 0.000 to 0.854 and 0.082 to 0.827, respectively. Newly redesigned primer confirmed that no null allele existed in most suspected loci. These microsatellite markers will be useful for future genetic analysis and conservation of this endangered species.
